La série SDB est issue des travaux de chimie médicinale de Samuel D. Banister et de ses collègues à l'université de Sydney sur les indole-3-carboxamides, dans le prolongement de l'APICA, composé vendu illicitement sous le nom 2NE1. Le 5F-SDB-006 en est l'analogue fluoré en bout de chaîne pentyle du SDB-006. Sa caractérisation pharmacologique publiée s'échelonne du milieu des années 2010 à nos jours : criblage de Hess et coll. en 2016, série stéréochimique d'Ametovski et coll. en 2020, puis mesure directe de son efficacité au CB1 par Yano et coll. en 2023.

Pharmacology

5F-SDB-006 is a cannabinoid receptor agonist of the indole-3-carboxamide type, combining a benzyl head group with a 5-fluoropentyl chain. The NCATS UNII record puts its activity at the human CB1 receptor at an EC50 of 50 nM (Ki of 71.9 nM) and at CB2 at 123 nM (Ki of 430 nM). The published work diverges markedly on affinity: Ametovski et al. (2020) report, for the benzyl series to which it belongs, CB1 Ki values ranging from 550 nM to more than 10 µM, far below the corresponding cumyl derivatives. Above all, Yano et al. (2023) measured its efficacy at CB1 directly by BRET: 5F-SDB-006 shows the lowest efficacy of all the 5F-pentylindoles tested, both for coupling to the Gi protein and for beta-arrestin 2 recruitment. That is precisely the point that matters for harm reduction. In this family, replacing the benzyl with a cumyl group or with a methyl ester is enough to turn a poorly efficacious molecule into a full agonist, or even a superagonist exceeding the CP55940 reference, whereas THC is itself only a partial agonist at CB1. It is this full agonism that makes synthetic cannabinoids dangerous, and nothing allows a consumer to tell one analogue from another in a plant mixture. Human data are entirely lacking for 5F-SDB-006.

Aucune parenté structurale avec les cannabinoïdes végétaux, qui sont des terpénophénols.

Pharmacokinetics

Aucune donnée de pharmacocinétique humaine publiée pour le 5F-SDB-006 : ni biodisponibilité, ni demi-vie, ni volume de distribution. Les cannabinoïdes de synthèse de cette famille sont très lipophiles, et Hess et coll. (2016) indiquent que la plupart des dérivés de leur série sont prédits franchir la barrière hémato-encéphalique par des outils bioinformatiques, prédiction qui ne remplace pas une mesure.

Metabolism

Aucune étude de métabolisme portant sur le 5F-SDB-006 lui-même n'a été retrouvée. Le composé parent non fluoré, le SDB-006, a fait l'objet d'une étude sur hépatocytes humains, mais ces résultats appartiennent à cet analogue et ne peuvent pas lui être transposés tels quels. Les métabolites urinaires de référence du 5F-SDB-006, indispensables au dépistage ciblé, ne sont donc pas établis dans la littérature consultée.

Toxicology and risks

Aucun cas clinique ni décès spécifiquement attribué au 5F-SDB-006 n'a été retrouvé dans la littérature. Les effets toxiques aigus décrits par l'ANSM valent comme repère de classe pour les cannabinoïdes de synthèse, pas comme description de cette molécule : troubles neurologiques et neuromusculaires (agitation, convulsions, somnolence, vertiges, paresthésies, ralentissement psychomoteur), troubles cardiovasculaires (tachycardie, palpitations, hypertension artérielle, plus rarement bradycardie ou hypotension) et troubles psychiatriques (troubles psychotiques, symptômes paranoïaques, confusion, anxiété, amnésie). L'ANSM signale également des cas de tolérance, de dépendance et de signes de sevrage, et rappelle que la littérature étrangère rapporte de nombreux décès par intoxication massive à ces produits. Le risque propre à ce marché tient à la substitution : un mélange végétal peut contenir un analogue bien plus efficace au CB1 que celui annoncé, sans aucun signe distinctif.

Detection and analysis

Le 5F-SDB-006 est enregistré au système d'alerte précoce de l'ONUDC, ce qui suppose son identification dans au moins un prélèvement transmis par un État membre. Son identification repose sur la chromatographie liquide ou gazeuse couplée à la spectrométrie de masse, avec un étalon de référence : les tests immunologiques usuels de dépistage du cannabis ne le détectent pas. Aucune méthode analytique dédiée ni cas médico-légal publié spécifiquement pour cette molécule n'ont été retrouvés.
