- Décrit en 1999 par Cecilia Hillard et ses collaborateurs du département de pharmacologie et de toxicologie du Medical College of Wisconsin, à Milwaukee, dans un article du Journal of Pharmacology and Experimental Therapeutics cosigné avec l'équipe de Roger Pertwee. Les auteurs cherchaient à combler un manque : la plupart des agonistes cannabinoïdes alors disponibles se liaient à CB1 et à CB2 avec des affinités voisines, ce qui interdisait d'attribuer un effet observé à l'un ou à l'autre récepteur. Ils ont préparé deux analogues de l'anandamide, l'arachidonylcyclopropylamide et l'arachidonyl-2-chloroéthylamide, et leur ont donné dans cette publication les sigles ACPA et ACEA. Le sigle ACPA est resté depuis la désignation d'usage de la molécule dans la littérature.

ACPA is a laboratory-made molecule modelled on anandamide, a cannabinoid the brain produces itself. It was created in 1999 as a research instrument: it acts on only one of the two cannabinoid receptors, which lets researchers work out which one does what. It does not occur in hemp, is present in no cannabidiol product, and has never been given to human beings: human data are entirely lacking.

Pharmacology
ACPA is a synthetic analogue of anandamide: the same twenty-carbon arachidonoyl chain with four cis double bonds, but a cyclopropylamide head in place of the ethanolamide group. This substitution gives it what the cannabinoid agonists of the 1990s lacked, a clear selectivity between the two receptors, with a Ki of 2.2 nM at CB1 against 0.70 µM at CB2, that is, roughly a 300-fold preference for the neuronal receptor. Its status as an agonist rests on three concordant functional measurements: inhibition of cAMP production in cells expressing human CB1, activation of G-protein coupling in cerebellar membranes, and inhibition of mouse vas deferens contractions. In mice, the molecule induces a hypothermia that the antagonist SR141716A abolishes, which confirms that it acts through CB1 in the whole animal. Relative efficacy, by contrast, has never been quantified, and it would be wrong to treat it as a full agonist comparable to the synthetic cannabinoids of the illicit market. Since 1999, ACPA has served essentially as a pharmacological probe in rodents, given intraperitoneally or by cerebral microinjection: the published work describes an anxiolytic-like effect in the elevated plus maze, a conditioned place preference abolished by CB1 blockade in the basolateral amygdala, an attenuation of the memory deficit in a streptozotocin-induced model of Alzheimer's disease, and antitumour activity in models of non-small-cell lung cancer. Two of these findings read as warnings rather than as promises: the place preference reflects a reinforcing potential, and stopping repeated administration reduces sleeping time in mice, which reproduces a withdrawal state. No clinical trial, no human pharmacokinetic data, no established therapeutic benefit: human data are entirely lacking.

Origin (research tool)
No biosynthetic pathway is known: ACPA is produced by no organism and does not arise from the plant metabolism of cannabis. It is a synthetic product obtained by forming an amide bond between arachidonic acid and cyclopropylamine, the original publication describing the preparation as a straightforward amidation of this polyunsaturated fatty acid.
Legal framework
France
ACPA is not listed by name on any French list of narcotics or psychotropic substances. It also escapes the generic clauses of annex IV of the decree of 22 February 1990: it is not a tetrahydrocannabinol, nor an ester, an ether or a salt of tetrahydrocannabinol; its structure does not contain the benzo[c]chromene core covered by the ANSM decision of 22 May 2024; and neither does it meet the clauses describing synthetic agonists built on an indole or indazole core bearing a carboxamide function. A fatty-acid amide derived from anandamide corresponds to none of these definitions, so that the applicable situation is that of an unscheduled substance, not that of a substance caught by a generic clause. This absence of scheduling does not amount to authorisation for use: the molecule is sold as a laboratory reagent, without any evaluation for human beings, and nothing allows it to be offered for consumption.
European Union
No harmonised control measure at Union level: the molecule does not appear among the substances added to the annex of Framework Decision 2004/757/JHA, and the European drugs agency has published on its subject neither an early warning system notification, nor a technical report, nor a risk assessment. Internationally, it is listed neither in the 1961 Single Convention nor in the 1971 Convention, and the WHO Expert Committee on Drug Dependence has never reviewed it. It therefore circulates within the Union as a research chemical, governed as such by chemicals regulation and not by narcotics law. National laws remain heterogeneous: several member states apply general psychoactive substances laws whose scope could cover it, without any published decision confirming this.

