Pharmacology

An analogue of arachidonic acid in which the acid function has given way to a trifluoroacetyl group, AACOCF3 acts as a transition-state analogue of serine hydrolases: the carbonyl carbon, made electron-poor by the three fluorines, forms a reversible hemiketal with the catalytic serine. Its original target, described in 1993, is the 85 kDa cytosolic phospholipase A2, which it inhibits with slow, tight-binding kinetics; it is still in this capacity that the molecule is most often used, as a means of cutting off the release of arachidonic acid upstream of prostaglandins and leukotrienes. Its entry into endocannabinoid pharmacology dates from 1994: among a series of anandamide analogues, it emerged as the best inhibitor of anandamide amidase, FAAH, and raises the anandamide level of neuroblastoma cells roughly twelvefold. It was also the only compound of that series to displace a cannabinoid radioligand at CB1, with a Ki of 0.65 µM, without its intrinsic efficacy at this receptor ever having been established. This accumulation of targets constitutes the tool's main limitation: an inhibitor of cPLA2, an inhibitor of FAAH, an inhibitor of calcium-independent phospholipase A2 with an IC50 of 15 µM, and a direct CB1 ligand in the same submicromolar range, the molecule does not allow an observed effect to be cleanly attributed to a specific pathway. The FAAH inhibitors published subsequently are markedly more selective and more potent: MAFP, described in 1997, surpasses it by a factor of about eight hundred at the amidase. Human data are lacking, with no clinical trial, no tolerability data and no established therapeutic benefit; in vivo work is confined to rodents, where neuroprotection after spinal cord injury and functional improvement after cauda equina compression have been reported.

Cétone trifluorométhylée dérivée de l'acide arachidonique : la fonction acide carboxylique du C20:4 est remplacée par un groupement trifluoroacétyle, la chaîne tétraénoïque et sa géométrie tout-cis restant intactes. Les trois fluors appauvrissent le carbone du carbonyle en électrons, ce qui fait de la molécule un analogue de l'état de transition des sérine hydrolases : en milieu aqueux le groupement s'hydrate largement, et c'est la forme cétone qui réagit avec la sérine catalytique en formant un hémicétal réversible. La molécule ne comporte aucun noyau terpénophénolique et n'appartient donc pas aux cannabinoïdes au sens chimique ; son rattachement au système endocannabinoïde est purement fonctionnel, par ses cibles enzymatiques.

Pharmacokinetics

Aucune donnée de pharmacocinétique humaine n'existe. Un point physico-chimique conditionne son usage : en milieu aqueux la cétone trifluorométhylée s'hydrate largement, et c'est la fraction non hydratée qui réagit avec l'enzyme, si bien que la puissance apparente dépend du milieu d'essai, ce que les auteurs de 1993 ont établi par résonance magnétique nucléaire du fluor. Les travaux chez le rat ont recouru à la voie intraveineuse après lésion médullaire et à la voie orale dans le modèle de compression de la queue de cheval, cette dernière montrant qu'une administration digestive suffit à produire un effet central. Aucune étude de distribution, de liaison aux protéines plasmatiques ni d'élimination n'a été publiée.

Metabolism

Aucune étude dédiée n'a caractérisé les voies de biotransformation ni identifié de métabolite. Le mécanisme d'action lui-même ne consomme pas la molécule : contrairement aux inhibiteurs covalents de type fluorophosphonate ou fluorure de sulfonyle, l'hémicétal formé avec la sérine catalytique est réversible, ce qui rend compte de la dissociation lente mesurée sur la cPLA2 plutôt que d'un blocage définitif. Le devenir de la molécule dans l'organisme reste donc inconnu.

Toxicology and risks

Aucune donnée de toxicologie humaine n'est disponible : la molécule n'a jamais été administrée à l'être humain. Chez le rat, les deux travaux in vivo publiés ne rapportent pas de toxicité, celui conduit par voie orale ne signalant aucun effet indésirable, mais il s'agit de séries courtes, sans étude de toxicité réglementaire ni suivi au long cours. Le risque théorique dominant tient à la cible elle-même : inhiber la phospholipase A2 cytosolique coupe la production des éicosanoïdes, y compris ceux qui participent à l'homéostasie normale, et le manque de sélectivité étend ce blocage à la phospholipase A2 indépendante du calcium et à la FAAH. Ce n'est pas un produit de consommation et il n'existe aucun usage alimentaire, cosmétique ou thérapeutique.

Detection and analysis

Aucune méthode de dépistage de routine ne cible cette molécule, ni en toxicologie médicolégale ni en contrôle antidopage : elle n'est pas classée, n'apparaît pas dans les produits de consommation et reste un réactif de paillasse. Les publications la manipulent comme un outil, sans qu'aucune méthode validée de quantification en matrice biologique ait été décrite.
