- Décrite en 1996 par Jakob Busch-Petersen, Marcus A. Tius, Alexandros Makriyannis et leurs collaborateurs, dans le Journal of Medicinal Chemistry. Le sigle AM renvoie aux initiales d'Alexandros Makriyannis, dont le laboratoire a produit toute la série. Le programme, soutenu par l'institut national américain sur l'abus des drogues, cherchait à préciser les exigences stéréochimiques de la chaîne latérale des cannabinoïdes en bloquant sa rotation autour de la liaison C1'-C2'. Cinq analogues ont été comparés dans la même série : l'isomère cis, l'AM-906, s'est révélé le plus affin pour CB1 et le plus sélectif du groupe.

In plain terms
AM-906 is a molecule made in the laboratory in the 1990s to study how the receptors on which cannabis acts work. It does not occur in hemp and has never been sold as a consumer product, but it binds about sixty times more strongly than THC to the main receptor in the brain. No study has been conducted in humans, and its name circulates chiefly on online forums.

Pharmacology
AM-906 is a classical cannabinoid of the AM series: a hexahydrocannabinol scaffold bearing a hydroxymethyl group at position 9, and a hept-1-enyl side chain of cis geometry at position 3, whose double bond blocks rotation about the C1'-C2' bond. This constraint was the purpose of the study, namely to measure what side-chain geometry imposes on receptor recognition. Of the five analogues prepared, AM-906 showed the highest affinity for CB1, with a Ki of 0.89 nM in displacement of [3H]CP-55940 on rat brain microsomes, and the widest separation between CB1 and CB2, the affinity for CB2 being 9.5 nM on a mouse spleen preparation. In the same assays, THC showed 47.6 nM at CB1. The limit of this finding must be stated plainly: only binding was measured. No functional assay has been published for this molecule, so that its efficacy, that is to say whether it is a full or a partial agonist, is not established. Synthetic cannabinoid receptor agonists are as a rule full CB1 agonists, where THC is only a partial agonist, and it is this difference that underpins their hazard; for AM-906 in particular, that property remains an inference drawn from neighbouring molecules and not a measurement. No action has been studied at TRPV1, GPR55, PPAR-gamma or 5-HT1A, and human data are entirely lacking.

No known biosynthetic pathway: the molecule is produced by no living organism. It arises from total organic synthesis on a classical cannabinoid scaffold, by addition of an organocuprate to a terpenoid ketone, closure of the dihydropyran ring, then stereospecific introduction of the hydroxymethyl group at position 9. France
AM-906 is named in no French text: neither the decree of 22 February 1990 nor the subsequent ANSM decisions cite it, whereas HU-210 and HU-243, built on the same scaffold, have appeared there by name since the decree of 3 October 2017. The so-called tetrahydrocannabinol clause, which covers THCs, their esters, their ethers and their salts, does not apply either: the molecule is a hexahydrocannabinol, hence neither a THC nor an ester, ether or salt of THC. The operative text is the generic clause added to annex IV by the decision of 22 May 2024, which covers any substance derived from the benzo[c]chromene core, non-, partially or fully hydrogenated on ring A, substituted at position 1 by a hydroxyl function, at position 3 by an alkyl, alkenyl or alkynyl chain, at position 6 by one or two alkyl groups, and at position 9 by a ketone, alkyl, hydroxyalkyl or alkoxy function. AM-906 satisfies each of these points, its hept-1-enyl chain falling explicitly under the term alkenyl and its hydroxymethyl under the term hydroxyalkyl. The French status is therefore that of a narcotic caught by a generic clause and not by listing by name: production, possession, transfer and use are criminally punishable.
European Union
No Union text covers this molecule. The survey published in 2020 by Zangani and colleagues, which compares the lists of the European drugs agency as at 1 April 2019 with those of the United Nations Office on Drugs and Crime as at 13 June 2019, places AM-906 outside both registers: it appears there only through the mentions recorded on consumer forums. No risk assessment has been conducted by the European scientific committee and no delegated directive has added it to the annex of Framework Decision 2004/757/JHA. Control therefore falls to each Member State, several of which have adopted generic clauses covering cannabinoid scaffolds, as France has. Internationally, the substance appears neither in the Single Convention of 1961 nor in the 1971 Convention.

Cannabinoïde classique de la série AM : noyau benzo[c]chromène dont le cycle carbocyclique est entièrement saturé, ce qui en fait un hexahydrocannabinol et non un tétrahydrocannabinol. Hydroxyle phénolique en position 1, gem-diméthyle en position 6, hydroxyméthyle d'orientation bêta en position 9, chaîne (Z)-hept-1-ényle en position 3. La double liaison de cette chaîne bloque la rotation autour de la liaison C1'-C2' : la géométrie du pharmacophore latéral est donc figée, ce qui était l'objet même de la conception de la molécule.
Pharmacokinetics
Aucune donnée de pharmacocinétique n'a été publiée, ni chez l'être humain ni chez l'animal. Le travail de 1996 s'est arrêté à la liaison sur préparations membranaires, sans administration à un organisme entier. La biodisponibilité, la demi-vie, le volume de distribution et le passage de la barrière hématoencéphalique restent donc inconnus pour cette molécule.
Metabolism
Le métabolisme de l'AM-906 n'a fait l'objet d'aucune étude publiée. La structure comporte deux sites de conjugaison attendus, l'hydroxyle phénolique en position 1 et l'alcool primaire porté par le carbone 9, ainsi qu'une double liaison latérale susceptible d'oxydation ; ces éléments relèvent de l'inférence structurale et non de résultats expérimentaux. Aucun métabolite n'a été identifié ni proposé comme marqueur analytique.
Toxicology and risks
Aucune étude de toxicologie n'a été conduite sur cette molécule et aucun cas d'intoxication humaine n'a été rapporté. Cette absence de signalements traduit l'absence du marché, non une innocuité démontrée. Le risque attendu est celui de la famille des agonistes synthétiques des récepteurs cannabinoïdes, où l'activation forte de CB1 s'accompagne de tachycardie, d'anxiété aiguë, d'agitation, de confusion et de vomissements répétés, et, dans les formes graves décrites pour d'autres composés de cette famille, de convulsions et de troubles du rythme cardiaque. Ces observations appartiennent à ces autres composés et non à l'AM-906, pour laquelle aucune donnée humaine n'existe. La très forte affinité mesurée pour CB1 place la substance parmi les ligands les plus puissants de sa famille, ce qui rend toute exposition non maîtrisée d'autant plus risquée.
Detection and analysis
Aucune méthode analytique dédiée n'a été publiée et la molécule ne figure pas parmi les cibles des criblages toxicologiques courants ; les tests immunologiques du cannabis ne la détectent pas. Son identification supposerait une chromatographie liquide ou gazeuse couplée à la spectrométrie de masse, de préférence à haute résolution. La difficulté propre à ce composé est isomérique : l'AM-905 ne diffère que par la géométrie de la double liaison latérale, partage la même formule brute et la même masse, et se distingue mal par la seule spectrométrie de masse. Une séparation chromatographique adaptée ou la résonance magnétique nucléaire est nécessaire pour trancher entre les deux.
