- Le 2-monopalmitine est un glycéride connu de longue date comme produit de la digestion des matières grasses. Son entrée dans la pharmacologie du système endocannabinoïde date de 1998 : Shimon Ben-Shabat, Raphael Mechoulam et leurs collègues de l'Université hébraïque de Jérusalem, associés aux équipes de Vincenzo Di Marzo à Naples et de Zvi Vogel à Rehovot, décrivent dans European Journal of Pharmacology les esters glycéridiques qui accompagnent le 2-arachidonoylglycérol dans la rate, l'intestin et le cerveau. Le 2-palmitoylglycérol et le 2-linoléoylglycérol comptaient parmi les plus abondants. Sans activité propre dans l'ensemble des tests employés, ils augmentaient la liaison du 2-AG et ses effets, ce que les auteurs ont baptisé effet d'entourage.

It is endogenous in mammals, where it accompanies 2-arachidonoylglycerol in the spleen, the intestine and nervous tissue. It also has a direct dietary origin: pancreatic lipase cleaves triglycerides at positions 1 and 3 and leaves the central monoester intact, so that the palmitate carried at the sn-2 position of fats, well represented in breast milk and in restructured OPO-type oils, reaches the intestine as 2-palmitoylglycerol. The mono- and diglycerides of fatty acids to which it belongs are moreover used as a food emulsifier under the code E 471. Laboratory samples come from synthesis.

2-palmitoylglycerol is a small fatty molecule that the body makes and that the digestion of fats also releases in the intestine. It resembles 2-AG, one of the cannabinoids the body produces itself, but it does not bind to the cannabis receptors. It was long presented as a companion that strengthens the action of 2-AG; more recent work does not find this effect, and no study has been conducted in human beings.

Pharmacology
2-palmitoylglycerol is the monoester of glycerol and palmitic acid at the central carbon, a saturated congener of 2-arachidonoylglycerol present in the spleen, the intestine and nervous tissue, and released in quantity by the digestion of triglycerides whose palmitate occupies the sn-2 position. It shows no measurable affinity for CB1 or for CB2. Its prominence comes from the 1998 article that introduced the notion of an entourage effect: combined with 2-AG, it increased the latter's binding and its inhibition of adenylate cyclase while remaining inactive on its own, a potentiation attributed to saturation of the degradative enzymes. This finding did not survive the re-evaluation published in 2016, in which the compound potentiated none of the parameters measured and behaved as a functional antagonist of 2-AG signalling. The best-documented aspect is today metabolic rather than cannabinoid: the molecule stimulates GLP-1 secretion by enteroendocrine cells through GPR119 and GPR40, in cells and in mice. Human data are lacking, no pharmacokinetic, efficacy or safety study addressing the molecule given on its own; the entourage effect so often invoked in the hemp trade cannot claim support from this compound.

Biosynthetic pathway (in vivo)
Two pathways converge on the same molecule. Within cells, hydrolysis of phosphoinositides by phospholipase C releases a diacylglycerol that diacylglycerol lipase cleaves at position 1, leaving a 2-monoacylglycerol whose chain depends on the fatty acid esterified at the central carbon, here palmitic acid. In the intestinal lumen, pancreatic lipase produces the same compound from dietary triglycerides carrying palmitate at the sn-2 position. The tissue pool therefore reflects both the remodelling of membrane phospholipids and lipid intake.
Legal framework
France
An endogenous substance and a common food constituent, not listed on the French lists of narcotics and psychotropic substances. The decree of 22 February 1990 covers in annex IV the tetrahydrocannabinols, their esters, ethers and salts as well as the salts of the aforementioned derivatives: 2-palmitoylglycerol is an ester of palmitic acid and glycerol, with no structural kinship to tetrahydrocannabinol, so it falls neither under this generic clause nor under any ANSM listing by name. No prohibition targets it, and the mono- and diglycerides of fatty acids of which it is part are authorised as a food additive under the code E 471.
European Union
No control measure at European Union level. The substance does not appear among the new psychoactive substances notified or assessed by the EUDA, it has been the subject of no Council implementing decision and is listed in no schedule of the 1961 and 1971 United Nations conventions. It circulates on the one hand as a research reagent and analytical standard, and on the other as a constituent of mono- and diglycerides of fatty acids (E 471), an authorised additive for which the EFSA scientific panel concluded in 2017 that no numerical acceptable daily intake was needed and that it raised no concern at the reported uses and use levels.

