Pharmacology
AM-4054 is a classic tricyclic cannabinoid: a hexahydrocannabinol in which the usual pentyl chain is replaced by an adamantyl group, with a hydroxymethyl function at position 9. Thakur and colleagues measured for it in 2013 a nanomolar affinity for the CB1 receptor (Ki of 4.9 nM at rat CB1) and a comparable affinity for CB2 (11.3 nM at human CB2), that is, a modest selectivity of about 2.5-fold. The decisive difference from THC concerns efficacy: THC is only a partial agonist at CB1, whereas AM-4054 behaves as a full agonist, both in cAMP inhibition (EC50 of 1.29 nM) and in beta-arrestin recruitment. In the rat it produces the full cannabinoid tetrad, antinociception, catalepsy, hypothermia and suppression of locomotor activity, effects abolished by the CB1 antagonists AM251 and rimonabant, which confirms an action mediated by this receptor. Discrimination assays place it about twenty-seven to thirty times more potent than THC. No activity is documented at TRPV1, GPR55, PPARγ or 5-HT1A. Human data are entirely lacking: neither pharmacokinetics, nor metabolism, nor clinical observation has been published.

No known biological pathway: this molecule is formed in no living organism. It is built on the classic tricyclic cannabinoid skeleton, obtained chemically, in which the pentyl chain at position 3 is replaced by an adamantyl group and the methyl at position 9 by a hydroxymethyl. It arises from optimisation work carried out around the compound AM411.
Legal framework
France
Controlled in France by a generic clause, and not under its own name. The ANSM decision of 22 May 2024, which came into force on 3 June 2024, added to the decree of 22 February 1990 a generic definition covering "any substance derived from the benzo[c]chromene ring", hydrogenated or not on ring A, substituted in particular "at position 1 by a hydroxyl function", "at position 3 by an adamantyl substituent" and "at position 9 by a ketone, alkyl, hydroxyalkyl or alkoxy function". AM-4054 combines precisely these three substitutions on a hexahydrobenzo[c]chromene ring and therefore falls under this clause. An important point: it is not the classic clause covering tetrahydrocannabinols, their esters and their ethers that applies here, since the molecule is a hexahydrocannabinol and not a tetrahydrocannabinol. AM-4054 appears under its own name in no ANSM list by name.
European Union
No control at European Union level. AM-4054 does not appear in the annex to Framework Decision 2004/757/JHA and has been the subject neither of a joint report nor of a risk assessment by the EUDA, formerly the EMCDDA. The sources consulted report no seizure or notification to the European Early Warning System: the molecule has remained a laboratory reagent. Nor is it listed in the schedules of the United Nations conventions of 1961 and 1971. National regimes differ, several Member States applying, as France does, generic definitions liable to encompass it.

Pharmacokinetics
Aucune étude de pharmacocinétique n'a été publiée pour cette molécule : ni concentrations plasmatiques, ni demi-vie, ni biodisponibilité ne sont documentées, chez l'animal comme chez l'humain. Les seules indications temporelles disponibles proviennent des essais chez le rat, où l'hypothermie et l'antinociception s'installent une à une heure et demie après l'administration et culminent quatre à six heures plus tard. Cette cinétique traduit une action nettement plus lente à s'établir et plus prolongée que celle du THC.
Metabolism
Le métabolisme de l'AM-4054 n'a pas été étudié : aucun profil de métabolites, aucune identification des enzymes impliquées et aucun marqueur urinaire n'ont été publiés. Les cannabinoïdes classiques sont, en règle générale, oxydés par les cytochromes P450 hépatiques, mais cette généralité de classe n'a jamais été vérifiée sur cette molécule, et le volume du groupe adamantyle en modifie vraisemblablement le devenir.
Toxicology and risks
Aucune donnée humaine, aucun cas clinique et aucune étude de toxicité réglementaire ne concernent l'AM-4054 : la molécule n'a pas été retrouvée sur le marché des produits vendus comme cannabinoïdes. Le risque se déduit donc de sa pharmacologie, et il est celui de sa classe. L'agonisme complet du récepteur CB1, par opposition à l'agonisme partiel du THC, est exactement ce qui rend les agonistes cannabinoïdes de synthèse dangereux : cette famille est associée dans la littérature à des convulsions, des états d'agitation et de confusion, des tachyarythmies, des vomissements incoercibles, des atteintes rénales aiguës et des décès. C'est ce mécanisme, ainsi que le potentiel d'abus et de dépendance qui l'accompagne, que l'ANSM a invoqué pour classer par clause générique les cannabinoïdes de ce squelette. Chez le rat, l'AM-4054 produit la tétrade complète, hypothermie et catalepsie marquées comprises, à une puissance environ trente fois supérieure à celle du THC, avec un délai d'action long qui expose au réajustement prématuré des prises. Ces molécules sont le plus souvent rencontrées pulvérisées sur des végétaux et vendues sous une autre étiquette, parfois présentée comme du CBD, sans que l'acheteur sache ce qu'il manipule.
Detection and analysis
Aucune méthode de dépistage de routine n'est publiée pour l'AM-4054, qui n'apparaît ni dans les bulletins de toxicologie médico-légale ni dans les rapports de saisies consultés. Les tests immunologiques usuels du cannabis ciblent le métabolite carboxylique du THC et ne reconnaîtraient pas cette structure. Son identification supposerait une chromatographie couplée à la spectrométrie de masse haute résolution, avec un étalon de référence dédié, et se heurterait à l'absence de spectres de métabolites publiés.
