In plain terms
AB-005 is a laboratory molecule with no connection to hemp: a pharmaceutical laboratory described it in 2010, after which clandestine manufacturers took it up to spray it onto plant material sold as smoking blends. It binds to the same receptors as THC, with a marked preference for the CB2 receptor. What it does in human beings remains unknown, no clinical study or published medical case concerns it, and it is classified as a narcotic in France.

Pharmacology
AB-005 is an indol-3-yl ketone: the indole nucleus carries a 2,2,3,3-tetramethylcyclopropyl group at position 3 and, on its nitrogen, a (1-methylpiperidin-2-yl)methyl chain. It is a chimeric compound, assembled from fragments of UR-144 and AM-1248, arising from a pharmaceutical programme in search of selective CB2 agonists (Frost and colleagues, 2010). The two available sets of measurements agree on a strong affinity for CB2 and a weaker one for CB1: 0.48 nM and 5.5 nM in Frost's work, 1.04 nM and 59.4 nM in the independent re-evaluation by Hess and colleagues (2016). The decisive point is functional: in that same study, most of the synthetic cannabinoids tested behaved as full agonists at the CB receptors, whereas AB-005 activated them with an efficacy lower than that of THC, itself a partial agonist in this system. Full agonism at the CB1 receptor, which accounts for the clinical severity of many compounds in this family, is therefore not established for AB-005. Its azepane isomer, also found on the illicit market, showed no CB1 affinity up to 10 µM and behaves as a moderate CB2 antagonist: two powders bearing the same name may have opposite pharmacologies. No human data, pharmacokinetic or clinical, have been published.

Origin (pharmaceutical research → illicit market)
No biological pathway: the molecule is entirely synthetic. Its framework combines two motifs already known from synthetic cannabinoids, the tetramethylcyclopropyl ketone of UR-144 and XLR-11 on the one hand, and the (1-methylpiperidin-2-yl)methyl chain of AM-1220 and AM-1248 on the other, grafted onto an indole nucleus.
Legal framework
France
A narcotic. AB-005 is cited by name in annex IV of the decree of 22 February 1990, within the generic family of indol-3-yl methanones introduced by the decree of 31 March 2017: this clause expressly covers the 1-(N-methylpiperidin-2-yl)methyl substituent on the indole nitrogen and a cyclopropyl group on the carbon of the methanone bridge. The heading also covers isomers, stereoisomers, esters, ethers and salts, which encompasses the azepane isomer described as a by-product. It is therefore not a mere attachment by generic clause: the substance appears under its own name in the consolidated list published by the ANSM.
European Union
Control rests with national legislation, which differs from one Member State to another, rather than with a measure designating AB-005 at Union level. The substance is recorded by the UNODC as a cannabinoid receptor agonist in its early warning system on new psychoactive substances. It does not appear in the schedules of the United Nations conventions of 1961 and 1971: the French annex IV brings together precisely those substances scheduled in France or in the Union without being scheduled internationally.

Pharmacokinetics
Aucune donnée humaine. Le composé est très lipophile, comme toute la série étudiée par Hess et ses collègues en 2016, dont les log P calculés vont de 3 à 7 ; ce travail conclut, par prédiction informatique et non par mesure, que la majorité de ces molécules, AB-005 comprise, franchiraient la barrière hémato-encéphalique. Un modèle rongeur publié en 2025 a administré la molécule par voie intraveineuse, puis recherché ses métabolites dans le sang, le cerveau, le foie et les fèces.
Metabolism
Caractérisé en 2025 sur microsomes hépatiques humains et chez le rat par Ning et ses collègues : dix-sept métabolites décrits, douze dans le modèle in vitro et cinq in vivo, avec un taux de conversion in vitro de 60,9 %. Les voies dominantes sont l'hydroxylation et la dihydroxylation, la déshydrogénation, la coupure de la chaîne latérale portée par l'azote indolique et la déméthylation de l'azote pipéridinique. Les enzymes responsables n'ont pas été identifiées.
Toxicology and risks
Les données humaines manquent : aucune intoxication, aucun cas clinique et aucun décès attribués spécifiquement à AB-005 ne figurent dans la littérature indexée. Ce que la classe des agonistes de synthèse a documenté, convulsions, délire, tachyarythmies, vomissements incoercibles, atteinte rénale aiguë et décès, provient d'autres composés, le plus souvent des agonistes complets très puissants, et ne peut pas être reporté sur celui-ci. Le risque concret tient au mode de diffusion : ces molécules circulent pulvérisées de façon inégale sur des végétaux, à teneur inconnue, sous des noms de fantaisie parfois présentés comme légaux, et fréquemment en mélange avec d'autres cannabinoïdes de synthèse dont la toxicité, elle, est établie. La circulation documentée de l'isomère azépane d'AB-005, inactif sur CB1, ajoute une incertitude sur le contenu réel d'un même lot.
Detection and analysis
AB-005 figure dans les panels médico-légaux : la méthode LC-MS/MS d'Ambroziak et Adamowicz (2018), qui couvre soixante-douze cannabinoïdes de synthèse en sang total, la suit avec un ion précurseur à m/z 353,3 et des ions produits à 125,1, puis 112,1 et 98,1. Comme les autres composés de la classe, elle est métabolisée rapidement, de sorte que la recherche de la molécule inchangée expose à des faux négatifs ; les métabolites hydroxylés et désalkylés décrits en 2025 fournissent les marqueurs correspondants. Son isomère azépane possède la même masse et ne s'en distingue donc pas sur le seul rapport masse sur charge.
