Pharmacology

A-40174 belongs to the classic cannabinoids: the tricyclic core of tetrahydrocannabinol is retained, but one carbon of the third ring is replaced by a nitrogen, which gives a chromeno[4,3-c]pyridine ring bearing a propargyl group, and the pentyl chain of THC gives way to the 1,2-dimethylheptyl chain of dimethylheptylpyran. The work published by Abbott between 1976 and 1983 describes a typically cannabinoid profile in animals: an analgesic effect in the tail-flick and writhing tests, reduced motor activity, ataxia in the dog, prolonged sleeping time in the cat, lowered blood pressure in the hypertensive rat, as well as an antisecretory and antidiarrhoeal action. A structure-activity comparison from 1978 provides the most important point for safety: tachycardia occurred in the conscious rat at doses far below those needed to obtain analgesia, and it was the water-soluble esters of dimethylheptylpyran, not the nitrogen-containing analogue, that offered the best ratio between the two effects. No affinity for CB1 or CB2 has ever been measured for this molecule, no metabolic data exist, and no human trial has been published: the series was abandoned without ever reaching the clinic.

No known biological pathway. A-40174 is produced by no plant and no animal: it is obtained exclusively by chemical synthesis, from a chromeno[4,3-c]pyridine ring, with no connection to the cannabigerolic acid pathway that gives rise to the cannabinoids of hemp.

Pharmacokinetics

La molécule est active par voie orale chez plusieurs espèces et atteint le système nerveux central, comme le montrent la sédation, la baisse d'activité motrice et l'ataxie observées après administration orale chez le rat et le chien. Aucune étude d'absorption, de distribution, de demi-vie ni de biodisponibilité n'a été publiée, et rien n'est connu chez l'être humain.

Toxicology and risks

Le signal le plus net est cardiovasculaire : chez le rat éveillé, A-40174 accélère le rythme cardiaque à des doses très inférieures à celles qui produisent une analgésie, et il abaisse la pression artérielle chez le rat hypertendu. Les études animales décrivent aussi une dépression marquée du système nerveux central, une réduction de l'activité motrice et une ataxie chez le chien, effets caractéristiques des dérivés N-propargyle de cette série. La létalité aiguë par voie orale chez la souris n'apparaît qu'à des niveaux d'exposition très élevés. Aucune donnée de toxicité humaine, aucun cas clinique et aucune étude de toxicité répétée n'ont été publiés : l'absence totale de recul rend toute consommation déraisonnable.

Detection and analysis

Aucune méthode analytique dédiée n'a été publiée pour cette molécule. Elle n'est pas recherchée par les tests immunologiques urinaires courants, y compris ceux destinés aux cannabinoïdes de synthèse, et n'a pas été signalée dans les saisies rapportées par les systèmes d'alerte européens. Une identification exigerait une chromatographie couplée à la spectrométrie de masse et un étalon de référence.
