Pharmacology

CUMYL-TSINACA, whose systematic name is 1-(4-methylphenylsulfonyl)-N-(2-phenylpropan-2-yl)indazole-3-carboxamide, is a synthetic cannabinoid distinguished by a tosyl group on the nitrogen at position 1 of the indazole ring. Described in 2022 by Pulver et al. from products seized in Germany, it behaves as a full agonist at CB1 (Emax of 228 %), but with a very low affinity (Ki of 292 nM) and low functional activity (EC50 of 31 µM). The authors judge its low thermal stability and low potency unfavourable for psychoactive use, which does not prove that it is harmless. No human, pharmacokinetic or toxicological data are published in the sources consulted.

History

Source
A fully synthetic molecule, absent from the cannabis plant. According to Pulver et al. (2022), it belongs to the newer synthetic cannabimimetics, a family that forms the largest category of new psychoactive substances in the European Union. It was identified in illicit products seized in Germany.

Origin (pharmaceutical research → illicit market)

No biosynthetic pathway: a chemically synthesised molecule that exists in no living organism. It belongs to the class of indazole-3-carboxamides, bearing a tosyl group on the N1 nitrogen and a cumyl group on the amide.
