Pharmacology

AM-1248 belongs to the aminoalkylindoles and bears an adamantyl group at position 3 of the indole. The literature describes it as an agonist at the CB1 receptor: in a functional assay measuring the opening of GIRK channels, its efficacy exceeds that of JWH-018, while its potency is the lowest of the panel compared. This efficacy is the decisive difference from THC, only a partial agonist at CB1, whose effect plateaus; a full agonist does not plateau, which explains the severity of the poisonings observed in this class. Nanomolar activity at the CB2 receptor is documented, with divergent responses depending on whether cyclic AMP or beta-arrestin recruitment is measured. At the concentration producing its maximal CB1 effect, AM-1248 did not activate the TRPV1 channel, unlike other compounds tested in parallel. Human data are lacking: neither a clinical study nor a published case series exists for this particular molecule.

No biological pathway: the molecule is produced by no living organism. It results from a laboratory synthesis by acylation of a substituted indole, with no known natural route.

Pharmacokinetics

Aucune donnée de pharmacocinétique publiée, ni chez l'animal ni chez l'humain, pour ce composé. Les bases de chimie le décrivent comme très lipophile, avec une violation de la règle de Lipinski, ce qui reste une valeur calculée et non une mesure in vivo.

Metabolism

Le devenir métabolique de l'AM-1248 n'a pas été cartographié et aucune étude de métabolites n'a été publiée. Un travail sur microsomes hépatiques de souris a montré que les dérivés adamantoyl-indoles, dont l'AM-1248, n'inhibent pas l'activité du cytochrome P450 1A, alors que les naphtoylindoles l'inhibent de façon compétitive. Ce résultat renseigne sur un risque d'interaction, pas sur les voies de transformation elles-mêmes.

Toxicology and risks

L'AM-1248 s'est révélé renforçant chez le rat dans un protocole d'auto-administration intraveineuse, avec un comportement maintenu de façon stable et une hausse des appuis sur le levier inactif, que les auteurs interprètent comme une composante impulsive. Ils concluent à un potentiel d'abus réel. Aucune donnée de toxicité humaine ne porte spécifiquement sur cette molécule. Pour la classe des agonistes complets de CB1, la littérature documente convulsions, delirium, tachyarythmies, vomissements incoercibles, atteintes rénales aiguës et décès ; ces observations concernent d'autres composés de la famille et ne peuvent pas être transposées telles quelles à l'AM-1248.

Detection and analysis

Aucune méthode publiée n'est dédiée spécifiquement à l'AM-1248 dans les matrices biologiques. Comme les autres cannabinoïdes de synthèse, ce composé échappe aux tests urinaires immunologiques du cannabis, qui ciblent les métabolites du THC, et son identification repose sur la chromatographie couplée à la spectrométrie de masse en laboratoire spécialisé. Sa présence sur un produit végétal ne se voit pas et ne se sent pas.
