- Molécule décrite en 2015. Elle n'a pas été repérée d'abord dans des produits vendus, mais conçue au laboratoire : Samuel D. Banister, dans l'équipe de Michael Kassiou à l'université de Sydney, a synthétisé les carboxamides d'indazole déjà identifiés sur le marché, dont l'AB-PINACA, ainsi que quatre analogues indoliques inédits destinés à mesurer séparément la contribution du noyau indazole. L'AB-PICA est l'un de ces analogues. Son profil fonctionnel a été établi par Jordyn M. Stuart à l'université Macquarie, puis publié la même année dans ACS Chemical Neuroscience, où la molécule porte le numéro 15. Elle figure depuis au répertoire des agonistes des récepteurs cannabinoïdes de synthèse du système d'alerte précoce de l'ONUDC.

AB-PICA has nothing to do with CBD or with hemp: it is a laboratory molecule, born of a pharmacology study, which acts on the same receptors as cannabis but far more intensely. It belongs to the synthetic cannabinoids, those powders found sprayed onto herbs to be smoked and resold under false names, sometimes even as a CBD product. In France it is classified as a narcotic, and what is known of its effects in humans is close to nil.

Pharmacology
AB-PICA is a synthetic agonist at the cannabinoid receptors belonging to the indole-3-carboxamide family: an N-pentylated indole nucleus bearing an L-valinamide motif, that is, the indole homologue of AB-PINACA. It did not circulate on the market before being studied, since it was synthesised in 2015 in a structure-activity relationship study intended to isolate the specific contribution of the indazole nucleus. In a membrane-potential functional assay on AtT20 cells expressing the human receptors, it behaves as a full agonist at both receptors: a pEC50 of 7.92 at CB1, corresponding to an EC50 of around 12 nM, for a maximal effect of 99 % of the response to WIN 55,212-2, and near-identical values at CB2. In the same set-up, Δ9-THC shows an EC50 of 235 nM and remains a partial agonist. The decisive difference is therefore not only potency, roughly twenty times greater, but maximal intrinsic efficacy, that is, the capacity to activate the CB1 receptor fully: it is this feature that underpins the risk profile of this whole family. Moving from indole to indazole, which gives AB-PINACA, multiplies potency by a further factor of about ten. No agonist activity is observed at TRPA1 or at TRPV1, and GPR55, PPARγ and 5-HT1A have not been explored. Beyond this mechanism, the literature is almost empty: no pharmacokinetics, no metabolism study, no clinical case attributable to this molecule alone. Human data are lacking and no therapeutic benefit is established.

No biological pathway: the molecule is biosynthesised by no organism and derives from no plant precursor. It belongs to synthetic chemistry alone, through formation of an amide bond between 1-pentylindole-3-carboxylic acid and valinamide.
Legal framework
France
A narcotic in France, by attachment to a generic clause and not by a nominative listing. AB-PICA is not cited by name in any list: it falls under the generic family of "indole 3-carboxamide derivatives" introduced into annex IV of the decree of 22 February 1990 by the decree of 31 March 2017. This family covers indole-3-carboxamides bearing on the indole nitrogen an alkyl-type substituent, and on the nitrogen of the carboxamide bridge a 1-amino-1-oxo-butan-2-yl-type group, whether or not that group is substituted at position 3 by one or two alkyl groups. The N-pentyl chain and the L-valinamide motif of AB-PICA match this definition exactly. Production, possession, transfer and use therefore fall under the criminal regime for narcotics. The generic clause covering tetrahydrocannabinols, their esters and their ethers is not the one that applies here, the molecule having no structural kinship with THC.
European Union
No control measure specific to AB-PICA at European Union level: the substance has been the subject of neither a joint report nor a risk assessment, and it does not figure among the synthetic cannabinoids named in the acts extending the European definition of "drug". No nominative listing in the schedules of the 1971 Convention on Psychotropic Substances has been found, unlike its indazole homologue AB-PINACA, listed in Schedule II: the two molecules do not share the same international status. Control therefore falls to national law, several Member States relying, as France does, on generic definitions of structural families that encompass this compound.

Agoniste synthétique des récepteurs cannabinoïdes de la famille des indole-3-carboxamides, sous-groupe des valinamides : noyau indole portant une chaîne N-pentyle, fonction carboxamide en position 3 reliée à un motif L-valinamide, c'est-à-dire l'amide primaire de la L-valine. C'est l'homologue indolique de l'AB-PINACA, dont le noyau est un indazole. Pharmacokinetics
Aucune donnée de pharmacocinétique publiée pour l'AB-PICA, ni chez l'être humain ni chez l'animal. Absorption, distribution, demi-vie et durée d'action restent inconnues, et aucune relation entre une quantité administrée et un effet n'a été établie. Les seules mesures disponibles proviennent d'essais sur cellules en culture.
Metabolism
Aucune étude de métabolisme n'a été consacrée à l'AB-PICA. Les voies décrites pour d'autres membres de la famille des indole-3-carboxamides, soit l'hydrolyse du carboxamide terminal, l'hydroxylation de la chaîne N-pentyle et la glucuronoconjugaison, appartiennent à ces autres molécules et n'ont pas été vérifiées expérimentalement sur celle-ci. Aucun métabolite marqueur n'est donc validé pour un dépistage.
Toxicology and risks
Aucun cas d'intoxication humaine attribuable à l'AB-PICA n'est documenté et aucune étude de toxicologie expérimentale ne lui a été consacrée : les données humaines manquent entièrement. Ce qui est établi est son mécanisme, et il suffit à situer le risque. Là où le Δ9-THC n'active le récepteur CB1 que partiellement, l'AB-PICA l'active pleinement, avec une puissance environ vingt fois supérieure dans le même essai. Cette activation complète est ce qui explique, pour l'ensemble des agonistes de synthèse, les tableaux rapportés en toxicologie d'urgence : vomissements incoercibles, agitation, confusion, delirium, convulsions, troubles du rythme cardiaque, atteinte rénale aiguë, coma, et des décès. Ces observations portent sur d'autres membres de la famille et non sur l'AB-PICA lui-même, mais rien dans son profil pharmacologique ne permet de l'en écarter. Deux facteurs aggravent le danger réel : les produits ne portent pas d'étiquetage fiable et la substance se répartit très inégalement sur un support végétal, si bien que la quantité reçue reste inconnue et varie d'une prise à l'autre. Enfin, le 5F-AB-PICA est une molécule différente, et les résultats obtenus sur l'un ne valent pas pour l'autre.
Detection and analysis
L'AB-PICA figure parmi les 182 nouvelles substances psychoactives d'une méthode de chromatographie liquide couplée à la spectrométrie de masse en tandem validée sur sang total par Giorgetti et ses collègues en 2022 ; la molécule inchangée y est recherchée directement, faute de métabolite marqueur validé. Les immunodosages cannabis de routine ne la reconnaissent pas. Deux pièges d'identification méritent d'être signalés : le 5F-AB-PICA, analogue portant un fluor en bout de chaîne, et l'AB-PINACA, homologue à noyau indazole, ont des masses et des fragmentations voisines, si bien que la confirmation exige un étalon de référence certifié. La distinction entre l'énantiomère (S) et le mélange racémique demande en outre une séparation chirale, les répertoires chimiques recensant les deux formes.
