- Le composé est apparu sur le marché avant toute publication scientifique. Il a été repéré en 2010 dans des produits vendus en boutiques spécialisées en Irlande, puis notifié en 2011 au système d'alerte précoce européen par les autorités allemandes. La même année, l'Institut national de la pharmacie de Budapest en a élucidé la structure dans des poudres de haute pureté saisies à un aéroport hongrois et étiquetées comme stéarate de calcium ou acide malique, travail publié par Jankovics et ses collègues dans Forensic Science International en 2012. Pharmacology
AB-001 is an adamantoylindole, a full agonist of the CB1 and CB2 receptors. In the functional assay published in 2013 by Banister's team, the EC50 is 35 nM at CB1 and 48 nM at CB2, with an efficacy comparable to that of the reference agonist WIN 55,212-2. The difference from cannabis lies less in affinity than in efficacy: Δ9-THC is only a partial agonist of CB1, whereas this compound fully activates the receptor, with no ceiling of effect, a mechanism that makes this class dangerous. The same study nevertheless observed weak cannabimimetic effects in the rat, with hypothermia and bradycardia remaining at the level of a trend, whereas the carboxamide analogue APICA produced them clearly; this gap between cellular potency and animal response is not explained. Human pharmacology has never been studied: human data are lacking on kinetics, dependence potential and the effects of repeated use, and no clinical series concerns this precise molecule. The only published human work bears on urine, where the unchanged compound is absent, detection resting on its metabolites hydroxylated on the adamantane cage.

Origin (pharmaceutical research → illicit market)
No biological pathway: this compound comes from no plant or animal metabolism and exists only through organic synthesis. Its preparation involves the acylation of an indole previously alkylated on nitrogen 1 with a derivative of adamantane-1-carboxylic acid, classic aminoalkylindole chemistry, first described in the 2013 publication that provided its pharmacological evaluation.
Legal framework
France
The molecule is listed by name in France. Annex IV of the decree of 22 February 1990 setting the list of substances classified as narcotics contains a generic family of "indol-3-yl methanones", defined by the nature of the substituent borne by the nitrogen of the indole ring and by that of the group borne by the carbon of the methanone bridge, the adamantyl group being among those admitted. The text expressly cites among its examples "AB-001 ou (1-pentyl-3-(adamant-1-oyl) indole)", a mention introduced by the decree of 31 March 2017 published in the Journal officiel of 6 April 2017 and specified by the decree of 3 October 2017. This is therefore a listing by name and not a mere generic clause: use, possession, transfer, acquisition and importation fall under the narcotics regime. The older clause covering "Tetrahydrocannabinols, their esters, ethers, salts as well as the salts of the aforementioned derivatives" is of no relevance here, this compound being neither a tetrahydrocannabinol nor a derivative of one.
European Union
There is no harmonised control measure at European Union level for this compound: no formal risk assessment has been initiated in respect of it and classification is a matter for the Member States, the European level being limited to the early warning system, to which the substance was notified in 2011 by the German authorities. Several countries have scheduled it nationally, either by name or through generic definitions covering indol-3-yl methanones; Germany has listed it in annex II of its narcotics law, a regime that permits regulated trade but forbids prescription. Internationally, it appears in no schedule of the United Nations conventions. Outside the Union, it falls under schedule II of the Canadian Controlled Drugs and Substances Act and under class B in the United Kingdom.

Adamantoylindole de la famille des aminoalkylindoles : noyau indole portant une chaîne pentyle sur l'azote 1 et, en position 3, un pont cétone relié à une cage adamantan-1-yle. Ce squelette n'a aucun rapport avec celui des cannabinoïdes du chanvre, bâtis sur un dibenzopyrane ou un résorcinol. La cage adamantane est symétrique et la molécule ne possède aucun centre stéréogène : il n'existe donc pas de stéréoisomères. Deux composés voisins se distinguent par un seul détail structural, l'APICA, où le pont cétone est remplacé par une liaison carboxamide, et l'AM-1248, qui porte un groupement N-méthylpipéridin-2-ylméthyle à la place de la chaîne pentyle ; les données publiées sur ces deux analogues leur appartiennent et ne décrivent pas l'AB-001.
Pharmacokinetics
La voie d'exposition documentée est l'inhalation, par combustion du produit pulvérisé sur un support végétal. Aucune étude d'absorption, de distribution ou d'élimination n'a été conduite chez l'humain, et les données humaines manquent sur la quantité produisant un effet comme sur la durée d'action. Le seul repère publié provient de la toxicologie urinaire : les métabolites restent détectables jusqu'à environ cent soixante heures après une prise, ce qui indique une élimination lente des produits de transformation. Chez le rat, le contraste entre une activité cellulaire nette et des réponses physiologiques faibles suggère une biodisponibilité ou une distribution défavorables, hypothèse qui n'a pas été mesurée.
Metabolism
Le métabolisme est hépatique et extensif. Les travaux de Grigoryev, Kavanagh et Melnik sur urines humaines, publiés en 2012 dans Drug Testing and Analysis, montrent que le composé inchangé ne se retrouve pas dans l'urine : les produits majoritaires sont des dérivés monohydroxylés sur la cage adamantane, seuls ou associés à la perte de la chaîne pentyle portée par l'azote de l'indole. La recherche urinaire doit donc viser ces métabolites et non la molécule mère. Les autres transformations attendues pour cette classe, hydroxylation de la chaîne latérale et conjugaison au glucuronide, n'ont pas fait l'objet d'une caractérisation dédiée pour ce composé.
Toxicology and risks
Aucune évaluation toxicologique systématique n'a été conduite et les données humaines manquent : la littérature ne comporte pour cette molécule précise ni série clinique, ni décès rapporté, ni donnée sur l'usage répété, ce qui reflète sa diffusion limitée et non une innocuité démontrée. Chez le rat, l'étude de 2013 n'a pas retrouvé les effets hypothermiques et bradycardiques caractéristiques des agonistes cannabinoïdes. Le risque tient d'abord à la classe : un agoniste complet de CB1 ne connaît pas le plafond d'effet du Δ9-THC, et les tableaux décrits pour d'autres cannabinoïdes de synthèse comportent tachycardie, vomissements répétés, agitation, confusion, convulsions et épisodes psychotiques. Ces observations appartiennent aux composés qui ont été étudiés et ne sont pas transposables telles quelles ici, car un fluor ou un carbone de différence suffit à changer le profil. Le danger propre à ce marché reste la tromperie sur le contenu : le produit se rencontre pulvérisé sur des plantes vendues comme article légal ou comme substitut du cannabis, la pulvérisation répartit la matière active de façon inégale sur le support, et la personne qui consomme ignore le plus souvent quelle molécule se trouve réellement dans le sachet.
Detection and analysis
La molécule échappe aux immunoessais cannabinoïdes courants, calibrés sur les métabolites du Δ9-THC. Sur les saisies, l'identification repose sur la chromatographie en phase gazeuse ou liquide couplée à la spectrométrie de masse, complétée par la spectrométrie de masse à temps de vol et par la résonance magnétique nucléaire, combinaison employée lors de l'élucidation structurale initiale en Hongrie. En toxicologie urinaire, la méthode publiée est une chromatographie en phase gazeuse couplée à la spectrométrie de masse ciblant les métabolites hydroxylés de la cage adamantane, la molécule mère étant absente de l'urine. La distinction avec les composés voisins, notamment l'analogue carboxamide APICA et l'AM-1248, exige un étalon de référence certifié.
