Pharmacology

JWH-368 belongs to the naphthoylpyrroles, a family of synthetic cannabinoids arising from the work of John W. Huffman. In the single published study that tested it, it binds strongly to both cannabinoid receptors: a Ki of 16 nM at the CB1 receptor and of 9.1 nM at the cloned human CB2 receptor. That work measured binding affinity only: the actual ability of JWH-368 to activate the receptor has never been assessed. The distinction matters, because what makes synthetic cannabinoids dangerous is their full agonism at the CB1 receptor, where THC is only a partial agonist; this property is established for the class, not for this molecule in particular. Human data are entirely lacking.

No biological pathway: JWH-368 is entirely synthetic. It is obtained by acylation of an N-alkylated pyrrole bearing an aryl group, with no plant precursor and no enzymatic step.

Aucune étude de toxicité, aucun cas clinique et aucun décès liés au JWH-368 lui-même ne sont publiés : la molécule est restée un outil de laboratoire et n'a pas été identifiée dans les mélanges végétaux vendus à fumer. Les dangers documentés pour la classe des cannabinoïdes de synthèse, convulsions, états confusionnels, troubles du rythme cardiaque, vomissements incoercibles, atteintes rénales aiguës et décès, concernent d'autres composés de cette famille, ceux qui ont réellement circulé, et non celui-ci. Le risque tient ici à la structure : forte affinité pour le récepteur CB1 et absence complète de données humaines.
