- La molécule est un dérivé du pravadoline, cet anti-inflammatoire aminoalkylindole abandonné dont l'activité cannabinoïde inattendue a ouvert toute une chimie. Elle a d'abord été décrite comme antagoniste faible des récepteurs cannabinoïdes du cerveau, avant que le laboratoire d'Alexandros Makriyannis et l'équipe de Roger Pertwee à l'université d'Aberdeen n'établissent sa véritable cible. Le travail de référence est celui de Ruth Ross, Heather Brockie, Lesley Stevenson et leurs collègues, publié en février 1999 dans le British Journal of Pharmacology, qui a confirmé la prédiction faite par ces auteurs : la molécule est un ligand sélectif de CB2, et elle s'y comporte non pas en simple antagoniste mais en agoniste inverse. Elle est depuis devenue l'outil pharmacologique standard employé dans le monde entier pour établir qu'un effet observé passe bien par le récepteur CB2.

In plain terms
AM630 is not a substance that is consumed: it is a laboratory reagent. Researchers use it as a switch, to turn off the CB2 receptor and check that an observed effect really does pass through it. It produces no sought-after effect and does not appear on the drug market. Its structure nevertheless brings it, in France, within a general definition covering synthetic cannabinoids.

Pharmacology
AM630 is the reference antagonist at the CB2 receptor, present in thousands of publications as a negative control: when an effect disappears in its presence, the conclusion is that the effect passed through CB2. Derived from pravadoline like the whole aminoalkylindole family, it was first taken for a weak antagonist at brain receptors before the Aberdeen work of 1999 established its selectivity for CB2, with an inhibition constant of 31.2 nM and an affinity ratio of 165 in favour of that subtype. The same work showed that it is not a neutral antagonist but an inverse agonist: far from merely occupying the site, the molecule lowers the constitutive activity of the receptor, which can be read in the 5.2-fold increase in the forskolin-stimulated production of cyclic adenosine monophosphate. The distinction is not academic, since an inverse agonist can produce an effect of its own where a neutral antagonist produces none, and attributing a result to the blockade of an endogenous signal then becomes ambiguous. Two reservations attend its use. The first concerns the residual activity at CB1, where it behaves as a weak partial agonist from micromolar concentration upwards, which requires staying within the selective window. The second is proper to these laboratory assays: the reported values were obtained on recombinant human receptors, and nothing guarantees that they transpose to rodent receptors, as the inter-species variability documented for other CB2 ligands has shown. No human data exist: the molecule has never been the subject of a clinical trial and no human dose is characterised.

No known biosynthetic pathway: the molecule is formed in no living organism. It arises from organic synthesis within the aminoalkylindole family, by acylation at position 3 of an iodinated indole bearing a morpholinoethyl chain on its nitrogen, the ketone bridge being linked to a methoxylated aromatic ring. Manufacture is described here only by its chemical class.
Legal framework
France
The French position is paradoxical and deserves to be known by laboratories. The molecule is not listed by name on any schedule, but its structure corresponds point by point to the generic clause on indol-3-yl methanones in annex IV of the decree of 22 February 1990, which covers methanones borne at position 3 of an indole ring whose nitrogen is substituted, in particular, by a 2-(4-morpholinyl)ethyl group, and whose methanone bridge carbon bears a naphthyl, benzyl, phenyl, cyclopropyl or adamantyl group, whether substituted or not. The indole nitrogen of this molecule bears precisely a morpholinoethyl chain, and the ketone bridge is linked to a methoxylated phenyl. A literal reading of the text therefore places it among narcotics, although it has no recreational use and serves exclusively as a laboratory tool. Classification by generic clause remains less robust than listing by name, but it carries the same practical consequences: possession and use for research purposes fall under the authorisation regime applicable to narcotics.
European Union
No control measure at European Union level covers this molecule, which has been the subject neither of a notification to the early warning system on new psychoactive substances, nor of a risk assessment, nor of a Council implementing decision. It appears in no schedule of the 1961 Convention on narcotic drugs or of the 1971 Convention on psychotropic substances. Its status therefore varies according to national generic clauses on aminoalkylindoles, which are very uneven from one Member State to another: where the structural definition of synthetic cannabinoids is broad, as in France, it is captured without ever having been targeted by the legislator, whereas elsewhere it circulates freely as a laboratory chemical.

