Deutsch et Alexandros Makriyannis, au sein d'une série de fluorures de sulfonyle gras allant de C12 à C20 évalués comme inhibiteurs de l'hydrolyse de l'anandamide. Le brevet correspondant, déposé par l'Université du Connecticut avec une priorité au 31 mai 1996, mentionne explicitement le fluorure de stéarylsulfonyle parmi les composés testés. Le code AM-381 provient de la nomenclature interne du laboratoire Makriyannis, la même qui a donné AM-374 et AM-404.

Pharmacology
AM-381, or stearylsulfonyl fluoride, belongs to the first generation of FAAH inhibitors developed at the end of the 1990s. The principle is straightforward: the sulfonyl fluoride function reacts with the catalytic serine of the enzyme and blocks it covalently, hence irreversibly; anandamide is no longer hydrolysed and its local concentrations rise. In the series published in 1997, the C12 to C18 analogues inhibited anandamide amidase at nanomolar concentrations, and the longest chains, including the C18 one, proved distinctly more selective for the enzyme than for the CB1 receptor. What the literature does not establish counts just as much: no behavioural study, no pharmacokinetic data and no human data bear on AM-381 itself. The results obtained in animals and frequently cited in this field concern the C16 analogue, AM-374, and cannot be transposed to this molecule. Nor has real selectivity with respect to the other serine hydrolases been characterised here.

No biosynthetic pathway exists for this molecule. It is obtained by synthetic chemistry, from a sulfonated derivative with an octadecyl chain, the sulfonyl fluoride function constituting the chemically reactive part of the compound.

France
AM-381 is neither a tetrahydrocannabinol, nor an ester, nor an ether, nor a salt of THC: the generic clause of annex IV of the decree of 22 February 1990 therefore does not cover it. Nor does the molecule appear by name on the French narcotics list. Its status is that of an unclassified substance, which does not mean that it would be authorised for consumption: it is a research chemical, devoid of the status of a foodstuff as much as of a medicine, and placing it on the market for the general public would come under quite different regulation.
European Union
No European or international control measure covering AM-381 is documented. The molecule does not appear in the control decisions adopted by the Council of the European Union, nor in the publications of the EUDA, formerly the EMCDDA, nor in the UNODC lists. It circulates solely as a laboratory reagent, governed as such by the general regulation of chemical products.

Fluorure d'alcanesulfonyle à longue chaîne saturée en dix-huit carbones, dérivé du squelette d'un acide gras. Structurellement, ce n'est pas un cannabinoïde : c'est un inhibiteur enzymatique du système endocannabinoïde, la fonction fluorure de sulfonyle jouant le rôle de groupe réactif électrophile.
Pharmacokinetics
Aucune étude de pharmacocinétique n'a été publiée sur AM-381 : absorption, distribution, demi-vie et biodisponibilité restent inconnues, et les données humaines manquent entièrement. La forte lipophilie de la chaîne en dix-huit carbones et la réactivité de la fonction fluorure de sulfonyle, qui s'hydrolyse en milieu aqueux, sont des propriétés chimiques établies, mais leur traduction en exposition systémique n'a jamais été mesurée pour cette molécule.
Metabolism
Le devenir métabolique d'AM-381 n'a pas été décrit. Par construction chimique, la fonction fluorure de sulfonyle est consommée lors de la réaction avec l'enzyme cible ou par hydrolyse, en libérant un ion fluorure et l'acide sulfonique correspondant. Aucune identification de métabolites, aucune étude d'implication des cytochromes P450 et aucun profil d'élimination n'ont été publiés pour ce composé.
Toxicology and risks
Aucune étude de toxicologie ne porte sur AM-381. Le seul élément solide est de nature chimique : les fluorures de sulfonyle forment une fonction électrophile qui inactive les sérine hydrolases en général, une famille qui comprend aussi bien la FAAH que des enzymes vitales comme les cholinestérases, et la sélectivité d'AM-381 au sein de cette famille n'a pas été mesurée. Cette molécule doit donc être considérée comme un produit chimique réactif destiné au laboratoire, à manipuler dans un cadre professionnel, et non comme une substance dont l'innocuité serait établie. Aucun cas humain et aucune donnée de tolérance ne figurent dans la littérature.
Detection and analysis
