Le THCP-O ne dispose pas d'une publication propre qui le décrirait pour la première fois. La molécule dont il dérive, le Δ9-THCP, a été isolée du cannabis en 2019 par l'équipe de Cinzia Citti et Giuseppe Cannazza. L'ester acétique, lui, est apparu ensuite sur le marché gris et ce sont les laboratoires de contrôle qui l'ont caractérisé : l'Institut national japonais des sciences de la santé l'a identifié en 2025 dans des huiles et un produit végétal, aux côtés d'esters propanoate et butanoate apparentés, et il figure parmi les substances relevées la même année dans des saisies analysées en Allemagne.

Pharmacology

THCP-O is the acetate ester of Δ9-THCP, formed on the phenol at position 1. Yet it is precisely this free hydroxyl that ensures recognition by the CB1 and CB2 receptors: once masked, direct action becomes improbable, and the molecule is regarded as a prodrug that must be hydrolysed by esterases to release THCP. No binding, pharmacokinetic or toxicological study has been published on the ester itself, and human data are entirely lacking. What is established concerns the released THCP: a CB1 agonist of very high affinity (Ki of 1.2 nM), active in animals in the tetrad test, and associated in humans with one reported case of psychosis after a single oral intake. To this is added a risk specific to the cannabinoid acetate family, the heating of which releases ketene, a toxic gas, demonstrated for the acetates of Δ8-THC, CBN and CBD under vaping conditions.

Pharmacokinetics

Aucune donnée publiée pour le THCP-O. Par analogie avec les autres acétates de cannabinoïdes, l'ester est très lipophile et doit être hydrolysé par les estérases avant toute activité, ce qui décale l'apparition des effets par rapport au cannabinoïde libre correspondant. Aucune demi-vie, aucune biodisponibilité et aucun volume de distribution ne sont établis, ni chez l'animal ni chez l'humain.

Metabolism

Le devenir métabolique de l'ester n'a pas été décrit. L'hydrolyse de la fonction acétate doit rendre le THCP, dont le métabolisme a été étudié in vitro : comme le Δ9-THC, il produit des marqueurs de phase I hydroxylés puis carboxylés sous l'action des cytochromes P450, la longueur de la chaîne latérale déplaçant l'équilibre entre ces deux voies, l'allongement réduisant la formation des dérivés hydroxylés et favorisant la conversion vers les dérivés carboxylés.

Toxicology and risks

Les données humaines manquent : ni essai clinique, ni série de cas propre au THCP-O. Deux signaux méritent d'être retenus. Le chauffage des acétates de cannabinoïdes produit du cétène, un gaz toxique pour le poumon, mis en évidence pour les acétates de Δ8-THC, de CBN et de CBD dans des conditions de vapotage ; une revue ultérieure souligne que le risque dépend du dispositif et de la température atteinte et qu'il reste mal quantifié. Par ailleurs, le THCP libéré est un agoniste CB1 de très forte affinité : un cas de psychose avec passage à l'acte auto-agressif a été publié après une prise orale unique de 8 mg chez un consommateur pourtant habitué au THC. La teneur réelle des produits du marché gris est en outre très variable d'un échantillon à l'autre, ce qui rend l'exposition imprévisible.

Detection and analysis

Le THCP-O s'identifie par chromatographie en phase gazeuse ou liquide couplée à la spectrométrie de masse, avec confirmation structurale par résonance magnétique nucléaire ; c'est selon ce schéma qu'il a été caractérisé dans des huiles et un produit végétal analysés au Japon, puis repéré dans des saisies allemandes par chromatographie liquide couplée à la spectrométrie de masse en tandem. Les immunoessais THC courants sont mal adaptés : la réactivité croisée mesurée est de 3 % pour l'acétate de THC et de 0,5 % pour le THCP, ce qui expose à des résultats faussement négatifs si aucune méthode ciblée n'est employée.
