L'inventaire critique des phytocannabinoïdes publié par Hanuš et ses collaborateurs en 2016 range cette molécule parmi les dérivés partiellement aromatisés apparentés au cannabinol. Elle a ensuite été analysée par voie informatique par Jagannathan en 2020, puis décrite en 2024 par Caprari et ses collaborateurs parmi les THC de synthèse circulant sur le marché récréatif. Aucune publication ne documente une première isolation datée ni un auteur identifié.

Pharmacology

7,8-dihydrocannabinol sits between delta-9-THC and cannabinol: its terpene ring carries an additional double bond at 6a,10a, which brings it closer to cannabinol without fully aromatising it. This structural kinship is the only argument available to the literature, since no binding assay at the CB1 or CB2 receptors has been published: the reference review on synthetic THCs (Caprari and colleagues, 2024) leaves the affinity box empty and confines itself to indicating that the effects are expected to be close to those of delta-9-THC. A computational analysis of 2020 predicts an increased sensitivity to hepatic metabolism, without experimental validation. No human data, no toxicology study, no validated analytical method. What is established amounts to two points: it is a semi-synthesis product sold outside any control of purity, and France has classified it as a narcotic since 3 June 2024.

Route of preparation (chemical process)

No plant biosynthetic pathway is described for this molecule. Its preparation involves the oxidative dehydrogenation of delta-9-tetrahydrocannabinol, that is to say a partial aromatisation of the terpene ring which stops short of the cannabinol stage. Cannabinoïde de type CBN partiellement réduit : le noyau benzo[c]chromène porte encore deux doubles liaisons sur le cycle terpénique, en 9,10 et en 6a,10a, là où le delta-9-THC n'en compte qu'une, avec une chaîne latérale pentyle en position 3. Par son degré d'insaturation, la molécule se situe exactement entre le delta-9-tétrahydrocannabinol et le cannabinol.

Pharmacokinetics

Aucune donnée de pharmacocinétique humaine n'est publiée : ni l'absorption, ni la distribution, ni la demi-vie de cette molécule ne sont documentées. Comme les autres cannabinoïdes de ce squelette, elle est très lipophile, ce qui laisse attendre une distribution tissulaire large, mais il s'agit d'une extrapolation et non d'une mesure.

Metabolism

Une analyse informatique publiée en 2020, reprise par la revue de 2024, prédit que cette molécule serait plus sensible que le delta-9-THC au métabolisme hépatique. Aucun métabolite n'a toutefois été identifié expérimentalement, et aucune étude sur microsomes ou sur matrice biologique n'a été publiée.

Toxicology and risks

Aucune étude de toxicologie, aucun rapport de cas et aucune donnée d'addictovigilance propres au 7,8-DHC n'ont été retrouvés. L'absence de caractérisation constitue ici le principal signal de risque : les produits vendus sous ce genre d'appellation échappent à tout contrôle de pureté, peuvent contenir des résidus de delta-9-THC ou d'autres isomères, et sont parfois présentés à tort comme du CBD légal. En cas de malaise après consommation d'un tel produit, il faut appeler les secours ou un centre antipoison.

Detection and analysis

Aucune méthode analytique validée n'est publiée pour le 7,8-DHC, la revue de 2024 laissant également cette colonne vide. Les dépistages courants ciblent le delta-9-THC et son métabolite THC-COOH et ne sont pas conçus pour identifier cette molécule. Son identification supposerait une chromatographie liquide couplée à la spectrométrie de masse haute résolution, avec un étalon de référence dédié.
