Décrit pour la première fois en 1978 par Garrett, Gouyette et Roseboom, qui l'identifient parmi les produits de dégradation du Δ9-THC dans leurs travaux sur la stabilité des tétrahydrocannabinols. Reggio et ses collègues en réalisent la synthèse ciblée en 1991, sous la forme 9β, dans une recherche de séparation entre analgésie et psychoactivité. 9-OH-HHC is a semi-synthetic molecule close to HHC, absent from cannabis. It appears above all as a by-product during the laboratory manufacture of hemp derivatives, and is therefore found as an impurity in certain products on sale. The few published assays, carried out in the mouse, indicate effects far weaker than those of THC, and no human data exist.

Pharmacology

9-OH-HHC retains the classic cannabinoid pharmacophore, a phenol at position 1, a pentyl chain at position 3 and a pyran ring, but bears a tertiary alcohol at position 9 on a fully saturated C ring. This combination was deliberately sought by Reggio and colleagues in 1991 in an attempt to dissociate analgesia from psychoactivity: the two epimers 9α and 9β proved markedly less active than Δ9-THC in the battery of behavioural and antinociceptive tests in the mouse, molecular modelling attributing this loss of activity to an unfavourable conformation. Watanabe and colleagues found a comparable profile in 2007 for 9α-OH-HHC, with a weak antinociceptive effect. No CB1 or CB2 affinity constant has been published for this molecule, nothing has been measured at TRPV1, GPR55, PPARγ or 5-HT1A, and human data are entirely lacking. Partial CB1 agonism therefore remains a hypothesis inferred from the behavioural profile, not a measurement.

No plant biosynthesis: the molecule is not produced by cannabis. It results from a hydroxylation at position 9 on a hexahydrocannabinol skeleton, obtained chemically during the acid cyclisation of CBD or the reduction of Δ9-THC, and recovered as an impurity rather than as an intended product.

Pharmacokinetics

Aucune étude de pharmacocinétique humaine n'a été publiée : absorption, demi-vie, volume de distribution et biodisponibilité restent inconnus. Les seules administrations documentées concernent la souris, dans des travaux de pharmacologie comportementale qui ne renseignent pas sur le devenir du composé chez l'être humain.

Metabolism

Aucune voie métabolique humaine n'est documentée pour le 9-OH-HHC. La littérature le présente comme un sous-produit chimique et non comme un métabolite formé par l'organisme, les métabolites hydroxylés du HHC décrits jusqu'ici portant plutôt sur les positions 8 et 11. Le devenir de l'alcool tertiaire de la position 9, notamment sa conjugaison éventuelle, n'a pas été étudié.

Toxicology and risks

Aucune étude de toxicologie n'a été conduite sur le 9-OH-HHC, ni chez l'animal au-delà de tests comportementaux aigus, ni chez l'être humain. Le risque documenté tient à son statut d'impureté non caractérisée : il accompagne des produits de Δ8-THC ou de HHC fabriqués hors de tout cadre pharmaceutique, dont la composition réelle diffère souvent de l'étiquetage. L'ANSM a rapporté pour cette famille de cannabinoïdes des intoxications graves, vomissements, perte de connaissance, convulsions, tachycardie, sans qu'aucun de ces cas soit attribuable au 9-OH-HHC en particulier.

Detection and analysis

Le 9-OH-HHC échappe aux tests immunochimiques urinaires courants, comme l'ensemble des cannabinoïdes de synthèse et semi-synthétiques. Son identification demande une chromatographie liquide couplée à la spectrométrie de masse : Schirmer et ses collègues ont validé en 2026 une méthode de criblage LC-QTOF-MS sur sang total incluant les étalons 9α-OH-HHC et 9β-OH-HHC. Au niveau du produit lui-même, la caractérisation repose sur la GC-MS et la résonance magnétique nucléaire, comme dans les travaux d'isolement publiés en 2023.
