Pharmacology
JJKK 048 is a synthetic triazole urea that inhibits monoacylglycerol lipase (MAGL), the enzyme responsible for ending the action of 2-arachidonoylglycerol (2-AG). First described in 2013 by a team at the University of Eastern Finland in Kuopio, it inhibits human MAGL with an IC50 below 0.4 nM (Aaltonen et al., 2013). ChEMBL data from 2014 give an IC50 above 4 000 nM on human FAAH (Korhonen et al., 2014). In laboratory animals, the 2016 study reports a massive rise in brain 2-AG without change in anandamide, and analgesia in the writhing test at a low dose, without cannabimimetic effects (Aaltonen et al., 2016). The abstracts consulted report no affinity constant at CB1 or CB2, and no pharmacokinetic or clinical data in humans.

History
- Source
- Fully synthetic molecule, part of a series of piperazine and piperidine triazole ureas developed at the University of Eastern Finland in Kuopio. No natural occurrence is known: it is a laboratory compound.
Origin (research tool)
No biosynthetic pathway: a chemically synthesised molecule belonging to the class of piperidine triazole ureas.
Legal framework
- European Union
No harmonised European Union control measure is known to target this molecule, according to the sources consulted; control, where it exists, falls to national legislation. It is also absent from the INCB Green List 2026, which lists the substances subject to the 1971 Convention.
- France
JJKK 048 is not named in any annex of the decree of 22 February 1990 fixing the list of narcotics, as consolidated by the ANSM on 16 February 2026. It is not a tetrahydrocannabinol, nor an ester, ether or salt of one: the generic clause covering them therefore does not capture it. Its structure, a piperidine triazole urea with no indole, indazole, naphthyl, cyclohexylphenol, thiazole or pyrrolopyridine core, meets none of the generic family definitions in that list. It is therefore unscheduled in France. This absence of classification does not amount to an authorisation: the sources consulted describe no use outside research, and a later classification by ministerial order remains possible.
