- Le squelette indazole-3-carboxamide à résidu valinamide provient de travaux pharmaceutiques de la fin des années 2000 portant sur des agonistes cannabinoïdes à visée analgésique, restés sans débouché thérapeutique. AB-CHMINACA est ensuite apparue sur le marché des produits fumables : Uchiyama et ses collègues la signalent dans des produits illégaux japonais analysés en 2014, et sa diffusion en Europe et en Amérique du Nord suit immédiatement, avec un pic de saisies en 2015.

AB-CHMINACA is a molecule made in a laboratory, with no connection whatsoever to hemp: it is sprayed onto dried plant material or onto paper, then sold under fanciful names. It acts on the same receptors as cannabis, but far more strongly, and the quantity deposited varies from one piece to another. Emergency services have reported convulsions, states of severe confusion and deaths after consumption of products containing it.

Pharmacology
AB-CHMINACA belongs to the synthetic cannabinoid receptor agonists, in the family of indazole-3-carboxamides with a valinamide residue. The work of Wiley and colleagues describes it as a full agonist of CB1, of nanomolar affinity, whose efficacy exceeds that of most known full agonists; in discrimination assays in rodents, it substitutes for THC at markedly lower doses. This maximal efficacy at CB1 explains the clinical gap with cannabis: where THC plateaus because it only partially activates the receptor, a full agonist does not plateau, and the interval between the sought-after effect and intoxication becomes very narrow. The literature solidly establishes the mechanism and the in vitro potency; it establishes neither a safe dose nor a predictable effect profile in humans, no controlled administration study having ever been conducted. The only human data available come from series of poisonings and from forensic investigations.

Origin (pharmaceutical research → illicit market)
No known biosynthetic pathway: the molecule exists in no living organism and derives from no plant precursor. It is obtained by chemical means, from an N-alkylated indazole-3-carboxylic acid coupled to an L-valine amide, with no connection whatsoever to the cannabigerol pathway that gives rise to the cannabinoids of hemp.
Legal framework
France
A substance classified by name as a narcotic in France. AB-CHMINACA appears explicitly in annex III of the decree of 22 February 1990 setting the list of substances classified as narcotics, under its name and its full chemical denomination, by the effect of the decree of 14 October 2019. This is therefore not an indirect attachment via the generic clause covering tetrahydrocannabinols and their esters or ethers, a clause that does not cover this chemical family: the molecule is targeted in its own right. Production, possession, transfer and use fall under the criminal regime for narcotics. This substance has no place in the wellness hemp or CBD trade.
European Union
AB-CHMINACA is listed in Schedule II of the 1971 Convention on Psychotropic Substances, as confirmed by the green list of the International Narcotics Control Board. This international classification binds all Member States of the European Union. Framework Decision 2004/757/JHA moreover defines drugs by reference to the substances covered by the 1971 Convention, so that AB-CHMINACA falls under the criminal law on narcotics throughout the Union. The molecule is also monitored by the European early warning system managed by the European drugs agency.

La molécule associe un noyau indazole, une chaîne cyclohexylméthyle portée par l'azote 1 et un résidu amide dérivé de la L-valine. Elle ne partage aucun squelette avec les cannabinoïdes du chanvre, qui reposent sur un motif dibenzopyrane ou résorcinol.
Pharmacokinetics
Les données humaines manquent : aucune étude d'administration contrôlée n'a été conduite. Chez le rongeur, une évaluation publiée en 2026 rapporte une élimination biphasique, une demi-vie d'environ 13,8 heures, un volume de distribution élevé et une clairance modérée, ce qui traduit une forte diffusion tissulaire. Une étude in vitro montre une fixation quasi complète aux protéines plasmatiques humaines, de l'ordre de 98,7 à 99,9 pour cent sur l'albumine sérique, avec compétition sur les sites de liaison de la warfarine et du tryptophane. La voie d'exposition habituelle étant l'inhalation, les effets s'installent en quelques minutes, mais la dose réellement absorbée reste inconnue de la personne qui consomme.
Metabolism
Le métabolisme est intense et la molécule mère disparaît rapidement de la circulation. Les incubations sur microsomes hépatiques humains identifient environ neuf métabolites, formés par hydrolyse de la fonction amide, oxydation, carboxylation et désalkylation de la chaîne cyclohexylméthyle ; deux d'entre eux ont été retrouvés dans des échantillons authentiques. Chez le rongeur, un dérivé dihydroxylé domine dans les urines entre huit et vingt-quatre heures après administration. Les isoformes précises du cytochrome P450 responsables de ces transformations n'ont pas été établies de façon consensuelle pour cette molécule.
Toxicology and risks
AB-CHMINACA compte parmi les agonistes de synthèse les plus souvent cités dans les revues de décès liés à ces produits, aux côtés du MDMB-CHMICA. Les tableaux cliniques rapportés associent tachycardie, convulsions, agitation, états confusionnels sévères et dépression du système nerveux central pouvant aller jusqu'au coma ; des épisodes psychotiques et des décès ont été documentés. Une série allemande portant sur des décès survenus entre 2014 et 2020 retrouve la molécule parmi les plus fréquemment détectées. Chez la souris, une administration répétée provoque des lésions histologiques du foie et du rein, y compris aux doses faibles. Aucune dose sûre ne peut être identifiée : la teneur des produits est hétérogène et l'efficacité maximale sur CB1 supprime le plafond d'effet qui limite le THC.
Detection and analysis
La recherche analytique repose principalement sur la chromatographie liquide couplée à la spectrométrie de masse en tandem, complétée par la spectrométrie de masse haute résolution pour le criblage non ciblé. La molécule mère n'est présente qu'à de très faibles concentrations dans le sang et la salive, et elle est souvent absente des urines : mettre en évidence une consommation suppose de rechercher les métabolites, donc de les avoir caractérisés au préalable. Les tests immunochimiques courants destinés au cannabis ne détectent pas cette substance.
