AM-411 is a molecule made in a laboratory, which hemp does not produce. Chemists took the structure of THC and replaced its small side chain with a rigid block, adamantane, which makes it markedly more active at the cannabinoid receptors of the brain. It serves solely as a research tool in animals and has never been studied in human beings.

Pharmacology
AM-411 is a so-called classical cannabinoid: the tricyclic skeleton of Δ8-tetrahydrocannabinol, whose pentyl chain at position 3 is replaced by a 1-adamantyl group. This bulky substitution strengthens affinity for the CB1 receptor, measured at 6.8 nM on rat brain membranes, against 52 nM for CB2, that is to say a modest preference by a factor close to eight. First presented as a full CB1 agonist, it was subsequently reclassified as a partial agonist, with the lowest β-arrestin recruitment efficacy of the adamantylated series. In rats it reproduces the behavioural profile of Δ9-THC and substitutes for it in stimulus discrimination assays, and its effects are reversed by the CB1 antagonist SR-141716, which confirms a central action mediated by CB1. Repeated administration causes profound tolerance in monkeys. Nothing is published on TRPV1, GPR55, PPARγ or 5-HT1A, and human data are entirely lacking.

Origin (pharmaceutical research → illicit market)
No known biological pathway: AM-411 is produced by no plant and no micro-organism. It is obtained solely by chemical synthesis, by grafting an adamantyl group onto the tricyclic benzo[c]chromene-type skeleton of Δ8-tetrahydrocannabinol.

AM-411 is named in no ANSM decision. It does, however, fall under a generic clause. Annex IV of the decree of 22 February 1990 classifies tetrahydrocannabinols, their esters, ethers and salts; the ANSM decision of 22 May 2024, applicable from 3 June 2024, added to it cannabinoids built on the benzo[c]chromene core, hydrogenated or not on ring A and substituted at positions 1 to 4. AM-411 is precisely a benzo[c]chromene of that kind, bearing a hydroxyl at position 1 and an adamantyl at position 3, and therefore falls under this family definition without appearing there under its own name. The status retained is that of a substance controlled by generic clause, and not of a substance classified by name.
European Union
No European text targets AM-411 by name. The molecule has been the subject of no risk assessment by the EUDA nor of a control measure at Union level, and it appears in no schedule of the international conventions of 1961 and 1971. National situations diverge: several Member States apply generic definitions of cannabinoid families that encompass it, others keep to lists by name. It is authorised nowhere in the Union as a food or cosmetic ingredient.

Pharmacokinetics
Aucune étude pharmacocinétique dédiée à l'AM-411 n'a été publiée. Les travaux animaux établissent seulement qu'une administration par voie systémique suffit à produire des effets centraux chez le rat, donc que la molécule franchit la barrière hémato-encéphalique, et que sa puissance in vivo est élevée. Le groupe adamantyle est très lipophile, ce qui favorise la fixation tissulaire, mais aucun paramètre chiffré, demi-vie, biodisponibilité ou volume de distribution, n'est disponible pour ce composé.
Metabolism
Le métabolisme de l'AM-411 n'a fait l'objet d'aucune publication : ni les métabolites, ni les enzymes impliquées, ni les voies d'élimination ne sont décrits. Les mesures de stabilité microsomale obtenues sur d'autres cannabinoïdes adamantylés portent sur ces molécules-là et ne se transposent pas à l'AM-411.
Toxicology and risks
Il n'existe aucune donnée humaine pour cette molécule précise : ni essai clinique, ni cas d'intoxication publié, ni décès rapporté. Chez l'animal, l'AM-411 produit les effets attendus d'un agoniste CB1, dont la suppression de l'activité locomotrice et du redressement, effets normalisés par l'antagoniste SR-141716 ; l'administration répétée entraîne une tolérance profonde chez le singe, signe d'une désensibilisation marquée du récepteur. Le risque documenté est celui de la classe : l'ANSM décrit, pour les cannabinoïdes de synthèse vendus au public, des vomissements, des convulsions, une paranoïa, une hypertension artérielle, une tachycardie, des douleurs thoraciques, une perte de connaissance pouvant aller jusqu'au coma, ainsi que des cas d'insuffisance rénale, avec des effets plus sévères que ceux du cannabis et un risque de dépendance. Une précaution s'impose enfin sur les codes : l'AM-411 partage son préfixe avec des molécules très différentes de la même série, comme l'AM-2201 ou l'AM-694, dont les données ne sont pas les siennes.
Detection and analysis
Aucune méthode analytique validée spécifique de l'AM-411 n'a été publiée, et la molécule n'appartient pas aux bibliothèques spectrales de routine des laboratoires de toxicologie. Pour les cannabinoïdes de synthèse en général, l'ANSM rappelle que les tests immunochimiques urinaires classiques sont insuffisants et que la recherche doit passer par une chromatographie liquide couplée à la spectrométrie de masse en tandem, sur sang ou sur urine, complétée si possible par l'analyse du produit lui-même.
