Pharmacology

ABD459 is a diarylpyrazole ketone derived from the skeleton of rimonabant, characterised as a neutral antagonist at the CB1 receptor. On mouse brain membranes, it fully displaces the radioligand [3H]CP55940 with a Ki of 8.61 nmol/l and blocks the [35S]GTPγS stimulation induced by CP55940 (KB of 7.7 nmol/l), while leaving basal binding unchanged: it is precisely this last point that sets it apart from rimonabant and AM251, inverse agonists which do lower the constitutive activity of the receptor. In the mouse, intraperitoneal administration reduces food intake and active food seeking for 5 to 6 hours, with no rebound after elimination, and suppresses paradoxical sleep without altering wakefulness, slow-wave sleep or locomotion. Beyond this single piece of work, the literature is very thin: nothing has been published on CB2 affinity, on pharmacokinetics, on metabolism or on toxicity, and human data are entirely lacking. The authors themselves state that they did not verify whether the molecule escapes the adverse effects associated with rimonabant and AM251.

No biological pathway: ABD459 is formed in no living organism. It is a product of organic synthesis, described here by its chemical class alone, a ketone built on the 1,5-diarylpyrazole skeleton of rimonabant.

Pharmacokinetics

Aucune étude d'absorption, de distribution ou d'élimination n'a été publiée. Le seul repère disponible est la durée d'action observée chez la souris après administration intrapéritonéale : la réduction de la prise alimentaire se maintient environ 5 à 6 heures et la suppression du sommeil paradoxal couvre les quatre premières heures, sans effet rebond ensuite. Ni la biodisponibilité orale, ni la liaison aux protéines plasmatiques, ni la demi-vie ne sont connues.

Metabolism

Aucune étude de métabolisme n'a été publiée : les métabolites, les enzymes impliquées et les voies d'élimination restent inconnus.

Toxicology and risks

Aucune étude de toxicologie n'a été conduite sur cette molécule. Dans le travail princeps, la locomotion et la prise de boisson des souris n'étaient pas affectées, mais les auteurs indiquent explicitement ne pas avoir cherché à savoir si ABD459 partage les effets indésirables décrits pour l'AM251 et le rimonabant, tels que nausées, vomissements ou syndromes de grattage et de toilettage compulsifs ; ces observations concernent ces deux analogues, pas ABD459. Pour la classe des antagonistes et agonistes inverses du CB1, le signal de sécurité le mieux documenté reste celui du rimonabant, dont l'autorisation européenne a été suspendue puis retirée en 2008 et 2009 en raison d'effets psychiatriques (anxiété, dépression, idées suicidaires) ; là encore, il s'agit du rimonabant et non d'ABD459. Aucune exposition humaine à ABD459 n'est documentée.
