Le code AMG-41 n'apparaît ensuite que dans les bases de données chimiques et dans une revue de 2016 consacrée aux cannabinoïdes classiques, qui le recense sous le numéro 54 de ses tableaux.

In plain terms
AMG-41 is a laboratory molecule, a cousin of THC modified on its side chain. It was made in 2002 to study how the receptors targeted by cannabis work, never to be consumed: it occurs neither in hemp nor in any commercial product. What is known comes down to one point: it binds very strongly to these receptors, about one hundred times better than THC. Human data are entirely lacking.

Pharmacology
AMG-41 is a classical synthetic cannabinoid: the benzo[c]chromene skeleton of Δ8-tetrahydrocannabinol, in which the benzylic carbon of the side chain is incorporated into a cyclopropane ring extended by a hexyl chain. Described in 2002 by the team of Papahatjis, Nikas and Makriyannis, it served to map a subsite of the binding domain of the CB1 and CB2 receptors located at that carbon. The result is a very high affinity: a Ki of 0.44 nM at CB1 and of 0.86 nM at CB2 by displacement of [3H]CP-55,940, against a Ki close to 44 nM for Δ8-THC, that is, a factor of about one hundred. The literature stops there. No functional assay says whether AMG-41 behaves as a full or a partial agonist, and a high affinity says nothing about efficacy; no animal study, no pharmacokinetics, no toxicology and no human data have been published. The later works that mention this compound are 3D-QSAR models reusing the same binding values, with no new measurement. AMG-41 is therefore to be read as an uncharacterised research tool, not as a substance whose effects are known.

No biosynthesis: AMG-41 is produced by no living organism. It is obtained by chemical synthesis from the benzo[c]chromene skeleton of Δ8-tetrahydrocannabinol: the side chain at position 3 is reworked so that its benzylic carbon (position 1') is incorporated into a cyclopropane ring extended by a hexyl chain, the tricyclic core remaining unchanged. The route described in 2002 involves the cyclopropanation of a side chain, with no other modification of the skeleton.
Legal framework
France
AMG-41 is named in no French text. It falls under a generic clause: since the decision of the Director General of the ANSM of 22 May 2024, annex IV of the decree of 22 February 1990 setting the list of substances classified as narcotics covers "any substance derived from the benzo[c]chromene core" carrying a hydroxyl at position 1, at position 3 an alkyl chain optionally substituted by a ring, at position 6 one or two alkyl groups, and at position 9 a methyl. AMG-41 meets these four criteria: it is therefore a narcotic in France without appearing on any list by name. The same decree separately classifies "tetrahydrocannabinols, their esters, ethers, salts as well as the salts of the aforementioned derivatives".
European Union
No control measure specific to AMG-41 exists at European Union level: the molecule has never been reported on the drug market and is not among the substances that have been subject to a risk assessment followed by a European control decision. It is not listed by name in the schedules of the 1961 Single Convention or of the 1971 Convention on Psychotropic Substances. Depending on the Member State, a generic definition covering dibenzopyran-core derivatives may nonetheless capture it, as is the case in France.

Cannabinoïde classique de type dibenzopyrane (squelette benzo[c]chromène), configuration (6aR,10aR) trans comme le Δ9-THC, double liaison en position Δ8. Sa particularité tient à la chaîne latérale en position 3 : le carbone benzylique (position 1') est inclus dans un cycle cyclopropane portant une chaîne hexyle, là où le THC porte une simple chaîne pentyle. Contrairement aux agonistes de synthèse des séries JWH, AM ou FUBINACA, bâtis sur un noyau indole ou indazole, AMG-41 reste structurellement un tétrahydrocannabinol.
Pharmacokinetics
Aucune donnée pharmacocinétique n'est publiée : ni biodisponibilité, ni délai d'action, ni demi-vie, ni distribution, chez l'animal comme chez l'humain. Aucune étude d'administration in vivo n'a été retrouvée dans la littérature évaluée par les pairs : les travaux disponibles se limitent à des essais de liaison sur préparations membranaires et à de la modélisation moléculaire.
Metabolism
Aucune voie métabolique n'a été caractérisée pour AMG-41 : ni enzymes de phase I ou II, ni métabolites, ni profil d'élimination. Le cycle cyclopropane a précisément été introduit pour contraindre la conformation de cette région de la chaîne latérale, mais son devenir métabolique n'a pas été étudié.
Toxicology and risks
Profil toxicologique inconnu : aucune étude de toxicité, aucun cas clinique, aucun décès rapporté, aucune exposition humaine documentée. Cette absence n'est pas un signal de sécurité. AMG-41 se fixe sur CB1 avec une affinité environ cent fois supérieure à celle du Δ8-THC, et les agonistes synthétiques de CB1 qui ont circulé comme drogues sont associés à des convulsions, des troubles du rythme cardiaque, des vomissements incoercibles, des états délirants, des atteintes rénales aiguës et des décès. Ces observations concernent ces autres composés, pas AMG-41, dont l'efficacité fonctionnelle n'a même pas été mesurée. Aucune dose humaine n'est caractérisée et aucune ne peut l'être.
Detection and analysis
Aucune méthode analytique dédiée à AMG-41 n'est publiée et le composé n'apparaît pas dans les bibliothèques de criblage des nouveaux produits de synthèse. Aucun numéro CAS ni matériau de référence certifié n'est référencé dans les bases consultées, ce qui interdit en pratique une identification de routine : il faudrait une chromatographie couplée à la spectrométrie de masse haute résolution et un étalon synthétisé pour l'occasion. Les immunoessais urinaires du cannabis ciblent le métabolite carboxylique du THC et ne sont pas conçus pour un analogue à chaîne latérale modifiée.
