- La série des cyclohexylphénols a été développée par les laboratoires Pfizer dans les années 1970, le chef de file CP 47,497 ayant été décrit comme agent cannabimimétique par Weissman et ses collaborateurs en 1982. Les homologues n-alkyle de cette série, dont celui à six carbones, sont issus du même programme de relations structure-activité, dont les résultats ont été publiés par Melvin et ses collaborateurs en 1993. L'homologue C6 réapparaît hors du laboratoire au début de 2009, lorsque les autorités autrichiennes l'identifient, avec les homologues C8 et C9, dans des mélanges de plantes à fumer de type Spice.

Pharmacology
(C6)-CP 47,497 belongs to the cyclohexylphenol series, cannabinoids described as non-classical: the pyran ring of THC is absent, and all that remains is a phenol bearing a 3-hydroxycyclohexyl group and a branched alkyl chain. It is this chain that governs affinity for the receptors, and the C6 homologue bears one of six carbons, below the optimum of the series, maximal affinity being reached with seven or eight carbons according to the structure-activity work of Melvin and his collaborators. The members of the series for which a measurement exists, the parent CP 47,497 and its C8 homologue, behave as full agonists at the CB1 receptor, whereas THC is only a partial agonist: this difference in efficacy, characteristic of synthetic agonists, is what makes these products unpredictable, because the effect does not plateau. For the C6 homologue itself, no affinity value, no measure of efficacy and no in vivo study have been published, and human data are entirely lacking: its own pharmacology is inferred from its place in the series, it has not been measured.

No known biosynthetic pathway: the molecule does not exist in living things and has never been isolated from hemp. It results from total chemical synthesis in the cyclohexylphenol family, the length of the branched alkyl chain being the only variable distinguishing the homologues from one another.

A narcotic designated by name. CP 47,497-C6 appears in full in annex IV of the decree of 22 February 1990, under the heading for cyclohexylphenols, in the wording "CP 47,497-C6 or (5-(1,1-dimethylhexyl)-2-[(1R, 3S)-3-hydroxycyclohexyl]-phenol". It is therefore not a matter of attachment through the generic clause covering tetrahydrocannabinols, their esters and their ethers: the substance is cited by its name. The classification dates back to 24 February 2009, when six synthetic cannabinoids were added to that annex on a proposal from the national commission on narcotics and psychotropic substances; the heading "derivatives of 2-(3-hydroxycyclohexyl) phenol" moreover covers the whole family generically. Possession, transfer and use fall under the narcotics regime, and no lawful CBD product may contain it.
European Union
No control measure at Union level: this molecule has been the subject neither of a formal risk assessment nor of a Council decision, and it is listed in none of the United Nations conventions of 1961 and 1971. It is monitored as a new psychoactive substance by the European early-warning system, control being a matter for each Member State. Besides France, Austria banned it as early as March 2009 by way of pharmaceutical legislation, Estonia placed it under control in July 2009 and Sweden in September 2009.

Pharmacokinetics
Aucune donnée de pharmacocinétique publiée pour cet homologue, ni chez l'animal ni chez l'être humain : absorption, distribution et demi-vie restent inconnues. La seule voie d'exposition documentée est l'inhalation de végétaux imprégnés, un mode d'administration où la quantité réellement reçue échappe à la personne qui consomme, l'imprégnation des mélanges étant hétérogène d'un point à l'autre du même sachet.
Metabolism
Le devenir métabolique de l'homologue C6 n'a pas été décrit. Les seules données de la série portent sur le chef de file CP 47,497, dont huit métabolites, issus surtout d'hydroxylations et d'oxygénations, ont été caractérisés sur microsomes hépatiques humains par Jin et ses collaborateurs en 2013 ; ce résultat appartient à cette molécule voisine et ne peut pas être transposé à l'homologue à chaîne plus courte.
Toxicology and risks
Aucun cas clinique n'est attribué dans la littérature à cet homologue pris isolément, et les données humaines manquent. Les risques documentés sont ceux de la classe des agonistes de synthèse des récepteurs cannabinoïdes, rappelés par l'agence européenne des drogues : intoxications plus profondes qu'avec le cannabis, convulsions, perte de connaissance rapide, dépression respiratoire, vomissements incoercibles, délire, agitation, psychose, comportements violents, atteintes cardiovasculaires graves et décès. Il n'existe aucun antidote. L'agonisme complet du récepteur CB1, caractéristique de cette classe, explique qu'un écart minime de quantité suffise à changer la nature de l'effet, là où le THC, agoniste partiel, plafonne. Un danger propre à cette famille est l'erreur d'identité : ces molécules se ressemblent à un carbone près, circulent sur des supports muets et ont déjà été retrouvées dans des produits présentés comme du cannabis ou du CBD.
Detection and analysis
Les quatre membres de la série CP 47,497 ne diffèrent que par la longueur de la chaîne latérale, ce qui rend leur distinction analytique délicate et impose des étalons de référence ainsi qu'une bonne séparation chromatographique. La spectrométrie de masse haute résolution les départage par leur formule brute, C20H32O2 pour l'homologue C6, et des spectres de référence figurent dans les bibliothèques médico-légales. Aucun marqueur urinaire validé n'est publié pour cet homologue, et les tests de dépistage usuels du cannabis ne le détectent pas.
