Pharmacology
(9R)-HHC is the 9R epimer of hexahydrocannabinol and the pharmacologically active constituent of marketed HHC, which is always a mixture of variable proportions of the 9R and 9S epimers. It activates the human CB1 receptor as a partial agonist, with a potency about five times lower than that of JWH-018 in vitro, while the 9S epimer is markedly less potent at comparable efficacy. This difference stems from the conformation adopted in the active site: the 9R form places its C9 methyl in a favourable equatorial position, where the 9S form oscillates between two conformations, one of which degrades binding. Preclinical studies attribute Δ9-THC-type properties to (9R)-HHC, but the human pharmacology of HHC remains to be established. HHC has been listed in Schedule II of the 1971 Convention since 6 December 2025 and classified as a narcotic in France since 2023.

Route of preparation (chemical process)
Route of production: (9R)-HHC is not a product of plant biosynthesis. Industrial manufacture starts from cannabidiol extracted from hemp, cyclised into a mixture of Δ8- and Δ9-THC and then subjected to catalytic hydrogenation. This reduction creates a stereogenic centre at C9 and simultaneously produces both the 9R and 9S epimers. The HHC sold commercially is therefore not a single molecule but an epimeric mixture of variable proportions, whose two constituents are not pharmacologically equivalent.
Legal framework
France
In France, hexahydrocannabinol is classified on the list of narcotics by ANSM decision of 13 June 2023, and the generic clause covering derivatives of the benzo[c]chromene nucleus (ANSM decision of 22 May 2024) covers its stereoisomers. The sale and possession of (9R)-HHC are prohibited.
European Union
HHC has been listed in Schedule II of the 1971 Convention on Psychotropic Substances since 6 December 2025, becoming the first semi-synthetic cannabinoid placed under international control. The EUDA (formerly the EMCDDA) previously monitored it as an emerging new psychoactive substance, and several Member States had already classified it at national level.

Cannabinoïde semi-synthétique de type dibenzopyrane (benzo[c]chromène), épimère 9R de l'hexahydrocannabinol. Le cycle cyclohexane est saturé, contrairement au Δ9-THC qui porte une double liaison en position 9 : cette hydrogénation confère au HHC sa stabilité chimique. En C9, le méthyle adopte la configuration R.
Pharmacokinetics
Molécule lipophile de la famille du THC, le (9R)-HHC suit une distribution tissulaire attendue proche de celle du Δ9-THC. La pharmacologie humaine du HHC, y compris ses paramètres pharmacocinétiques, reste à documenter : aucune étude clinique dédiée à l'épimère isolé n'est publiée.
Metabolism
Le métabolisme animal du HHC est partiellement élucidé ; le métabolisme humain reste incomplètement caractérisé. Des métabolites hydroxylés du HHC ont été identifiés dans l'urine humaine, et des travaux de profilage métabolique in vitro ont porté sur le HHC et ses analogues semi-synthétiques.
Toxicology and risks
Aucune étude de sécurité humaine dédiée au (9R)-HHC isolé n'est disponible. Le composé étant un agoniste partiel du CB1 de type THC, les effets indésirables attendus rejoignent ceux des cannabinoïdes psychoactifs. La composition variable des produits, en particulier le ratio 9R sur 9S, rend la dose active imprévisible pour le consommateur. Aucune dose humaine caractérisée ne peut être proposée.
Detection and analysis
La séparation des épimères 9R et 9S est un enjeu documenté du contrôle des produits. Des méthodes bioanalytiques stéréosélectives par LC-MS/MS permettent de quantifier séparément le (9R)- et le (9S)-hexahydrocannabinol ainsi que leurs métabolites dans le sang et le fluide oral. Les immunoessais cannabis usuels ne sont pas conçus pour ces composés.
