Papahatjis, Alexandros Makriyannis et leurs collaborateurs, dans un article du Journal of Medicinal Chemistry consacré à une série d'analogues du Δ8-tétrahydrocannabinol dont la chaîne latérale porte une insaturation destinée à limiter les conformations accessibles. In plain terms
AMG-1 is a molecule made in a laboratory, absent from hemp and from every living organism. It is a modified THC: chemists replaced its side chain with a longer, more rigid one in order to understand how this part of the molecule sits in the receptors. It binds very strongly to the brain's CB1 receptor, it has never been studied in humans and it enters into no consumer product.

Pharmacology
AMG-1 is a classical synthetic cannabinoid: the dibenzopyran skeleton of Δ8-tetrahydrocannabinol, whose pentyl chain at position 3 has been replaced by a hept-1-ynyl chain. This modification belongs to a programme carried out in the late 1990s to map the receptor sub-site that accommodates the side chain; it yields a very high affinity for both cannabinoid receptors, with an inhibition constant of 0.65 nM at CB1 and 3.1 nM at CB2 in the original publication, that is, a modest preference for CB1. Another laboratory reported 36 nM at CB1 with its own protocol, a discrepancy that argues for comparing only measurements obtained under the same conditions. Behavioural assays in mice — reduced spontaneous activity, analgesia and hypothermia — confirm functional activation of CB1 in vivo, of a potency close to that of Δ9-tetrahydrocannabinol. Beyond this core of results, everything is missing: intrinsic efficacy unquantified, pharmacokinetics and metabolism unstudied, human data non-existent. AMG-1 remains a laboratory tool serving to understand the receptor, not a drug candidate nor an ingredient.

Origin (research tool)
No biosynthetic pathway: AMG-1 arises from no plant or animal metabolism. The compound is obtained by organic synthesis from the dibenzopyran skeleton of the classical cannabinoids, the pentyl chain of Δ8-tetrahydrocannabinol being replaced by a heptynyl chain by means of a palladium-catalysed coupling with an alkyne.
Legal framework
France
AMG-1 is not listed by name in any French text: it does not appear in the consolidated list of substances classified as narcotics maintained by the ANSM. It does, however, fall under the generic clause of annex IV of the decree of 22 February 1990, broadened by the ANSM decisions of 22 May 2024 and 3 June 2024, which classifies any substance derived from the benzo[c]chromene ring substituted at position 1 by a hydroxyl function, at position 3 by an alkyl, alkenyl or alkynyl chain, at position 6 by one or two alkyl groups and at position 9 by an alkyl group. The hydroxyl at position 1, the hept-1-ynyl chain at position 3, the two methyls at position 6 and the methyl at position 9 of AMG-1 match this definition point by point, and the molecule falls under none of the exclusions provided for (CBN, CBNA, THCV, THCVA, THCA). Classification therefore proceeds through a generic clause, not through a nominative listing; the older entry in the same annex, which covers tetrahydrocannabinols, their esters, ethers and salts, points the same way, the molecule being a tetrahydrocannabinol with a modified side chain.
European Union
There is no harmonised control measure at European Union level: placing under control is a matter for national laws, the European tier being confined to the early warning system and the risk-assessment procedures provided for by Regulation (EU) 2017/2101 and Directive (EU) 2017/2103. AMG-1 has been the subject of no European risk assessment; the inventory of synthetic cannabinoids published in 2020 placed it among the molecules absent from the lists of the EMCDDA (now the EUDA) as from those of the UNODC. Nor does it appear in the schedules of the United Nations conventions of 1961 and 1971, which name the isomers of tetrahydrocannabinol but not this heptynyl-chain analogue. Several Member States apply generic definitions whose scope differs from the one adopted in France: status therefore varies appreciably from one country to another.

Cannabinoïde classique de type dibenzopyrane (noyau benzo[c]chromène), analogue du Δ8-tétrahydrocannabinol dans lequel la chaîne pentyle en position 3 est remplacée par une chaîne hept-1-ynyle. La triple liaison placée au départ de la chaîne allonge et rigidifie le pharmacophore latéral : c'est exactement la contrainte de conformation que cherchait à imposer l'étude d'origine. Le reste du squelette est celui du Δ8-THC, avec l'hydroxyle phénolique en position 1, deux méthyles en position 6 et un méthyle en position 9.
Pharmacokinetics
Aucune étude de pharmacocinétique n'a été publiée : absorption, distribution, demi-vie et biodisponibilité ne sont documentées ni chez l'animal ni chez l'humain. Les seules observations in vivo disponibles sont des mesures d'effet comportemental chez la souris après administration systémique, sans dosage plasmatique ni cérébral associé.
Toxicology and risks
Aucune étude de toxicité réglementaire, aucun essai clinique, aucun cas d'exposition humaine publié : les données humaines manquent entièrement. Ce qui est établi tient en peu de mots : AMG-1 se lie très fortement au récepteur CB1 et produit chez la souris le tableau cannabimimétique complet, avec une puissance du même ordre de grandeur que celle du Δ9-tétrahydrocannabinol dans le seul protocole où les deux ont été comparés. La marge entre effet et toxicité, le risque cardiovasculaire, neurologique ou psychiatrique, les effets d'une exposition répétée ne sont pas documentés. Il s'agit d'un réactif de laboratoire, jamais formulé ni évalué pour un usage humain, et sa forte activité centrale suffit à écarter toute idée d'ingestion.
Detection and analysis
Aucune méthode médico-légale dédiée n'est publiée pour AMG-1, et le composé ne figure pas dans les panels toxicologiques de routine. Les immunoessais cannabinoïdes, calibrés sur les métabolites du Δ9-tétrahydrocannabinol, ne sont pas conçus pour lui ; une identification supposerait une chromatographie couplée à la spectrométrie de masse et un étalon de référence certifié. Une analyse des forums d'usagers publiée en 2020 relève le nom AMG-1 parmi les cannabinoïdes de synthèse commentés en ligne, tout en constatant qu'il ne figurait alors ni sur la liste de l'ONUDC ni sur celle de l'EMCDDA : le nom circule donc dans les discussions bien avant toute donnée clinique.
