Pharmacology

JWH-057, or 1-deoxy-Δ8-THC-DMH, is a synthetic cannabinoid designated by the JWH code, used in the work of John W. Huffman at Clemson University. It is an analogue of Δ8-THC that lacks a hydroxyl at position 1, and whose side chain at position 3 is a 1′,1′-dimethylheptyl. Huffman et al. (2010) report an affinity constant of Ki 23 ± 7 nM for CB1 and Ki 2.9 ± 1.6 nM for CB2, a selectivity of nearly ten-fold for CB2. The authors describe it as potent in vivo. No human data were found in the sources consulted, and no therapeutic indication is established. In France, it falls under a generic clause of the narcotics list.

History

Source
Synthetic molecule designed in laboratories studying cannabinoid receptors, with no established therapeutic indication. No source consulted reports it in hemp or in any other living organism. It appears in the literature around 1996, in parallel with the work of Huffman's team at Clemson University.

Origin (pharmaceutical research → illicit market)

No biosynthetic pathway: JWH-057 is produced by no living organism identified in the sources consulted. It belongs to chemical synthesis alone, which assembles a partially hydrogenated benzo[c]chromene nucleus and a 1′,1′-dimethylheptyl side chain on the aromatic ring.
