In plain terms

AM-1235 is a synthetic cannabinoid made in the laboratory: it exists in no plant and has nothing in common with CBD. It was created in the late 1990s by an American university team to study cannabis receptors, never to be consumed. Products of this family are sometimes found sprayed onto plant material sold as cannabis or CBD, which makes their actual composition unpredictable.

Pharmacology

AM-1235 belongs to the naphthoylindoles, the family of cannabinoid receptor agonists opened by WIN 55,212-2 and the JWH compounds. Its structure combines an indole nucleus nitrated at position 6, a 5-fluoropentyl chain on the indole nitrogen and a naphthalene-1-carbonyl group at position 3. The original patent reports an affinity of 1.5 nM for the CB1 receptor and of 20.4 nM for the CB2 receptor, hence a selectivity of about thirteenfold in favour of CB1. An affinity, however, says nothing about functional efficacy, and no published measurement of intrinsic activity relates specifically to AM-1235. The toxicological literature describes the marketed synthetic cannabinoids as full CB1 agonists, whereas THC is only a partial agonist: this difference in efficacy, more than potency alone, explains the severity of the poisonings observed in this class. For AM-1235 itself, human data are entirely lacking, and the only studies that have actually analysed the molecule are analytical work.

Pharmacokinetics

Aucune étude de pharmacocinétique publiée ne porte sur l'AM-1235. Ni l'absorption, ni la distribution, ni la demi-vie d'élimination n'ont été décrites, chez l'animal comme chez l'humain.

Metabolism

Le métabolisme de l'AM-1235 n'a pas été caractérisé dans la littérature publiée. Les voies décrites pour d'autres cannabinoïdes de synthèse porteurs d'une chaîne 5-fluoropentyle sont des données propres à ces analogues et ne peuvent pas être transposées à l'AM-1235, dont le groupement nitro modifie le comportement chimique.

Toxicology and risks

Aucun cas clinique, aucune intoxication et aucun décès publiés ne mettent en cause l'AM-1235 : les données humaines manquent totalement. Les risques connus sont ceux de la classe des agonistes cannabinoïdes de synthèse, documentés notamment par une revue systématique portant sur 74 décès publiés : convulsions, troubles du rythme cardiaque, agitation et confusion, dépression respiratoire, rhabdomyolyse, atteintes rénales et hépatiques. Ces observations concernent d'autres molécules de la même famille et non l'AM-1235. Le danger propre à cette classe tient à son agonisme complet du récepteur CB1, alors que le THC n'en est qu'un agoniste partiel, et au fait que les produits vendus échappent à toute vérification de composition.

Detection and analysis

L'AM-1235 fait partie des cannabinoïdes de synthèse pour lesquels il existe des données analytiques propres. Une méthode UHPLC-MS/MS validée pour 182 nouvelles substances psychoactives dans le sang total l'inclut, avec une limite de détection et de quantification de 1,25 nanogramme par millilitre et un coefficient de détermination de 0,995, les auteurs précisant que la validation complète n'a pas été atteinte au niveau de contrôle bas. En chromatographie gazeuse couplée à la spectrométrie de masse par impact électronique, le groupement nitro de l'indole supprime le fragment attendu vers m/z 232 ou 233, ce qui a fait échouer un modèle de classification automatique sur cette molécule : les analogues de cette famille ne se distinguent pas de façon fiable sans spectrométrie haute résolution. Les immunoessais de routine, eux, ne détectent pas les cannabinoïdes de synthèse.
