- La cannabichromanone a été décrite pour la première fois en 1975 par Friedrich-Fiechtl et Spiteller. Trois dérivés supplémentaires, les cannabichromanones B, C et D, ont été isolés et caractérisés en 2008 par l'équipe du National Center for Natural Products Research de l'Université du Mississippi.

Pharmacology
Cannabichromanone is a minor cannabinoid of Cannabis sativa L., first described in 1975 and then joined in 2008 by three congeners, cannabichromanones B, C and D, isolated from cannabis with a high Δ9-THC content. Its chromanone skeleton, bearing a ketone function, sets it clearly apart from the benzo[c]chromene core of the classical psychoactive cannabinoids, and no affinity for the CB1 or CB2 receptors has been measured to date. The only documented biological activities come from the 2008 screening: no antimicrobial activity, moderate antimalarial activity for congeners A and C, moderate antileishmanial activity for all four, and strong free-radical scavenging power in the DPPH test. Its abundance in the plant is too low to credit it with any role in the effects of cannabis.

Biosynthetic pathway
Biosynthetic pathway: cannabichromanone derives from the cannabichromene branch of the cannabigerol pathway. Its formation involves an oxidation of the CBC-type skeleton leading to the ketonic chromanone core. Its very low abundance in the plant is consistent with a secondary oxidation product rather than with a primary metabolite.

France
Cannabichromanone is not listed by name on the French list of narcotics. Its chromanone core does not match the benzo[c]chromene core covered by the generic clause of the ANSM decision of 22 May 2024. Its presence in trace amounts in a product remains governed by the regulations applicable to hemp.
European Union
Not listed in the European Union narcotics schedules. Concentrated preparations intended for food use fall under Regulation (EU) 2015/2283 on novel foods (Novel Food).

Cannabinoïde à squelette chromanone (2,2-diméthylchromén-4-one), oxygéné et cétonique, structurellement distinct des cannabinoïdes classiques de type benzo[c]chromène. Il porte une chaîne pentyle en C7 et une chaîne 3-oxobutyle en C3. Quatre congénères sont décrits : cannabichromanones A, B, C et D.

Toxicology and risks
Aucune étude de toxicologie dédiée n'est disponible. Les tests menés en 2008 sur les quatre congénères n'ont révélé aucune activité antimicrobienne. Une activité antipaludique modérée a été mesurée pour la cannabichromanone A (IC50 de 3,7 à 3,8 µg/mL) et pour la cannabichromanone C (IC50 de 3,4 à 4,7 µg/mL), ainsi qu'une activité antileishmanienne modérée pour les quatre composés (IC50 de 9,0 à 14,0 µg/mL). Les quatre congénères ont montré un fort effet de piégeage des radicaux libres au test DPPH.
Detection and analysis
La cannabichromanone se détecte par chromatographie liquide couplée à la spectrométrie de masse dans les extraits végétaux. Sa caractérisation structurale a reposé sur le dichroïsme circulaire électronique et l'analyse des esters de Mosher pour établir la configuration absolue.
