Pharmacology
A-796260 is an indol-3-yl ketone of the aminoalkylindole family, bearing a 2,2,3,3-tetramethylcyclopropyl group on the carbonyl bridge and a 2-(morpholin-4-yl)ethyl chain on the indole nitrogen. Yao and colleagues measured an affinity of 4.37 nM at the human CB2 receptor against 845 nM at human CB1, that is a selectivity of about 190-fold, and potent activation of CB2 in cAMP inhibition, with an EC50 of 0.710 nM and a partial efficacy of 78 %. In four rodent pain models — inflammatory, postoperative, neuropathic and osteoarthritic — the effect was suppressed by a selective CB2 antagonist but not by a CB1 antagonist or by a mu opioid antagonist, with no impairment of locomotor activity. This molecule is therefore not a full CB1 agonist like the synthetic cannabinoids of the illicit market, and the observations made on its street analogues UR-144 and XLR-11, which differ from it only by the side chain, belong to those analogues and not to it. Set against this, the only toxicology study available attributes to it a strong cytotoxic potential on cultured human liver cells. Human data are entirely lacking: no clinical trial, no pharmacokinetic data and no published case of intoxication.

Origin (research tool)
No known biological pathway: the molecule is entirely synthetic and absent from every living organism. Chemically it belongs to the indol-3-yl ketones, obtained by acylation of the indole nucleus followed by alkylation of its nitrogen.
Legal framework
France
Prohibited in France, without being named there. A-796260 does not appear by name in annex IV of the decree of 22 February 1990, but it falls under the generic definition of indol-3-yl ketones introduced into that annex by the decree of 31 March 2017. This clause covers any indol-3-yl methanone bearing on the indole nitrogen a substituent of the 2-(4-morpholinyl) ethyl type, which corresponds exactly to its side chain, and on the carbon of the methanone bridge a group of the cyclopropyl type, whether or not further substituted, which covers its 2,2,3,3-tetramethylcyclopropyl group. Two immediate analogues, A-834,735 and AB-005, are for their part cited by name in the same annex. The generic clause relating to tetrahydrocannabinols, their esters and their ethers does not apply here, the molecule not being a THC derivative. Production, transfer, possession and use are therefore prohibited under the narcotics regime.
European Union
No control measure specific to this molecule exists at European Union level: no EUDA risk assessment, formerly EMCDDA, has been found concerning it, and it appears in no schedule of the United Nations conventions of 1961 and 1971. Control therefore falls to national laws, and several member states apply, as France does, generic definitions covering the indol-3-yl ketone family: its status may thus differ from one country to another. The molecule moreover holds no status as a food or cosmetic ingredient in the Union.

Aminoalkylindole, plus précisément cétone d'indol-3-yle : le noyau indole est acylé en position 3 par un groupe 2,2,3,3-tétraméthylcyclopropyle, et l'azote indolique porte une chaîne 2-(morpholin-4-yl)éthyle. Pharmacokinetics
Les données humaines manquent : ni biodisponibilité, ni demi-vie, ni volume de distribution n'ont été publiés. Chez le rongeur, l'effet analgésique a été obtenu après administration systémique par voie intrapéritonéale. Sur microsomes hépatiques humains, le métabolisme de phase I est extensif et réparti entre plusieurs isoenzymes du cytochrome P450, ce qui rend, selon Gampfer et ses collègues, une interaction médicamenteuse peu probable.
Metabolism
Gampfer et ses collègues ont identifié en 2021, sur microsomes hépatiques humains, les métabolites de phase I de la molécule. Les plus abondants résultent d'une coupure oxydative du cycle morpholine, d'une ouverture oxydative de ce même cycle, d'une hydroxylation, et d'une dihydroxylation suivie d'une déshydrogénation. Plusieurs isoenzymes du cytochrome P450 y participent, sans qu'une voie unique ne domine.
Toxicology and risks
Le seul travail de toxicologie disponible est un criblage à haut contenu sur cellules hépatiques humaines HepG2, publié par Gampfer et ses collègues en 2021 : A-796260 y montre un fort potentiel cytotoxique, du même ordre que celui du naphtoylindole JWH-200 et supérieur à celui de 5F-EMB-PINACA. L'emploi d'un inhibiteur large du cytochrome P450 indique que le métabolisme joue un rôle mineur dans cet effet, qui semble donc porté par la molécule elle-même. Aucun cas d'intoxication humaine, aucune série clinique et aucun décès associés à cette molécule n'ont été retrouvés dans la littérature. L'absence d'effet de type cannabique et l'absence d'atteinte de l'activité locomotrice chez le rongeur ne valent pas démonstration d'innocuité chez l'humain, et la faible affinité pour le CB1 ne dit rien de la cytotoxicité hépatique observée in vitro.
Detection and analysis
Les métabolites de phase I les plus abondants, décrits par Gampfer et ses collègues au moyen de la chromatographie liquide couplée à la spectrométrie de masse haute résolution en tandem, sont proposés comme cibles analytiques pour le criblage urinaire. Aucune méthode de dépistage validée sur des échantillons humains réels n'a été publiée à ce jour, et aucune détection de cette molécule dans des produits saisis n'a été retrouvée.
