- La molécule a été synthétisée et caractérisée par Samuel Banister, Michael Kassiou, Iain McGregor et leurs collaborateurs, dans un travail publié en 2015 dans ACS Chemical Neuroscience consacré à huit carboxamides d'indole et d'indazole porteurs de chaînes valinamide ou tert-leucinamide. Les auteurs partaient du constat que ces composés étaient rencontrés avec une fréquence croissante par les laboratoires médico-légaux et associés à des effets indésirables graves, mais qu'ils étaient pour la plupart inédits dans la littérature scientifique au moment de leur découverte, sans aucune pharmacologie connue.

Pharmacology
At the CB1 receptor, 5F-ADB-PINACA is the most potent compound of a series that Samuel Banister, Michael Kassiou and their co-workers characterised in 2015 in order to fill a gap: these indole and indazole carboxamides were arriving in growing numbers in forensic laboratories, associated with serious adverse effects, without any pharmacology being known for them. The eight compounds tested all behave as high-potency agonists, with EC50 values from 0.24 to 21 nM at CB1 and from 0.88 to 15 nM at CB2, and this one lies at the most potent bound of that range, whereas tetrahydrocannabinol is itself only a partial agonist. A nomenclature caution is called for before any reading of the literature: the terminal function here is a primary amide, whereas the corresponding methyl ester compound, 5F-MDMB-PINACA, circulates under the shorthand 5F-ADB. Two different molecules therefore bear very similar names, and the confusion extends even into databases. The second issue is analytical, and it was addressed by Jérémy Carlier, Marilyn Huestis and their co-workers in 2017: this molecule and its non-fluorinated analogue, ADB-PINACA, do not have the same legal status depending on the country, which makes it necessary to distinguish between the two forms of use, even though their major metabolites are positional isomers. The authors recommend the 5-hydroxypentyl and pentanoic acid metabolites of ADB-PINACA as markers of intake of the fluorinated compound, while specifying that optimised chromatography and the monitoring of specific product ions remain indispensable. No controlled study in human beings exists.

5F-ADB-PINACA is classified as a narcotic in France, and it is so under its own name. Annex IV of the decree of 22 February 1990 mentions it explicitly, in the form "5F-ADB-PINACA or N-(1-Amino-3,3-dimethyl-1-oxobutan-2-yl)-1-(5-fluoropentyl)-1H-indazole-3-carboxamide", within the generic family of derivatives of 3-carboxamide indazole. The official designation incidentally removes the ambiguity with the methyl ester 5F-MDMB-PINACA: it is indeed the primary amide that is covered under this name. The corresponding generic clause covers molecules bearing on the indazole nitrogen a haloalkyl substituent and, on the carboxamide bridge nitrogen, a group of the 1-amino-1-oxo-butan-2-yl type substituted at position 3 by one or two alkyl groups: the molecule matches it point by point. Production, possession, transfer, transport and use fall under the criminal law on narcotics.
European Union
No harmonised control measure at Union level covers 5F-ADB-PINACA: the substance does not appear in the annex to Framework Decision 2004/757/JHA, to which other, more recent carboxamides have been added by delegated acts. Control remains national and varies from one member state to another; the authors of the metabolism study noted, moreover, that this molecule and its non-fluorinated analogue, ADB-PINACA, had a different legal status depending on the country, which made the ability to distinguish between the two forms of use critical. France has listed it by name in its annex IV.

Indazole-3-carboxamide à chaîne tert-leucinamide : noyau indazole portant une chaîne 5-fluoropentyle sur son azote en position 1, relié en position 3, par une fonction carboxamide, à un motif 1-amino-3,3-diméthyl-1-oxobutan-2-yle dérivé de la tert-leucine. Un point de nomenclature demande de la vigilance : la fonction terminale est ici un amide primaire, et non un ester méthylique. Le composé à ester méthylique correspondant est le 5F-MDMB-PINACA, largement diffusé sous le raccourci 5F-ADB, si bien que deux molécules différentes circulent sous des noms voisins. La confusion est courante et n'est pas anodine, les deux composés n'ayant ni la même formule brute ni le même profil métabolique.
Pharmacokinetics
Aucune étude de pharmacocinétique humaine n'est publiée : biodisponibilité, demi-vie plasmatique et volume de distribution restent inconnus. Les seules données in vivo de la série de caractérisation concernent l'AB-FUBINACA et l'AB-PINACA, évaluées chez le rat par biotélémétrie, et ne peuvent être transposées à cette molécule. La stabilité du carboxamide, sans point de coupure hydrolytique comparable à l'ester des carboxylates quinoléinyles, distingue cette génération de la précédente et allonge la fenêtre de détection des métabolites. La voie d'exposition documentée est l'inhalation de la fumée d'un support végétal imprégné, avec un délai d'action de quelques minutes.
Metabolism
Le métabolisme humain a été caractérisé en 2017 sur hépatocytes humains cryoconservés par chromatographie liquide et spectrométrie de masse haute résolution, dans un travail conduit en parallèle avec l'analogue non fluoré. Douze métabolites majeurs ont été identifiés pour cette molécule, contre dix-neuf pour l'ADB-PINACA. La réaction dominante est la défluoration oxydative suivie de carboxylation, voie caractéristique des chaînes fluoropentylées, là où l'analogue non fluoré subit surtout des hydroxylations de la chaîne pentyle, leur oxydation en cétone et des glucuroconjugaisons. Les auteurs recommandent le 5-hydroxypentyl et l'acide pentanoïque de l'ADB-PINACA comme marqueurs d'une prise du composé fluoré. Le point délicat est que les métabolites principaux des deux substances sont des isomères de position : leur distinction impose de suivre des ions produits spécifiques et d'optimiser les conditions chromatographiques.
Toxicology and risks
Aucune étude contrôlée chez l'être humain n'est publiée et il n'existe pas de donnée de toxicité systématique propre à cette molécule. Deux éléments cadrent néanmoins le risque. D'abord la puissance : ce composé est le plus puissant de sa série sur CB1, dans une famille dont les auteurs de l'étude de métabolisme rappellent qu'elle compte parmi les cannabinoïdes de synthèse les plus puissants testés à ce jour, avec plusieurs cas d'intoxication sévère documentés. Ensuite l'absence de plafond d'effet, qui distingue ces agonistes complets du tétrahydrocannabinol et explique les tachycardies, convulsions, vomissements, agitations sévères et décompensations psychiatriques aiguës rapportés dans cette catégorie. La poudre est pulvérisée sur un support végétal, à répartition très inégale d'une prise à l'autre, ce qui expose à des surdoses accidentelles.
Detection and analysis
L'identification repose sur la chromatographie couplée à la spectrométrie de masse, en phase gazeuse ou liquide, par comparaison à un étalon de référence certifié. Les immunoessais destinés au cannabis ne réagissent pas à cette structure, et un dépistage négatif ne vaut pas absence. Dans l'urine, la recherche porte sur le 5-hydroxypentyl et l'acide pentanoïque de l'ADB-PINACA. La difficulté centrale est de départager une prise de 5F-ADB-PINACA d'une prise d'ADB-PINACA, puisque les métabolites majeurs des deux molécules sont des isomères de position, de masse identique : cela exige le suivi d'ions produits spécifiques et des conditions chromatographiques optimisées, faute de quoi l'attribution est impossible. Une seconde vigilance porte sur le nom lui-même, le 5F-MDMB-PINACA circulant sous le raccourci 5F-ADB, ce qui peut égarer une recherche documentaire ou une transcription de résultat.
