- La chimie dont AM-087 est issu remonte au début des années 1990 : Charalambous et coll. (1991) évaluent une série d'analogues halogénés du Δ8-THC et montrent que les halogènes volumineux placés en bout de chaîne latérale augmentent nettement la puissance, résultat systématisé ensuite par Nikas et coll. (2004) sous la forme d'un modèle de pharmacophore CB1. AM-087, qui combine cette bromation terminale avec une ramification diméthyle en C1', n'a pas de publication princeps identifiable sous ce code dans les bases indexées : les valeurs qui lui sont associées apparaissent dans des travaux de modélisation moléculaire des années 2000.

Pharmacology
AM-087 belongs to the classical cannabinoids, the structural family of THC: the dibenzopyran skeleton of Δ8-THC is retained, with the side chain bearing two methyl groups at C1' and a bromine on its terminal carbon. These two modifications sharply increase affinity for the CB1 receptor, with a reported value of 0.43 nM, roughly two orders of magnitude below the values usually measured for THC, while a [35S]GTPγS functional assay on rat cerebellum places the EC50 at around 10 nM with marked activation of G proteins. The pharmacological point that matters here is efficacy: THC is only a partial agonist at CB1, whereas synthetic cannabinoid agonists most often behave as full agonists, which makes their effects far harder to contain and explains the severity of the poisonings observed in this family. For AM-087 itself, CB2 affinity, selectivity, pharmacokinetics and toxicology remain absent from the literature: human data are entirely lacking, and results obtained on closely related halogenated analogues, which differ by a single substituent, cannot be attributed to it.

No biosynthetic pathway: the molecule does not exist in the plant. It is obtained by synthesis from the tetrahydrocannabinol skeleton, the side chain being branched at C1' and then halogenated on its terminal carbon.
Legal framework
France
AM-087 is named in no ANSM decision, so no nominative listing concerns it. It is the generic text that applies: annex IV of the decree of 22 February 1990 classifies as narcotics tetrahydrocannabinols, their esters, ethers and salts, and AM-087 is a tetrahydrocannabinol of which only the side chain has been modified, which brings it within the scope of that generic entry. In practice, the consequence is simple: a laboratory reagent, no sale, no lawful food or consumption use, and no presence in the products of a CBD shop.
European Union
No control measure at Union level targets AM-087: the sources consulted mention neither a joint report nor a EUDA risk assessment concerning it, unlike several synthetic cannabinoid agonists placed under European control after such an assessment. Its status therefore depends on national law, some Member States classifying tetrahydrocannabinols by generic families and others by nominative lists. Internationally, AM-087 is not an isomer of THC, its molecular formula being different, and it does not appear by name in the schedules of the 1961 and 1971 conventions.

Pharmacokinetics
Aucune étude d'absorption, de distribution ou d'élimination n'a été publiée pour AM-087. Le seul repère voisin porte sur une molécule différente, le 5'-18F-Δ8-THC, dont la biodistribution chez la souris et l'imagerie chez le babouin montrent une captation cérébrale rapide suivie d'une élimination rapide (Charalambous et coll., 1991) : ces données appartiennent à cet analogue et non à AM-087.
Metabolism
Le métabolisme d'AM-087 n'a pas été décrit : ni voies d'oxydation, ni métabolites, ni enzymes impliquées ne figurent dans les sources consultées.
Toxicology and risks
Aucune donnée de toxicologie propre à AM-087, chez l'animal comme chez l'humain, n'a été retrouvée. Le risque se déduit de la pharmacologie du récepteur : une affinité CB1 de l'ordre de 0,4 nM associée à une efficacité élevée place cette molécule parmi les agonistes cannabinoïdes de synthèse, famille dont les intoxications documentées associent tachycardie, agitation, vomissements, convulsions et troubles de la conscience, avec des décès rapportés pour les composés réellement diffusés (Yano et coll., 2023). Le facteur aggravant reconnu de cette famille est l'agonisme plein sur CB1, très supérieur en efficacité au THC qui n'est qu'un agoniste partiel (Zagzoog et coll., 2021), et le fait qu'une modification structurale minime suffise à faire varier fortement cette efficacité. AM-087 ne figure pas dans les relevés de produits saisis ni dans les inventaires de substances circulant en ligne qui ont été consultés : le retour d'expérience clinique est donc nul, ce qui constitue en soi une inconnue et non une garantie.
Detection and analysis
Aucune méthode analytique validée spécifique d'AM-087 ne figure dans les sources consultées, et la molécule n'appartient à aucun panel de dépistage courant. Les immunoessais urinaires visant le cannabis reconnaissent les métabolites du THC et ne sont pas conçus pour un analogue à chaîne latérale modifiée ; une identification formelle exigerait une chromatographie couplée à la spectrométrie de masse avec un matériau de référence certifié.
