Isolé et caractérisé en 2015 par Radwan, ElSohly et leurs collègues de l'université du Mississippi, à partir des parties aériennes d'un Cannabis sativa à forte teneur en THC. La même publication décrit ensemble les deux épimères 10α et 10β ainsi que plusieurs autres cannabinoïdes hydroxylés mineurs, et fournit les premières et, à ce jour, seules mesures d'affinité pour les récepteurs CB1 et CB2.

A natural constituent of Cannabis sativa, present only in trace amounts: it could be obtained only after extensive chromatographic fractionation of a high-THC variety, which rules out any appreciable presence in an ordinary flower or extract. The molecule derives from the Δ8-THC skeleton and remains accessible chemically from it, but it has not been reported among the impurities of the Δ8-THC products analysed on the North American market, nor among the semi-synthetic cannabinoids recorded on the European market.

Pharmacology

10α-OH-Δ8-THC retains the cannabinoid pharmacophore in full, a phenol at position 1 and a pentyl chain at position 3, with an additional hydroxyl at position 10 on the Δ8 skeleton. Radwan and colleagues, who isolated it from cannabis in 2015, measured an affinity of 31 nM for CB1 and 30 nM for CB2, comparable to that of Δ9-THC tested in the same assay, and described it as the most active of the minor cannabinoids in this series in the tetrad test in mice, with hypolocomotion, catalepsy, hypothermia and antinociception. The 10β epimer, isolated in the course of the same work, loses more than twentyfold in affinity at CB1 and about a hundredfold at CB2: the orientation of the hydroxyl decides the activity. Beyond this publication the literature is very thin; functional efficacy at CB1 has never been measured, no secondary target has been explored, and human data are entirely lacking.

Biosynthetic pathway

The plant first forms cannabigerolic acid, converted by THCA synthase into tetrahydrocannabinolic acid, then into Δ9-THC and Δ8-THC. 10α-OH-Δ8-THC corresponds to this Δ8 skeleton bearing an additional hydroxyl on carbon 10. No enzyme dedicated to this hydroxylation has been identified, and non-enzymatic formation through slow oxidation during drying and ageing of the plant remains the simplest explanation.

La configuration (6aR,10R,10aR) définit l'épimère alpha ; l'épimère beta correspondant, le 10β-OH-Δ8-THC, est une molécule distincte aux propriétés très différentes.

Pharmacokinetics

Aucune donnée de pharmacocinétique n'a été publiée, ni chez l'être humain ni chez l'animal : absorption, demi-vie, distribution et biodisponibilité restent inconnues. Les seules administrations documentées sont des injections intrapéritonéales chez la souris dans un test comportemental aigu, qui renseignent sur l'effet observé mais pas sur le devenir de la molécule dans l'organisme.

Metabolism

Le devenir métabolique du 10α-OH-Δ8-THC n'est pas documenté. Aucune étude ne l'identifie comme métabolite du Δ8-THC chez le mammifère, les voies décrites pour ce dernier portant sur les positions 11 et 7 ; la littérature le présente comme un constituant de la plante et non comme un produit de l'organisme. La conjugaison éventuelle de l'hydroxyle en position 10 n'a pas été étudiée.

Toxicology and risks

Aucune étude de toxicologie n'a porté sur cette molécule et aucun cas d'intoxication humaine ne lui est attribué. Le risque envisageable découle de son profil pharmacologique : une affinité pour CB1 comparable à celle du Δ9-THC laisse attendre les mêmes effets indésirables que le THC, anxiété, tachycardie, désorientation, sans qu'aucune mesure chez l'être humain ne vienne le confirmer ni l'infirmer. La molécule n'ayant pas été signalée dans les produits du commerce, l'exposition réelle paraît pour l'instant très limitée.

Detection and analysis

Aucune méthode de dépistage de routine ne cible le 10α-OH-Δ8-THC, et sa réactivité croisée avec les tests immunochimiques urinaires n'a jamais été mesurée. Son identification dans la plante a reposé sur un fractionnement chromatographique poussé, complété par spectrométrie de masse haute résolution et résonance magnétique nucléaire, seule combinaison capable de séparer les épimères 10α et 10β et de les distinguer. Aucune donnée de détection en matrice biologique n'a été publiée.