Amide d'acide gras polyinsaturé : chaîne arachidonoyle en C20 portant quatre doubles liaisons cis en positions 5, 8, 11 et 14, liée par une fonction amide à la cyclopropylamine. C'est un analogue de l'anandamide dont la tête éthanolamide est remplacée par un cycle cyclopropyle à trois carbones. Aucune parenté structurale avec les cannabinoïdes terpénophénoliques du chanvre, ni avec les agonistes de synthèse à noyau indole, indazole ou benzo[c]chromène.
Pharmacokinetics
Aucune donnée humaine, par aucune voie d'administration. Chez l'animal, la molécule est donnée par voie intrapéritonéale, par voie intracérébroventriculaire ou par microinjection directe dans une structure cérébrale, hippocampe, cortex préfrontal ou amygdale basolatérale selon les protocoles, ce qui contourne la question de l'absorption plus qu'il ne la résout. L'hypothermie observée chez la souris après administration systémique atteste au moins d'une exposition cérébrale effective. Le seul élément publié touchant la biodisponibilité est indirect mais parlant : une équipe a conçu une plateforme nanoparticulaire injectable dans le but explicite d'augmenter la biodisponibilité de l'ACPA et son efficacité antitumorale, ce qui signale que la molécule libre s'y prête mal. Aucune demi-vie, aucune concentration plasmatique, aucun volume de distribution n'ont été publiés, ni chez l'animal ni chez l'être humain.
Metabolism
Aucune étude de métabolisme n'a été publiée pour cette molécule : ni sur microsomes hépatiques, ni chez l'animal, ni chez l'être humain, et aucun métabolite n'a été identifié ni dosé. Le site de coupure attendu est la liaison amide, dont l'hydrolyse libérerait de l'acide arachidonique et de la cyclopropylamine, mais cette réaction n'a pas été démontrée pour l'ACPA et sa stabilité vis-à-vis de l'amide hydrolase des acides gras, la FAAH, n'a fait l'objet d'aucune mesure publiée. Les données disponibles sur d'autres amides d'acides gras appartiennent à ces molécules et ne peuvent pas lui être transposées.
Toxicology and risks
Aucune étude de toxicologie réglementaire, aucun cas d'intoxication humaine, aucun décès rapporté : la molécule n'a jamais circulé comme produit de consommation et n'apparaît pas dans les signalements de l'observatoire européen. Ce vide documentaire ne vaut pas innocuité, et deux observations animales appellent à la prudence. La première est un potentiel de renforcement : chez la souris, l'administration intrapéritonéale produit une préférence de place conditionnée, supprimée par le blocage des récepteurs CB1 dans l'amygdale basolatérale, ce qui est la signature d'un effet gratifiant passant par ce récepteur. La seconde est un sevrage : l'arrêt d'une administration répétée diminue le temps de sommeil total pendant la phase claire, augmente les redressements et réduit les comportements de toilettage, tableau que les auteurs présentent comme un modèle murin du sevrage cannabique. À cela s'ajoutent les effets attendus d'un agoniste CB1 puissant chez le rongeur, l'hypothermie au premier rang. Le travail de formulation nanoparticulaire en oncologie expérimentale mentionne une fenêtre de doses jugée sûre chez l'animal, mais il ne s'agit pas d'une étude de sécurité et cela ne dit rien de la tolérance humaine. Enfin, l'affinité de l'ACPA pour CB1, de l'ordre du nanomolaire, dépasse largement celle du THC, ce qui rend toute extrapolation depuis l'expérience du cannabis trompeuse.
Detection and analysis
Aucune méthode de dépistage ne cible cette molécule et aucune publication de médecine légale ne lui est consacrée. Elle ne figure pas dans les panels toxicologiques courants, urinaires, sanguins ou salivaires, et les tests immunologiques destinés au cannabis ne la reconnaissent pas. Aucune méthode analytique dédiée, aucun marqueur urinaire, aucun seuil de détection n'ont été publiés, ce qui découle de l'absence d'étude de métabolisme et de l'absence de circulation de la substance en dehors du laboratoire.
