La forme racémique remonte aux travaux des années 1940 sur la synthèse des tétrahydrocannabinols, menés par les équipes de Todd puis de Roger Adams, qui dépose en 1947 un brevet américain décrivant le composé. Les deux énantiomères n'ont été séparés qu'en 1984 par Srebnik, Lander, Breuer et Mechoulam, à partir des stéréoisomères du Δ10-THC. Rosati et ses collaborateurs ont publié en 2014 un accès direct à chaque énantiomère par condensation d'un résorcinol avec la pulégone, et c'est ce travail qui a fourni la première mesure d'affinité de la molécule pour le récepteur CB1.

(S)-Δ3-THC is not a product of hemp: no plant enzyme forms it and it is not among the cannabinoids isolated from Cannabis sativa. It comes exclusively from laboratory chemistry. In the field, Δ6a,10a-THC appears as a by-product of the conversion processes used to make other THC isomers, and official laboratories have found it in seized vaping liquids, where it most often accompanies Δ10-THC. Since routine analyses do not separate the two enantiomers, the exact share of the (S) form in these products remains unknown.

In plain terms

(S)-Δ3-THC is a chemical cousin of THC made in a laboratory: the same molecular formula, with the double bond placed elsewhere on the skeleton. Hemp does not produce it, and the molecule exists only by synthesis; official laboratories sometimes find it as a by-product in vaping liquids. The few trials conducted in humans, in the 1970s and 1980s, describe effects of the same nature as those of THC but several times weaker, and the substance is scheduled as a narcotic.

Pharmacology

(S)-Δ3-THC, also written (S)-Δ6a,10a-THC, shifts the double bond of Δ9-THC towards the ring junction: the skeleton flattens and the two asymmetric junction carbons disappear, leaving only one stereogenic centre at C9. This modification does not abolish affinity for the CB1 receptor, quite the contrary: the only published measurement, in 2014, gives 5 nM at human CB1 for the (S) form and 29 nM for the (R) form, and the [35S]GTPγS assays classify the series among the partial agonists, the same status as Δ9-THC and distinct from the full agonism of the synthetic cannabinoids. Stereoselectivity is the best-established fact in this file: in human volunteers, the (S) enantiomer produced psychic effects qualitatively comparable to those of Δ9-THC but three to six times less potent, whereas the (R) enantiomer proved inactive and the mixture of the two did not increase the effect. In pigeons trained to discriminate Δ9-THC, the acetate esters of both enantiomers substitute for Δ9-THC, the (1S) form being distinctly the more potent. Beyond this older work, the file is thin: human pharmacokinetics has never been described, the metabolism specific to this isomer has not been characterised, and neither affinity for CB2 nor the TRPV1, GPR55, PPARγ and 5-HT1A targets are documented for this molecule.

Route of preparation (chemical process)

No known enzymatic pathway: hemp does not produce this isomer. The molecule is obtained by chemical means, either by condensation of a resorcinol with pulegone, or by isomerisation of other tetrahydrocannabinols under acidic conditions. These routes give the racemate, and obtaining the (S) enantiomer alone requires starting from an already resolved precursor.

Cannabinoïde de type THC : squelette dibenzo[b,d]pyrane, chaîne latérale pentyle en position 3, double liaison portée par la jonction des cycles 6a,10a. Ce déplacement aplanit le système et supprime les deux carbones asymétriques de jonction du Δ9-THC : il ne reste qu'un seul centre stéréogène, le carbone 9 porteur du méthyle, si bien que la molécule existe en deux énantiomères, (9S) et (9R). Cette fiche décrit uniquement la forme (S). L'ancienne numérotation monoterpénique appelle ce composé Δ3-THC, la numérotation dibenzopyrane Δ6a,10a-THC.

Pharmacokinetics

Aucune étude de pharmacocinétique humaine n'a été publiée pour cet isomère. Le comité d'experts de l'OMS relève que les données propres aux isomères du THC sont rares et raisonne par analogie : comme les autres tétrahydrocannabinols, la molécule est très lipophile, donc absorbée et distribuée vers le cerveau par voie orale, inhalée ou parentérale, avec un métabolisme initial hépatique assuré par les cytochromes P450. Le fait que la forme fumée ait produit des effets psychiques chez le volontaire humain établit au moins qu'elle atteint le système nerveux central. Demi-vie, biodisponibilité et volume de distribution restent inconnus.

Metabolism

Le métabolisme propre au Δ6a,10a-THC n'a été caractérisé ni chez l'humain ni chez l'animal. Le comité d'experts de l'OMS considère que le squelette tétrahydrocannabinol commun laisse attendre les mêmes réactions que pour les autres isomères, c'est-à-dire une hydroxylation initiale par les cytochromes P450 hépatiques, suivie d'une oxydation puis d'une conjugaison. Mais aucun métabolite de ce composé n'a été identifié à ce jour, et aucun marqueur urinaire spécifique n'a été validé.

Toxicology and risks

Le profil toxicologique n'est pas établi, et les seules données disponibles portent sur la forme racémique. Chez la souris, l'administration intraveineuse à forte dose n'a pas entraîné de mortalité, ce que le comité d'experts de l'OMS interprète comme une toxicité aiguë faible. Chez le lapin génétiquement sensible aux convulsions induites par le Δ9-THC, le composé s'est montré environ quinze fois moins puissant que le Δ9-THC. Chez l'humain, les quelques volontaires exposés par voie fumée dans les années 1970 et 1980 ont rapporté des effets moins marqués et plus brefs que ceux du Δ9-THC, avec sensations vertigineuses, engourdissements du visage et des extrémités, fatigue, sueurs froides et somnolence ; aucune étude systématique de tolérance ou de sécurité n'a jamais été conduite. Le risque concret aujourd'hui ne tient pas à la molécule pure mais aux produits : lorsqu'un Δ6a,10a-THC se trouve dans un liquide de vapotage, il n'est pas déclaré sur l'étiquette et s'accompagne d'autres isomères ainsi que de résidus de réaction non caractérisés.

Detection and analysis

La chromatographie en phase gazeuse couplée à la spectrométrie de masse, après dérivation triméthylsilylée, sépare le Δ6a,10a-THC des autres isomères du THC : son spectre de masse se distingue par une fragmentation nettement plus faible et moins d'ions de masse élevée. C'est par cette voie qu'un laboratoire fédéral américain l'a identifié dans des liquides de vapotage saisis, à des teneurs notables dans une partie des échantillons et à l'état de traces dans beaucoup d'autres. Limite importante : sur colonne achirale, les énantiomères (9S) et (9R) coéluent complètement, si bien qu'une analyse de routine confirme la présence du Δ6a,10a-THC sans dire lequel des deux énantiomères est en cause. Les méthodes plus anciennes reposaient sur la chromatographie sur couche mince, la chromatographie liquide et les tests microcristallins.
