- L'AB-FUBINACA, dont cette molécule est un isomère de position, a été mise au point et brevetée par Pfizer en 2009 comme agoniste du récepteur CB1, avant d'être détournée puis retrouvée dans des produits à fumer. L'isomère 2-fluorobenzyle a lui-même été identifié dans des produits au Japon et à l'étranger selon le ministère japonais de la santé, du travail et du bien-être, et inscrit comme substance désignée par la loi japonaise en décembre 2015. Le laboratoire spécialisé de la Drug Enforcement Administration américaine en a publié une monographie de référence établie sur matériel structuralement confirmé, révisée en mars 2016, et l'équipe de Murakami et collaborateurs l'a caractérisé par spectrométrie de masse en 2016 puis en 2018.

The 2-fluorobenzyl isomer of AB-FUBINACA is a synthetic cannabinoid, made in a laboratory and absent from hemp. It closely resembles AB-FUBINACA, from which it differs only in the position of one fluorine atom on the ring, and it has been found in smoking products sold under other names. Almost nothing is known about its effects in human beings, since no study has assessed it; molecules of the same family, by contrast, cause severe poisonings.

Pharmacology
The 2-fluorobenzyl isomer of AB-FUBINACA is a presumed cannabinoid receptor agonist, built on the indazole-3-carboxamide skeleton common to the whole FUBINACA series. What makes this family dangerous is full agonism at the CB1 receptor: where THC activates that receptor only partially, the synthetic carboxamides can activate it maximally, without the ceiling effect that limits cannabis. For this particular isomer, however, the pharmacology remains undocumented: no binding study, no functional assay, no metabolism or pharmacokinetic data have been published, and human data are lacking. What is established concerns it as a substance encountered in the field and as an analytical problem: the Japanese Ministry of Health, Labour and Welfare reports its identification in products in Japan and abroad, it was listed as a designated substance in Japan in December 2015, and two mass spectrometry studies have developed the methods that allow it to be distinguished from AB-FUBINACA and from its other positional isomers. That distinction is no detail: narcotics lists name the para isomer, and confusing two isomers has direct consequences, judicial as well as health-related.

Origin (pharmaceutical research → illicit market)
No known biological pathway: this molecule comes from no living organism. It arises entirely from chemical synthesis, through the assembly of an indazole-3-carboxamide bearing a 2-fluorobenzyl group on the N1 nitrogen and a valinamide residue on the nitrogen of the amide function. It has never been demonstrated in hemp nor in any other plant.
Legal framework
France
In France, this isomer is not listed by name on the narcotics list, but it is captured by a generic clause. Annex IV of the decree of 22 February 1990 classifies any molecule belonging to the family of 3-carboxamide indazole derivatives bearing, on the nitrogen at position 1 of the indazole core, a substituent of the halobenzyl type, and bearing, on the nitrogen of the carboxamide bridge, a group of the 1-amino-1-oxo-butan-2-yl type, whether or not that group is substituted at position 3 by one or two alkyl groups. The 2-fluorobenzyl group and the valinamide residue of this molecule fulfil both conditions: classification as a narcotic therefore arises through a generic clause, and not through listing by name. As a point of reference, AB-FUBINACA, the para isomer, does appear by name in annex III, which reproduces the schedules of the 1971 Convention. Possessing, supplying or consuming a substance classified as a narcotic is a criminal offence in France; this paragraph describes the state of the law and does not constitute legal advice.
European Union
At international level, AB-FUBINACA, the para isomer, was placed in Schedule II of the 1971 Convention on psychotropic substances by decision of the United Nations Commission on Narcotic Drugs at its 63rd session, in March 2020, on the recommendation of the World Health Organization expert committee that met in 2019; the decision entered into force on 3 November 2020. That listing covers the named substance and its stereoisomers, not its positional isomers: the 2-fluorobenzyl isomer is therefore not caught by that text. No measure applicable across the European Union names it, and its status depends on the law of each Member State, several countries including France applying generic definitions that encompass it. The United Nations Office on Drugs and Crime records it among the synthetic cannabinoid receptor agonists in its early warning system.

Toxicology and risks
Aucun cas d'intoxication ni aucune étude toxicologique portant spécifiquement sur cet isomère n'a été retrouvé, et les données humaines manquent. Les éléments disponibles concernent l'AB-FUBINACA, l'isomère para, et appartiennent à cette molécule para, non à celle décrite ici : l'examen critique du comité d'experts de l'Organisation mondiale de la santé y rapporte, chez l'animal, une suppression de l'activité motrice, une hypothermie, une atteinte de l'équilibre, ainsi que des signes neurologiques dont convulsions, hyperréflexie, myoclonies et agressivité spontanée ; chez l'être humain, des tableaux de confusion, agitation, somnolence, hypertension et tachycardie, et au moins un décès dans lequel l'AB-FUBINACA était le seul cannabinoïde de synthèse détecté. Le risque propre à cette famille, agonisme complet du récepteur CB1 sans plafond d'effet, teneurs très variables d'un lot à l'autre et absence de toute étiquette fiable, s'applique par construction à toute molécule diffusée de cette manière.
Detection and analysis
Cet isomère est bien documenté sur le plan analytique. Le laboratoire spécialisé de la Drug Enforcement Administration a publié pour la collection SWGDRUG une monographie de référence établie sur matériel structuralement confirmé, réunissant un spectre de résonance magnétique nucléaire du proton, un spectre de masse par impact électronique après chromatographie en phase gazeuse et un spectre infrarouge. Deux travaux japonais ont ensuite traité la difficulté centrale, séparer cet isomère de l'AB-FUBINACA et de ses autres isomères de position, dont les spectres comportent les mêmes ions. En impact électronique couplé à un triple quadripôle, le rapport d'abondance des ions à m/z 109 et m/z 253 classe les trois isomères fluorés dans l'ordre méta, ortho, para, et cet écart se creuse avec l'énergie de collision. En ionisation par électronébulisation, l'association d'une trappe ionique linéaire et d'un triple quadripôle différencie les six positions étudiées à partir des rapports d'abondance des ions produits.