Aminoalkylindole de la famille des indol-3-yl méthanones, dérivé du pravadoline. Un noyau indole porte en position 2 un méthyle, en position 6 un atome d'iode et, sur l'azote, une chaîne éthyle terminée par un cycle morpholine ; la position 3 porte un pont cétone relié à un noyau benzénique méthoxylé en para. L'iode en position 6 est l'élément qui fait basculer la sélectivité de CB1 vers CB2 par rapport au pravadoline dont la molécule dérive, et le nom d'usage 6-iodopravadoline en garde la trace. Cette classe chimique est la même que celle des cannabinoïdes de synthèse de première génération, ce qui explique sa capture par les clauses génériques.
Pharmacokinetics
Aucune donnée chez l'être humain, la molécule n'ayant jamais fait l'objet d'un développement clinique. Chez l'animal, elle est employée par voie intrapéritonéale ou intraveineuse comme outil de blocage, à des doses choisies empiriquement pour couvrir la durée de l'expérience, et non caractérisées par des études d'exposition dédiées. Aucune biodisponibilité orale, aucune demi-vie plasmatique, aucun volume de distribution et aucune donnée de passage de la barrière hémato-encéphalique n'ont été publiés pour cette molécule. Cette lacune est réelle : elle limite l'interprétation des expériences in vivo, où la concentration effectivement atteinte au contact du récepteur reste inconnue.
Metabolism
Le devenir métabolique n'a pas été caractérisé. Aucun métabolite n'a été identifié et aucune enzyme n'a été mise en cause expérimentalement pour cette molécule. Les sites de transformation attendus par analogie avec les aminoalkylindoles seraient la déméthylation oxydative du groupe méthoxy, l'oxydation du cycle morpholine et la coupure de la chaîne portée par l'azote, mais aucune publication ne le documente, et il ne s'agit donc que d'une extrapolation structurale. La question de savoir si des métabolites conservent une activité sur CB2 n'a jamais été posée.
Toxicology and risks
Aucune étude de toxicité réglementaire, aucun essai clinique, aucun cas d'intoxication humaine. La molécule n'est pas commercialisée pour la consommation et n'apparaît pas dans les signalements des systèmes d'alerte précoce. Les travaux animaux publiés l'emploient comme outil de blocage et non comme objet d'étude toxicologique : ils ne rapportent pas de mortalité aux doses utilisées, mais cette absence de signalement ne constitue pas une évaluation de sécurité. Aucune dose humaine n'est caractérisée. Le point de vigilance propre à cette molécule est d'ordre expérimental plutôt que sanitaire : son statut d'agoniste inverse et son agonisme partiel résiduel sur CB1 aux concentrations élevées peuvent produire des effets propres que l'expérimentateur risque d'attribuer par erreur au blocage d'un signal endogène.
Detection and analysis
Aucune recherche de routine ne vise cette molécule en toxicologie médicolégale ou en contrôle antidopage, faute d'usage récréatif documenté. Son dosage relève des laboratoires de pharmacologie, par chromatographie liquide couplée à la spectrométrie de masse en tandem, l'atome d'iode conférant une signature isotopique et une masse élevée qui facilitent l'identification. Elle est en revanche susceptible d'apparaître dans les criblages larges de cannabinoïdes de synthèse, qui ciblent la classe des aminoalkylindoles : un laboratoire qui la détecterait dans un prélèvement devrait garder à l'esprit qu'elle relève en France d'une clause générique sur les stupéfiants, alors même que son contexte d'usage est un laboratoire de recherche et non un marché illicite.
