- Décrite en 2021 par Shan Jiang, Christos Iliopoulos-Tsoutsouvas, Spyros P. Nikas, Alexandros Makriyannis et leurs collaborateurs, dans le Journal of Medicinal Chemistry. Le travail portait sur une série de sondes pharmacologiques bâties sur un squelette hexahydrocannabinol et fonctionnalisées par un isothiocyanate électrophile, un azide photoactivable ou un groupe cyano polaire, ces fonctions pouvant être combinées pour viser deux sites du récepteur. Le composé 27a de la série a reçu le code de laboratoire AM-11245 ; les auteurs l'ont qualifié de mégagoniste, terme employé dans ce laboratoire pour les ligands produisant une activation maximale du récepteur, et l'ont présenté comme l'un des agonistes cannabinoïdes les plus puissants décrits à ce jour. La molécule n'a jamais fait l'objet d'un développement pharmaceutique.

In plain terms
AM-11245 is a molecule made in a university laboratory in order to study the receptors on which cannabis acts. It does not exist in the plant and is used in no cannabidiol product: it is a research reagent, not a consumer product. It binds to the same receptors as THC, but activates them far more strongly and at much lower concentrations; what is known comes from experiments in cells and in mice, and human data are lacking.

Pharmacology
AM-11245 is a pharmacological probe of the AM series, built on a hexahydrocannabinol skeleton whose side chain is terminated by a nitrile and whose 9-position bears a hydroxymethyl group. Its established characteristic is potency: an affinity of 0.4 nM at rat brain CB1 and at human CB2, and a median effective concentration of 0.088 nM at human CB1 in a cyclic AMP assay, with a maximal effect of 103 % of that of CP-55,940. In other words, it is a full agonist at the CB1 receptor, where delta-9-THC is only a partial agonist, and this full activation combined with sub-nanomolar potency is what sets the molecule apart from the cannabinoids of hemp. In the mouse, the original publication describes antinociception dependent on the quantity administered, a fall in body temperature still measurable twenty-four hours after a single injection, a marked reduction in locomotor activity and a slowing of transit assessed by colonic expulsion. Beyond this mechanism and these animal observations, the file is empty: no pharmacokinetic study, no work on metabolism, no regulatory toxicology, no clinical trial. Human data are entirely lacking, and nothing warrants attributing any therapeutic interest to this molecule.

Legal framework
France
In France, AM-11245 is not listed under its own name on the narcotics list maintained by the ANSM. It does, however, fall under the generic clause of annex IV of the decree of 22 February 1990, introduced by the decision of 22 May 2024 and supplemented by that of 3 June 2024, which classifies as a narcotic any substance derived from the benzo[c]chromene nucleus, not, partially or fully hydrogenated on ring A, substituted at position 1 by a hydroxyl function, esterified or not, or by an alkoxy group, at position 3 by an alkyl, cyanoalkyl, haloalkyl, alkenyl, alkynyl or alkoxy chain, itself substituted or not by alkyl groups, at position 6 by one or two alkyl groups, and at position 9 by a ketone, alkyl, hydroxyalkyl or alkoxy function. AM-11245 matches this definition point by point: a benzo[c]chromene nucleus fully hydrogenated on ring A, a phenol at position 1, a dimethylated cyanoalkyl chain at position 3, two methyl groups at position 6, a hydroxymethyl group at position 9. None of the exclusions provided for by the clause, namely CBN, CBNA, THCV, THCVA and THCA, applies. This is therefore a classification by generic clause and not a listing by name. The older clause, which covers tetrahydrocannabinols, their esters, their ethers and their salts, does not apply here: the molecule is a hexahydro derivative, neither an ester nor an ether of THC. Production, possession, transfer and use are criminally punishable.
European Union
No control specific to the European Union: AM-11245 does not appear in the annex to Framework Decision 2004/757/JHA and has been the subject of no risk assessment by the European drugs monitoring agency, the literature consulted reporting no appearance of this molecule on the market for new psychoactive substances. Internationally, it is listed neither in the 1961 Single Convention nor in the 1971 Convention, and it does not appear in the reviews of the WHO Expert Committee on Drug Dependence. Its status nevertheless varies from one Member State to another: several countries apply, as France does, generic clauses covering derivatives of the benzo[c]chromene nucleus, so that the absence of a European measure does not amount to lawfulness at national level.

La molécule fait partie des sondes fonctionnalisées de la série AM : elle correspond à la variante à groupe cyano des composés de la même série porteurs d'un isothiocyanate, dont l'AM-841.
Pharmacokinetics
Aucune étude de pharmacocinétique n'a été publiée : ni absorption, ni distribution, ni demi-vie ne sont chiffrées, chez l'animal comme chez l'être humain. Le seul indice de durée disponible vient des essais chez la souris, où l'hypothermie consécutive à une injection unique était encore mesurée vingt-quatre heures plus tard, ce qui oriente vers une action prolongée. Les auteurs indiquent que la molécule agit aussi bien au niveau central qu'en périphérie.
Metabolism
Aucune étude de métabolisme n'a été publiée pour cette molécule : les voies de transformation, les métabolites formés et leur activité éventuelle sur les récepteurs cannabinoïdes restent inconnus.
Toxicology and risks
Aucune toxicologie réglementaire, aucun essai clinique, aucune donnée humaine. Les seuls signaux disponibles proviennent des essais chez la souris rapportés dans la publication princeps : baisse de la température corporelle après une injection unique, encore mesurable à vingt-quatre heures, réduction marquée de l'activité locomotrice et ralentissement du transit intestinal évalué par l'expulsion colique, trois observations que la base ChEMBL indexe parmi les effets indésirables. Le profil de risque tient à la combinaison de deux traits : l'activation complète du récepteur CB1, là où le delta-9-THC reste partiel, et une puissance sub-nanomolaire qui rend toute exposition humaine imprévisible. Il s'agit d'un réactif de laboratoire, sans aucune place dans un usage humain.
Detection and analysis
Aucune méthode analytique dédiée n'a été publiée, et la littérature consultée ne rapporte aucune identification de cette molécule dans des saisies ni dans des prélèvements biologiques humains. Elle n'est donc pas recherchée en routine par les laboratoires de toxicologie, et les seules données expérimentales la concernant proviennent d'essais de liaison sur récepteurs et de travaux chez l'animal.
