Pharmacology

A-41988 belongs to a family of azacannabinoids developed at Abbott Laboratories: the carbocyclic ring of Δ6a,10a-tetrahydrocannabinol is replaced by a piperidine whose nitrogen bears a propargyl group, and the pentyl chain of THC by a 4-(4-fluorophenyl)-1-methylbutyl chain. In animals, the molecule reproduces the profile of the classical cannabinoids: analgesia in the tail-flick test in rats and in the abdominal writhing test in mice, a marked fall in motor activity, prolonged sleeping time in cats, loss of coordination in dogs at low oral doses, a lasting fall in blood pressure in hypertensive rats, as well as digestive effects, reduced gastric acid secretion and reduced experimental diarrhoea. The limitation of these data must be stated plainly: they were produced before the cannabinoid receptors were identified, and no affinity or functional activity at CB1 or CB2 has ever been measured for this molecule. In humans, the only published trial, placebo-controlled in patients with ocular hypertension, showed no fall in intraocular pressure at the doses tested, unlike the comparator compound. Beyond that trial, human data are lacking.

No biosynthetic pathway: A-41988 is obtained by total chemical synthesis, built around construction of the benzopyrano[4,3-c]pyridine nucleus, a cannabinoid-type skeleton whose carbocyclic ring is replaced by a piperidine. Hemp, whose enzymatic pathway starts from CBGA, produces nothing comparable.

Azacannabinoïde tricyclique de la série 5H-[1]benzopyrano[4,3-c]pyridine : un analogue du Δ6a,10a-tétrahydrocannabinol dont le cycle carboné central est remplacé par une pipéridine portant un groupe propargyle sur l'azote. Le phénol occupe la position 10, un gem-diméthyle ferme le cycle pyrane, et la chaîne pentyle du THC cède la place à une chaîne 4-(4-fluorophényl)-1-méthylbutyle.

Pharmacokinetics

Le composé est actif par voie orale chez le rongeur, le chat, le chien et chez l'homme, ce qui indique une absorption digestive suffisante. Chez le rat hypertendu, la baisse de pression artérielle reste mesurable vingt-quatre heures après une prise unique, ce qui suggère une durée d'action longue, cohérente avec le caractère très lipophile de la molécule. Aucune étude d'absorption, de distribution, de demi-vie ou d'élimination n'a été publiée.

Metabolism

Aucune étude de métabolisme n'a été publiée pour A-41988, ni chez l'animal ni chez l'homme. Les voies de transformation de cette molécule, y compris le devenir du groupe propargyle porté par l'azote, restent inconnues.

Toxicology and risks

La toxicité aiguë par voie orale rapportée chez la souris dans l'article princeps est faible. Les effets indésirables attendus découlent du profil dépresseur central de la série : sédation marquée, perte de coordination chez le chien dès de faibles doses orales, réactivité anormale au toucher chez le rat, baisse de la pression artérielle et modifications du rythme cardiaque. Dans le seul essai clinique publié, les patients ayant reçu A-41988 ont rapporté des effets indésirables subjectifs légers et ont présenté une altération d'un test de production du temps, c'est-à-dire de la perception des durées. Il n'existe aucune donnée de toxicité chronique, de reprotoxicité, de tolérance ou de dépendance chez l'homme, et aucun cas d'intoxication n'a été décrit.

Detection and analysis

Aucune méthode analytique dédiée à A-41988 n'a été publiée : les travaux disponibles sont chimiques et pharmacologiques, pas médico-légaux. Le composé n'est pas recherché dans les dépistages de routine, et son squelette azoté le distingue nettement des cannabinoïdes de synthèse rencontrés sur le marché illicite, ce qui rend improbable une détection fortuite par les méthodes visant ces derniers.
