In the glandular trichomes of cannabis, two functionally characterised enzymes, CsTPS18VF and CsTPS19BL, are nerolidol/linalool synthases (Zager 2019). Content depends on the variety: the hemp « Finola » contains few monoterpene alcohols such as linalool, although they are common in other varieties (Booth 2017).","originNote":"A monoterpene alcohol found in the essential oils of many aromatic plants. It made up 23.8 to 33.0 % of true lavender essential oil (Lavandula angustifolia) in 20 authentic batches, against a range of 20.0 to 45.0 % set by the European Pharmacopoeia (Capetti 2021), and about 70 % of coriander fruit essential oil (Burdock 2009). It is among the most common terpenoids of cannabis (Santiago 2019), without being one of the three most abundant on average in US commercial samples, which are myrcene, β-caryophyllene and limonene (Smith 2022). Hemp makes it itself through terpene synthases in its trichomes (Zager 2019).","discovered":""},"pharmacology":{"summaryFr":"Linalool is a terpene, not a cannabinoid: an acyclic ten-carbon monoterpene alcohol, it has neither the skeleton of phytocannabinoids nor an established affinity for their receptors. It exists as two enantiomers, (R)-(−)-linalool, isolated from lavender oil, and (S)-(+)-linalool, isolated from coriander oil (Sugawara 2000); in 20 authentic lavender oils, the (R) form made up 93.8 % of the linalool (Capetti 2021). Its action at cannabinoid receptors remains disputed. In cells expressing human CB1 or CB2, it produced no effect up to 100 µM, alone or combined with Δ9-THC (Santiago 2019); the study by Finlay et al. (2020), often cited alongside it, did not test linalool. LaVigne et al. (2021), by contrast, described rimonabant-sensitive CB1 activation, at concentrations above 10 µM and up to 500 µM depending on the terpene, with no measurable binding at the orthosteric site; in mice, 200 mg/kg given intraperitoneally produced hypothermia, catalepsy and hypolocomotion, as well as weak antinociception blocked by rimonabant. Other targets appear only at millimolar concentrations: potentiation of GABA-A receptors (Milanos 2017), non-competitive inhibition of binding to the NMDA receptor (Brum 2001). In mice, linalool odour produced an anxiolytic-like effect without motor impairment, absent in animals made anosmic and abolished by flumazenil (Harada 2018); inhaled for one hour at 1 or 3 %, it prolonged pentobarbital-induced sleep without impairing motor coordination (Linck 2009). In humans, pharmacokinetic data are limited to a few measurements, including a peak plasma level of 121 ng/ml 19 minutes after a massage with lavender oil in one volunteer (EMA 2012). In air, linalool oxidises into allergenic hydroperoxides: tested at 6.0 % in 2 900 patients from six countries, oxidised linalool gave 6.9 % positive reactions (Bråred Christensson 2012). In the United States, it is listed among synthetic flavouring substances generally recognised as safe (21 CFR 182.60).","receptorSummaryFr":"Linalool is not an established cannabinoid ligand, and the two studies that tested it at these receptors disagree. In AtT20 cells expressing human CB1 or CB2, racemic linalool, from 1 nM to 100 µM, changed neither the membrane potential nor the response to CP55,940 or Δ9-THC (Santiago 2019). In CHO cells expressing human CB1, by contrast, it activated ERK in a rimonabant-sensitive manner and partially inhibited cAMP production, but only at high micromolar concentrations, without measurably displacing [3H]CP55,940 or recruiting β-arrestin 2; the authors do not rule out an indirect mechanism, through the membrane or through 2-AG (LaVigne 2021). Outside the cannabinoid system, at 2 mM it potentiates the current of recombinant α1β2γ2 GABA-A receptors (Milanos 2017), non-competitively inhibits [3H]MK801 binding to the NMDA receptor complex (IC50 = 2.97 mM) without changing [3H]muscimol binding (Brum 2001), and blocks the voltage-gated sodium current of dissociated dorsal root ganglion neurons (Leal-Cardoso 2010).","binding":[{"target":"CB1","efficacy":"unknown","measure":null,"valueNm":null,"species":"human","assay":"membrane potential (FLIPR), AtT20 cells expressing human CB1; racemic linalool from 1
