- La série RCS est apparue dans les produits à fumer vendus en ligne autour de 2010. La Nouvelle-Zélande a interdit nommément cette molécule, désignée comme l'isomère 2-méthoxy de l'homologue butyle du RCS-4, le 16 août 2011. Sa pharmacologie n'a été caractérisée qu'en 2015, par Banister et ses collègues, dans une étude consacrée au RCS-4, à ses régioisomères et à ses homologues en C4.

In plain terms
RCS-2-butyl is a wholly laboratory-made molecule that does not occur in hemp. It belongs to the family of synthetic cannabinoids, those products sprayed onto herbs and then resold under misleading names. Its presence in an article sold as CBD would be a fraud, never a legitimate ingredient.

Pharmacology
RCS-2-butyl acts on both receptors of the endocannabinoid system. In the only pharmacological work available, carried out on AtT-20 cells expressing the human receptors and the GIRK channel, it behaves as an agonist with an EC50 of 178 nM at CB1 and of 4.5 nM at CB2, that is a preference of about fortyfold for CB2. This imbalance is the notable point: the potency measured at CB2 clearly exceeds that measured at CB1. Maximal efficacy has not been reported separately for this compound, so it cannot be asserted that it behaves as a full CB1 agonist as many molecules of its class do, whereas THC is only a partial agonist. No activity has been studied at TRPV1, GPR55, PPARγ or 5-HT1A. Human data are entirely lacking: no pharmacokinetics, no metabolism and no clinical series concern this particular molecule.

None. This molecule arises from no plant or animal biosynthetic pathway: it belongs to the chemistry of the aminoalkylindoles and is obtained by acylation of an indole bearing a butyl chain on the nitrogen.
Legal framework
France
This molecule is not cited by name in the decree of 22 February 1990 setting the list of substances scheduled as narcotics. The generic clause of annex IV covers tetrahydrocannabinols, their esters, ethers and salts: it does not reach it, since this is an aminoalkylindole and not a THC derivative. France has so far proceeded by successive listings by name for synthetic cannabinoids, starting with the scheduling of JWH-018, CP 47,497 and its homologues and HU-210 in February 2009. Absence from the list amounts to neither authorisation nor safety: this molecule is an ingredient of no authorised consumer product, and its presence in an article sold as CBD constitutes adulteration.
European Union
No control measure at European Union level specifically covers this molecule. The Union proceeds substance by substance, following a risk assessment conducted by the EUDA, which has been done for several far more widespread synthetic agonists, but not for this one. Scheduling therefore depends on national laws, several Member States covering this type of structure through generic definitions of chemical families. Outside the Union, New Zealand banned it by name as early as 16 August 2011.

Pharmacokinetics
Aucune étude de pharmacocinétique humaine ou animale n'a été publiée sur cette molécule: absorption, distribution, demi-vie et biodisponibilité ne sont pas documentées. Les cannabinoïdes de synthèse de ce type circulent pulvérisés sur un support végétal, ce qui rend la quantité réellement reçue très irrégulière d'un fragment à l'autre d'un même sachet.
Metabolism
Aucune étude de métabolisme ne porte sur cette molécule. Les voies de biotransformation, les métabolites majeurs et les enzymes impliquées ne sont pas caractérisés, ce qui complique aussi l'interprétation des analyses biologiques, celles-ci reposant le plus souvent sur la recherche de métabolites connus.
Toxicology and risks
Aucun cas clinique, aucune série d'intoxication et aucun décès ne sont rattachés de façon documentée à cette molécule précise, qui a peu circulé et n'a pas fait l'objet d'un suivi toxicologique propre. Le risque s'apprécie donc au niveau de sa classe. Les agonistes cannabinoïdes de synthèse sont associés dans la littérature à des convulsions, une agitation et un état confusionnel, des épisodes psychotiques parfois prolongés, une tachycardie et des troubles du rythme, des vomissements répétés, une atteinte rénale aiguë, ainsi qu'à des décès. Leur dangerosité tient à l'agonisme complet sur le récepteur CB1, sans plateau d'effet, là où le THC n'est qu'un agoniste partiel. Ce point n'a pas été vérifié expérimentalement pour le RCS-2-butyl, dont seule la puissance a été mesurée. Le danger le plus concret reste le mode de diffusion: ces molécules sont pulvérisées sur des végétaux, s'y répartissent de façon très inégale et sont vendues sous une autre identité, parfois celle d'un produit au CBD.
Detection and analysis
Cette molécule échappe aux tests urinaires courants, qui ne ciblent pas les cannabinoïdes de synthèse. Son identification repose sur la chromatographie couplée à la spectrométrie de masse, avec la difficulté propre à cette série: l'isomère ortho, l'isomère para et les homologues à chaîne butyle ou pentyle donnent des réponses très voisines, si bien qu'un étalon de référence certifié est nécessaire pour trancher. C'est précisément cette question de différenciation qui a motivé l'étude de 2015 sur le RCS-4, ses régioisomères et ses homologues en C4.
