- Des chercheurs japonais conduits par Uchiyama décrivent la molécule en 2015 dans Forensic Toxicology, après l'avoir identifiée par chromatographie en phase gazeuse, chromatographie liquide et résonance magnétique nucléaire dans des produits illégaux à base de végétaux séchés. Ce sont eux qui forgent le code AB-CHFUPYCA, acronyme construit sur son nom systématique. En 2016, McLaughlin et ses collègues obtiennent le composé auprès d'un vendeur britannique en ligne qui l'annonçait à tort comme AZ-037, une autre molécule de la même série, et publient sa préparation, sa structure cristalline et les critères analytiques permettant de le séparer de son régio-isomère. Sa pharmacologie est restée inexplorée jusqu'aux travaux de Deventer et de ses collègues, publiés en 2023.

AB-CHFUPYCA is a laboratory-made molecule with no connection to hemp: it has been found sprayed onto dried plant material that is sold as incense or as smoking herb, and also in the form of a powder. Unlike most products of this kind, laboratory assays show that it activates the cannabis receptors very little and that it would tend instead to block them. No human data exist: neither its effects nor its real risks are known.

Pharmacology
AB-CHFUPYCA belongs to the synthetic cannabinoid receptor agonists, within the family of pyrazole-carboxamides with a valinamide residue, where the pyrazole core replaces the indazole of the PINACA and CHMINACA series. The only pharmacological study published to date, that of Deventer and colleagues in 2023, reaches an unusual result for this class: in a β-arrestin 2 recruitment assay, the molecule activates CB1 only weakly and only at the highest concentrations, with no usable plateau, does not activate CB2, and behaves rather as a blocker of both receptors. It therefore does not display the full CB1 agonism that characterises the dangerous synthetic cannabinoids and that separates them from THC, a mere partial agonist. The authors conclude that the immediate health threat is probably limited, while stressing that they can exclude neither toxicity nor effects independent of the cannabinoid receptors. What remains established is slight: a mechanism measured in vitro, a metabolism described in human microsomes, and nothing else. Human data are entirely lacking, no study in humans or in animals having been conducted, and the weak activity observed in vitro says nothing about the safety of a clandestine product whose real composition is unknown.

Origin (pharmaceutical research → illicit market)
No known biosynthetic pathway: the molecule exists in no living organism and derives from no plant precursor. It is obtained chemically, by coupling an N-alkylated pyrazole-carboxylic acid bearing a fluorophenyl group to an amide derived from valine, with no relation whatsoever to the cannabigerol pathway that gives rise to the cannabinoids of hemp. The authors who reproduced this chemistry showed that one of the possible routes leaves a chlorinated by-product, liable to accompany the substance in a clandestine batch.
Legal framework
France
A substance classified by name as a narcotic in France. AB-CHFUPYCA, cited under this code and under its synonym AB-CHMFUPPYCA, was entered in annex IV of the decree of 22 February 1990 setting the list of substances classified as narcotics, by the decree of 31 March 2017 published in the Journal officiel of 6 April 2017. This is therefore not an indirect attachment through the generic clause covering tetrahydrocannabinols, their esters and their ethers, a clause that does not cover this chemical family: the molecule is targeted in its own right. The entry extends to isomers, stereoisomers, esters, ethers and salts, which encompasses the 5,3-AB-CHMFUPPYCA regioisomer. The published text nonetheless associates with this code an erroneous systematic name, copied from the substance preceding it in the list, whereas the designation AB-CHFUPYCA appears there unambiguously. Production, possession, transfer and use fall under the criminal regime for narcotics. This substance has no place in the wellness hemp or CBD trade.
European Union
No control measure specific to the European Union: the substance has been the subject of neither a joint report nor a risk assessment by the European drugs agency, and it appears in no schedule of the United Nations Conventions of 1961 and 1971. Classification therefore depends on national laws, which diverge: France has named it since 2017, while other States rely on general laws covering psychoactive substances. The series remains monitored by the European early warning system, which received in June 2015 a French notification concerning the analogue 5F-AB-FUPPYCA, also called AZ-037, then in 2021 a German notification concerning the 5,3-AB-CHMFUPPYCA regioisomer. Outside the Union, the Chinese generic classification of synthetic cannabinoids adopted in 2021 covers only regioisomers of the 5,3 type: the 3,5 form, that of AB-CHFUPYCA, escapes it, which explains the renewed interest observed for these pyrazoles on the illicit market.