Monoacylglycérol saturé : l'acide palmitique, chaîne linéaire à seize carbones sans double liaison, est estérifié sur le carbone central du glycérol, dont les deux fonctions alcool restent libres. La molécule appartient aux médiateurs lipidiques apparentés au système endocannabinoïde, aux côtés du 2-AG et des autres 2-monoacylglycérols, et non aux cannabinoïdes terpénophénoliques du chanvre. La littérature des corps gras la désigne aussi sous le nom de 2-monopalmitine.
Pharmacokinetics
Aucune donnée humaine issue d'une administration de la molécule seule. Le composé suit le devenir des monoacylglycérols alimentaires : capté par l'entérocyte à partir des micelles mixtes, il est en grande partie réestérifié en triglycéride puis exporté dans les chylomicrons, ce qui explique que le palmitate porté par la position centrale soit mieux absorbé que l'acide palmitique libre, lequel forme des savons calciques peu solubles. La fraction qui échappe à cette réestérification est hydrolysée en glycérol et en acide palmitique, deux composants du pool lipidique général. Aucune concentration plasmatique de référence n'est établie, en partie parce que le dosage se heurte à une contamination de fond importante.
Metabolism
L'inactivation repose sur les hydrolases à sérine du système endocannabinoïde, au premier rang desquelles la monoacylglycérol lipase, qui accepte largement les monoacylglycérols quelles que soient la longueur et l'insaturation de la chaîne et libère ici du glycérol et de l'acide palmitique. Les hydrolases ABHD6 et ABHD12, également actives sur les monoacylglycérols, participent à cette dégradation dans d'autres compartiments cellulaires. Cette compétition de substrat avec le 2-AG constituait précisément le mécanisme proposé pour l'effet d'entourage. Les produits d'hydrolyse rejoignent le métabolisme lipidique ordinaire, bêta-oxydation ou réincorporation dans les glycérolipides. Aucun métabolite spécifique, oxydé ou conjugué, n'a été décrit pour ce composé.
Toxicology and risks
Aucun signal de toxicité propre n'est documenté, et aucune étude clinique ne porte sur la molécule isolée. Le contexte d'exposition est celui d'un constituant alimentaire : les mono- et diglycérides d'acides gras, dont fait partie le 2-palmitoylglycérol, sont ingérés quotidiennement avec les graisses et employés comme émulsifiant sous le code E 471. Lors de la réévaluation de 2017, le groupe scientifique de l'EFSA a conclu qu'aucune dose journalière admissible chiffrée n'était nécessaire et que l'additif ne soulevait pas de préoccupation aux usages et niveaux d'emploi déclarés, en retenant que l'hydrolyse digestive libère du glycérol et des acides gras déjà évalués par ailleurs. Cette conclusion vise un mélange alimentaire et non l'administration pharmacologique de la molécule pure, pour laquelle les données humaines manquent. Aucun cas d'intoxication n'est rapporté par les centres antipoison ni par les systèmes d'alerte précoce européens.
Detection and analysis
Il n'existe pas de recherche médico-légale de ce composé, qui n'est contrôlé nulle part et ne figure dans aucun criblage toxicologique de routine. Sa mesure relève de la lipidomique, par chromatographie liquide couplée à la spectrométrie de masse en tandem après extraction. Deux difficultés dominent. La première est la migration de l'acyle : le monoester central s'isomérise spontanément vers la forme portée par un carbone terminal, ce qui impose des conditions d'extraction ménagées et la séparation chromatographique des deux isomères. La seconde est la contamination de fond, les monoglycérides palmitiques étant omniprésents dans les plastiques, les solvants et les milieux de laboratoire : une méthode validée de quantification des endocannabinoïdes plasmatiques a dû retirer le 2-palmitoylglycérol de son panel, les niveaux de blanc dépassant les teneurs endogènes attendues.
