- La molécule porte le code SGT-25 dans la série de carboxamides de la société néo-zélandaise Stargate International. Elle a été identifiée entre mai 2014 et juin 2015 par Verena Angerer, Volker Auwärter et leurs collaborateurs de l'institut de médecine légale de Fribourg, dans trois liquides de cigarette électronique sur vingt et un achetés en ligne : la substance inconnue détectée dans ces produits a été caractérisée par résonance magnétique nucléaire. C'était la première fois qu'un cannabinoïde de synthèse de ce type était trouvé dans un tel vecteur, ce qui explique l'alias C-Liquid, repris jusque dans le texte réglementaire français. Sa pharmacologie a été précisée en 2019 par Samuel Banister et ses collaborateurs, dans une série comparant des analogues à changement de squelette.

Pharmacology
5F-CUMYL-PINACA brought synthetic cannabinoids into an unexpected vector. Between May 2014 and June 2015, the forensic medicine team in Freiburg analysed twenty-one electronic cigarette liquids bought online and detected an unknown substance in three of them, identified by nuclear magnetic resonance as this molecule. Nothing on the label mentioned it, and the alias C-Liquid, carried through even into the French regulatory text, preserves the trace of that episode. The molecule belongs to the SGT series of the New Zealand company Stargate International, where it bears the code SGT-25, and it is the fluorinated analogue of CUMYL-PINACA. Its potency is considerable: the work of Samuel Banister and co-workers published in 2019 assigns it an EC50 of 0.43 nM at CB1, the highest of the three analogues compared, ahead of 5F-CUMYL-PICA at 2.2 nM and 5F-CUMYL-P7AICA at 4.7 nM, whereas tetrahydrocannabinol is only a partial agonist of much lower potency. The terminal fluorine also has a direct analytical consequence: in the urine of a user, the hydrolytic defluorination route is very abundant, and the authors of the characterisation retained three metabolites as reliable markers. The question of the vector remains the most worrying. In a liquid intended to be vaporised, a substance active at a fraction of a nanomole is all the more poorly distributed for being diluted in a volume, and nothing informs the consumer of its presence. No clinical study exists: dose, tolerance and dependence are not characterised.

5F-CUMYL-PINACA is classified as a narcotic in France, and it is so under its own name. Annex IV of the decree of 22 February 1990 lists it in the form "5F-CUMYL-PINACA or SGT-25 or C-Liquid or 1-(5-fluoropentyl)-N-(1-methyl-1-phenylethyl)-1H-indazole-3-carboxamide", within the generic family of 3-carboxamide indazole derivatives. One drafting peculiarity deserves mention: the same name appears a second time in the text, this time in the list of 3-carboxamide indole derivatives, placed alongside CUMYL-BICA, which is nonetheless a distinct molecule. This redundancy changes nothing in substance, since the generic clause covering indazole carboxamides explicitly targets cumyl substituents on the nitrogen of the carboxamide bridge and haloalkyl substituents on the indazole nitrogen. Production, possession, transfer, transport and use fall under the criminal law on narcotics.
European Union
No harmonised control measure at Union level targets 5F-CUMYL-PINACA: the substance does not appear in the annex to Framework Decision 2004/757/JHA, to which other, more recent carboxamides have been added by delegated acts. Control remains national and is effective in many Member States by means of generic definitions covering indazole carboxamides with a cumyl head. The molecule is monitored by the Union's Early Warning System as a new psychoactive substance, its detection in electronic cigarette liquids bought online having constituted a signal of a new vector.

Indazole-3-carboxamide à tête cumyle et chaîne fluorée : noyau indazole portant une chaîne 5-fluoropentyle sur son azote en position 1, relié en position 3 à un groupement 2-phénylpropan-2-yle par une fonction carboxamide. La molécule est l'analogue fluoré du CUMYL-PINACA, le fluor terminal étant une substitution bioisostère déjà éprouvée ailleurs dans la classe, du JWH-018 vers l'AM-2201 puis du PB-22 vers le 5F-PB-22. Le carboxamide lui donne la stabilité chimique qui manquait aux carboxylates quinoléinyles.
Pharmacokinetics
Aucune étude de pharmacocinétique humaine n'est publiée : biodisponibilité, demi-vie plasmatique et volume de distribution restent inconnus. Les données humaines disponibles proviennent d'un échantillon d'urine authentique, qui a permis d'établir le profil métabolique sans que la dose ni le moment de la prise soient connus. Le vecteur documenté, un liquide de cigarette électronique, implique une absorption pulmonaire rapide, avec un délai d'action de quelques minutes. La stabilité du carboxamide distingue cette molécule des cannabinoïdes de synthèse à lien ester, dont la mère est fugace : ici, rien n'impose une hydrolyse préalable à l'absorption.
Metabolism
Le métabolisme de phase I a été caractérisé in vitro et sur un échantillon d'urine authentique par l'équipe de médecine légale de Fribourg. Six métabolites ont été identifiés. La voie distinctive est la défluoration hydrolytique, détectée avec une abondance élevée : le fluor terminal est remplacé par une fonction hydroxyle, ce qui rapproche le profil métabolique de celui du composé non fluoré et impose une séparation chromatographique soignée pour ne pas confondre les deux consommations. Trois métabolites ont été retenus par les auteurs comme marqueurs urinaires fiables. Cette convergence métabolique entre analogue fluoré et non fluoré est un problème récurrent de la classe, déjà rencontré entre le PB-22 et le 5F-PB-22.
Toxicology and risks
Aucune étude contrôlée chez l'être humain n'est publiée et il n'existe pas de donnée de toxicité systématique propre à cette molécule. Le profil de risque est dominé par la conjonction d'une puissance considérable et d'un vecteur trompeur. À une CE50 de 0,43 nM sur CB1, la quantité active est infime ; diluée dans un liquide de cigarette électronique vendu sans mention de son contenu, elle expose un consommateur à une substance qu'il n'a pas choisie et dont il ne peut estimer la dose. La classe est associée chez les consommateurs à des tachycardies, des convulsions, des vomissements, une agitation sévère et des décompensations psychiatriques aiguës. L'analogue non fluoré figure par ailleurs parmi les cannabinoïdes de synthèse qui inhibent le plus grand nombre de récepteurs non cannabinoïdes à concentration élevée, observation qui invite à ne pas réduire le risque au seul récepteur CB1.
Detection and analysis
L'identification repose sur la chromatographie couplée à la spectrométrie de masse, complétée par la résonance magnétique nucléaire lorsqu'aucun étalon n'existe encore, comme ce fut le cas lors de la caractérisation initiale dans les liquides de cigarette électronique. Les immunoessais destinés au cannabis ne réagissent pas à cette structure. Dans l'urine, la recherche porte sur les trois métabolites retenus par les auteurs de cette caractérisation, en gardant à l'esprit que la défluoration hydrolytique produit des composés proches de ceux du CUMYL-PINACA non fluoré. La détection en spectrométrie de mobilité ionique, employée pour cribler les papiers imprégnés en milieu carcéral, a été évaluée pour les cannabinoïdes à tête cumyle, avec des valeurs de mobilité réduite qui diffèrent parfois de celles rapportées antérieurement.
