Pharmacology

Methanandamide, or AM-356, is the chiral alpha-methylated analogue of anandamide. The methyl placed alpha to the amide function hinders the approach of fatty acid amide hydrolase (FAAH): in rat brain microsomes, the relative rate of hydrolysis falls to around 2.4 per cent of that of anandamide. This stability is accompanied by an increased affinity for the CB1 receptor, with a Ki of 17.9 to 20 nM on rat brain membranes against about 78 nM for anandamide, while affinity for CB2 remains low, of the order of 815 to 868 nM in mouse spleen. In rodents, the molecule reproduces the classic cannabinoid tetrad: hypothermia, hypokinesia, ring immobility and antinociception. Other targets have been reported: activation of the orphan receptor GPR55 in dorsal root ganglion neurons, blockade of the background potassium channels TASK-1 and TASK-3, and a capsazepine-sensitive component suggesting a contribution from TRPV1 in certain tissues. Methanandamide also induces the expression of cyclooxygenase-2 in human neuroglioma cells, through a mechanism independent of the cannabinoid receptors. All this pharmacology is preclinical: human data are lacking, no clinical trial having been conducted on this substance.

Origin (research tool)

No known biosynthetic pathway: the molecule comes from no living organism. It is obtained by chemical coupling of arachidonic acid with (R)-2-amino-1-propanol, which installs the chiral methyl borne by the ethanolamide head.

Amide d'acide gras appartenant aux N acyléthanolamines : une chaîne arachidonoyle de vingt carbones portant quatre doubles liaisons cis, reliée à une tête éthanolamide qui porte un méthyle de configuration R en alpha de l'azote. Ce méthyle constitue la seule différence structurale avec l'anandamide.

Pharmacokinetics

Les données humaines manquent : aucune étude de pharmacocinétique clinique n'a été publiée. Chez le rongeur, l'administration par voie intraveineuse ou intrapéritonéale produit des effets cannabinoïdes plus durables que ceux de l'anandamide, conséquence directe de la résistance à l'hydrolyse enzymatique. La molécule est très lipophile et se manipule en solution alcoolique ou en émulsion. Aucune donnée de biodisponibilité orale, de distribution tissulaire ou de demi-vie plasmatique n'est disponible chez l'humain.

Metabolism

La molécule a été conçue pour échapper à l'amide hydrolase des acides gras (FAAH). Sur microsomes de cerveau de rat, la vitesse relative d'hydrolyse se situe autour de 2,4 pour cent de celle de l'anandamide, avec une constante de Michaelis voisine de 33 micromolaire. La comparaison des affinités CB1 mesurées en présence et en l'absence de PMSF, un inhibiteur de protéases à sérine, confirme cette stabilité : l'écart observé reste faible, alors qu'il est marqué pour l'anandamide. Le méthanandamide augmente aussi l'expression de la cyclo-oxygénase-2 dans des cellules de neurogliome humain, par une voie indépendante des récepteurs cannabinoïdes. Le devenir métabolique chez l'humain n'a jamais été décrit.

Toxicology and risks

Aucune donnée de toxicologie humaine n'existe : le méthanandamide n'a jamais fait l'objet d'un essai clinique et n'est pas destiné à l'ingestion. Chez l'animal anesthésié, il provoque une réponse tensionnelle biphasique, une hausse de la pression artérielle suivie d'une baisse, largement dépendante du récepteur CB1. Sur cellules épithéliales alvéolaires de rat, il augmente la perméabilité paracellulaire sans réduire la viabilité cellulaire. Chez le rat, une injection unique diminue l'activité des cellules C thyroïdiennes et la calcitonine circulante. Ces signaux précliniques ne permettent aucune extrapolation à l'humain, et la marge de sécurité reste inconnue.

Detection and analysis

Aucune méthode de dépistage de routine ne cible cette molécule. Les immunoessais cannabis usuels visent le métabolite carboxylé du THC et ne la détectent pas. L'identification repose sur la chromatographie couplée à la spectrométrie de masse : des spectres de référence en GC-MS et en spectrométrie de masse en tandem sur l'ion protoné sont déposés dans les banques publiques. Aux États-Unis, AM-356 figure depuis avril 2011 dans la catégorie de rapportage NFLIS consacrée aux cannabinoïdes de synthèse, ce qui reflète une convention de classement des laboratoires de police scientifique et non une mesure réglementaire de classement.
