Pharmacology

JTE-7-31 is a synthetic ligand of cannabinoid receptors with an isoindolinone structure: 2-[2-(4-hydroxyphenyl)ethyl]-5-methoxy-4-(pentylamino)isoindolin-1-one. Described in patents filed by Japan Tobacco in 1996 and 1997, it is presented by US patent 6 017 919 (published in 2000) as one of the compounds of a series of selective ligands of peripheral cannabinoid receptors. That patent reports a Ki of 0.088 nM at CB2 (rat splenocytes) and 11 nM at CB1 (rat cerebellum), a ratio of 125. It also reports that, in mice, the compound reduces carrageenan-induced paw oedema, with a median effective dose of 0.012 mg/kg orally. No human study and no pharmacokinetic profile are published in the sources consulted, and the agonist or antagonist nature of the molecule is not measured there.

History
