In plain terms
AM-679 is a molecule made entirely in the laboratory: it does not exist in hemp and has nothing in common with CBD. Designed at the end of the 1990s for research, it was found from 2011 onwards in powders and plant mixtures sold as 'legal' products. It acts on the same receptors as THC, and no data on its use in humans have been published.

Pharmacology
AM-679 is a benzoylindole: a synthetic cannabinoid receptor agonist built on an indole ring bearing a pentyl chain on the nitrogen and a 2-iodobenzoyl group at position 3. The only pharmacological characterisation to appear in a peer-reviewed journal is that of Marusich and collaborators (2018), which measures an affinity of 39.6 nM at the human CB1 receptor and 131.7 nM at CB2, with low-amplitude G protein activation at CB1 (EC50 of 880.6 nM). In mice, the molecule substitutes for THC in stimulus discrimination, which marks cannabinoid-type effects, but it proved the least potent of the eight substances tested, slightly below THC itself. This point deserves to be stated plainly: synthetic cannabinoid agonists are dangerous because they behave as full agonists at the CB1 receptor whereas THC is only a partial agonist, and the data specific to AM-679 place it at the bottom of the affinity and efficacy scale of this series. Higher affinity values, on the order of 13.5 nM at CB1, also circulate, but they come from the patent literature and not from a peer-reviewed publication. Nothing has been published on any possible TRPV1, GPR55, PPARγ or 5-HT1A activity, and human data are entirely lacking.

No known biosynthetic pathway: the molecule is entirely synthetic, obtained by acylation of an N-alkylated indole, and has no plant equivalent.

France
AM-679 is classified as a narcotic in France, and it is so by name. It appears in annex IV of the decree of 22 February 1990, in the wording arising from the decree of 19 May 2015 and then the decree of 31 March 2017, under the entry 'AM-679 or (2-iodophenyl)(1-pentyl-1H-indol-3-yl)-methanone', within the benzoylindole family; the generic definition of (indol-3-yl)methanone derivatives also covers it. This is therefore not a case of applying the generic clause covering tetrahydrocannabinols, their esters and ethers: this molecule is not a THC and it is listed under its own name.
European Union
No control measure specific to the European Union level targets AM-679: the substance has not been the subject of a risk assessment by the EUDA (formerly the EMCDDA) or of a listing under Directive (EU) 2017/2103, and it does not appear in the schedules of the United Nations conventions. Control therefore falls to national laws, with situations differing from one country to another: listing by name in France, coverage by the German law on new psychoactive substances (NpSG), Class B classification in the United Kingdom.

Pharmacokinetics
Aucune donnée de pharmacocinétique humaine n'a été publiée. Les seules expériences in vivo disponibles ont été conduites chez la souris par voie intrapéritonéale et ne renseignent ni l'absorption par inhalation, qui est pourtant la voie d'exposition réelle, ni la demi-vie, ni la distribution.
Metabolism
Le métabolisme de l'AM-679 n'a pas été caractérisé : aucune étude sur microsomes ou hépatocytes humains, aucun profil de métabolites urinaires n'a été publié pour cette molécule. Les schémas décrits pour d'autres benzoylindoles, comme l'AM-694, appartiennent à ces molécules et ne peuvent pas lui être transposés.
Toxicology and risks
Aucune série de cas et aucun décès spécifiquement attribués à l'AM-679 n'ont été publiés, ce qui traduit sa rareté et l'absence de recherche ciblée en toxicologie analytique plutôt qu'une innocuité démontrée. Pour la classe des agonistes cannabinoïdes de synthèse, la littérature clinique documente convulsions, agitation et états délirants, tachycardie et troubles du rythme, vomissements incoercibles, atteintes rénales aiguës et décès ; ces observations concernent la classe et d'autres molécules, pas l'AM-679 lui-même. Le risque concret tient au produit fini : la teneur y est inconnue et très inégale d'un endroit à l'autre du même sachet, et l'étiquette ne correspond pas au contenu.
Detection and analysis
L'AM-679 a été identifié dans des saisies par chromatographie en phase gazeuse couplée à la spectrométrie de masse, par chromatographie liquide couplée à la spectrométrie de masse en tandem et à temps de vol, ainsi que par résonance magnétique nucléaire (Jankovics et al., 2012). Une seconde équipe a confirmé cette identification dans des herbes et des poudres et a proposé un dosage par chromatographie en phase gazeuse avec détection par ionisation de flamme (Gregori et al., 2013). La molécule échappe aux tests urinaires courants du cannabis, qui ciblent les métabolites du THC, et aucune méthode validée en matrice biologique ne lui est spécifiquement consacrée.