Agoniste de synthèse des récepteurs cannabinoïdes de la famille des pyrazole-carboxamides, sous-groupe des valinamides. La molécule associe un noyau pyrazole monocyclique, un groupe 4-fluorophényle en position 3, une chaîne cyclohexylméthyle portée par l'azote 1 et un résidu amide dérivé de la valine. Le pyrazole y remplace, par bio-isostérie, le noyau indazole des séries PINACA, FUBINACA et CHMINACA : AB-CHFUPYCA est l'analogue pyrazole d'AB-CHMINACA. Elle ne partage aucun squelette avec les cannabinoïdes du chanvre, bâtis sur un motif dibenzopyrane ou résorcinol. Le noyau pyrazole engendre un régio-isomère, le 5,3-AB-CHMFUPPYCA, dans lequel la fonction carboxamide et le groupe fluorophényle sont permutés ; la littérature désigne pour cette raison AB-CHFUPYCA sous le nom 3,5-AB-CHMFUPPYCA.
Pharmacokinetics
Les données humaines manquent entièrement : aucune étude d'administration contrôlée, aucun paramètre pharmacocinétique publié, ni chez l'humain ni chez l'animal. Un seul élément touche à l'exposition réelle : soumise aux conditions du fumage, la molécule n'est pas totalement stable et une fraction se dégrade par rupture de la liaison amide, de sorte qu'une part de ce qui est inhalé n'est plus la substance de départ. La quantité effectivement absorbée reste inconnue de la personne qui consomme, d'autant que la répartition du produit sur son support est inégale.
Metabolism
Le métabolisme de phase I a été décrit par Franz et ses collègues sur microsomes hépatiques humains, pour AB-CHFUPYCA comme pour son régio-isomère : les profils des deux isomères sont qualitativement voisins et dominés par l'oxydation de la chaîne cyclohexylméthyle. Les métabolites monohydroxylés y sont abondants et les auteurs les proposent comme cibles de recherche urinaire. Les produits de dégradation formés à la chaleur ont également été retrouvés dans ces incubations, ce qui brouille l'interprétation. Les isoformes du cytochrome P450 impliquées n'ont pas été identifiées. Une étude ultérieure sur les carboxylestérases humaines a porté sur un analogue fluoré de la même série et non sur cette molécule : ses conclusions ne lui sont pas transposables.
Toxicology and risks
Il n'existe aucune donnée de toxicité propre à cette molécule : les auteurs de la seule étude pharmacologique disponible relèvent explicitement l'absence de toute information sur ses effets chez l'humain comme chez l'animal, et aucun cas clinique ni décès ne lui est attribué dans la littérature. La classe des agonistes de synthèse des récepteurs cannabinoïdes est associée à des convulsions, des états d'agitation et de confusion, des hallucinations, une toxicité cardiovasculaire, des rhabdomyolyses, des comas et des décès, mais ces observations concernent des composés bien plus puissants et ne sont pas transposables à AB-CHFUPYCA, dont l'activité sur CB1 mesurée in vitro est faible. Les auteurs estiment le risque sanitaire immédiat probablement limité pour cette raison, sans pouvoir exclure une toxicité ni des effets ne passant pas par les récepteurs cannabinoïdes. Le danger documenté est ailleurs, dans le produit plutôt que dans la molécule : un cas d'étiquetage erroné, un produit secondaire chloré propre à l'une des voies de préparation, une répartition inégale sur le support, et le fait que des analogues de cette série ont été saisis en mélange avec un agoniste de synthèse réellement puissant, l'ADB-BUTINACA, observation qui porte sur ces analogues et non sur AB-CHFUPYCA elle-même.
Detection and analysis
La recherche analytique repose sur la chromatographie liquide couplée à la spectrométrie de masse, complétée par la haute résolution pour le criblage non ciblé. La difficulté propre à cette molécule est son régio-isomère, dont la masse et la formule sont identiques : les critères chromatographiques, spectroscopiques et spectrométriques publiés par McLaughlin et ses collègues, appuyés sur la structure cristalline des deux formes, permettent de les distinguer. Mettre en évidence une consommation suppose de viser les métabolites monohydroxylés décrits sur microsomes humains. Un piège d'interprétation a été signalé : des produits de dégradation thermique se retrouvent aussi parmi les métabolites, ce qui pèse notamment sur l'analyse capillaire, où la présence d'un métabolite est habituellement lue comme la preuve d'une consommation plutôt que d'une contamination externe. Les tests immunochimiques courants destinés au cannabis ne détectent pas cette substance.
