In plain terms
AZ-11713908 is a laboratory molecule developed by the pharmaceutical company AstraZeneca. It has no connection with hemp and is found in no plant: it was designed to activate the same receptors as THC while remaining largely outside the brain, the idea being to relieve pain in animals without marked psychoactive effect. No study in humans has been published.

Pharmacology
A cannabinoid receptor agonist from a pharmaceutical programme, AZ-11713908 binds to human CB1 with nanomolar affinity and activates it with a maximal effect greater than that of WIN 55,212-2: it is a full agonist, where THC is only a partial agonist. Its affinity for human CB2 is stronger still, but the direction of the effect changes with species, partial agonism in the mouse and inverse agonism in the rat, which forbids any conclusion for humans. The originality of the molecule lies elsewhere: it penetrates the brain poorly, so that the analgesia observed in rodents in inflammatory and neuropathic pain models works through peripheral CB1 receptors. Three converging arguments establish this: the effect disappears in mice lacking CB1 and persists in those lacking CB2, in vivo electrophysiology places the site of action in the periphery, and local application suffices to produce it. The file stops there: nothing is published on TRPV1, GPR55, PPARγ or 5-HT1A, no toxicology study exists, and human data are entirely lacking.

Origin (research tool)
No biological pathway: the molecule is produced by no organism. It is obtained by total synthesis, through assembly of a substituted benzimidazole nucleus followed by sulfonylation of the aromatic amine with a thiophene derivative, without any step of plant origin.
Legal framework
France
Not listed by name on the French narcotics list. The decree of 22 February 1990 covers, in annex IV, tetrahydrocannabinols, their esters, ethers and salts, together with a generic family of indol-3-yl methanone derivatives and a long list of synthetic cannabinoids designated one by one. AZ-11713908, a benzimidazol-5-yl sulfonamide, falls neither under the tetrahydrocannabinol nucleus nor under the generic indole definition, and its name does not appear in the list: it is therefore not classified in France to date. The fact that benzimidazoles such as FUBIMINA had to be added by name confirms this reading. This status is that of a laboratory compound never marketed, and it may change if the ANSM decides to list it.
European Union
No classification at European Union level. The substance appears neither in the lists of the European drugs agency (EUDA, formerly the EMCDDA) nor in those of the UNODC, and it has been the subject of no European risk assessment and no control decision. A survey published in 2020, comparing these two official lists with the results of a crawler run over user forums, places it precisely among the molecules absent from the lists and mentioned only online. It is not placed under control by the international conventions. Several Member States nevertheless apply generic provisions on synthetic cannabinoid agonists, whose scope depends on the chemical scaffolds they describe.

Benzimidazole-5-sulfonamide N-méthylée, portant un cyclohexylméthyle sur l'azote 1 et, en position 2, un méthylène relié à une pyridine éthoxylée. Agoniste cannabinoïde dit non classique, sans parenté structurale avec les cannabinoïdes du chanvre.
Pharmacokinetics
Les seuls paramètres publiés viennent de l'animal. Le rapport des concentrations cerveau sur plasma mesuré chez le rat après administration sous-cutanée est d'environ 0,07, et de l'ordre de 5 pour cent chez la souris : l'exposition centrale reste faible, ce qui fonde la notion de restriction périphérique. La molécule est peu soluble en milieu aqueux à pH neutre et franchit faiblement une monocouche de cellules Caco-2, deux éléments cohérents avec cette faible pénétration. Aucune donnée de pharmacocinétique humaine n'existe.
Metabolism
Les microsomes hépatiques humains et de rat dégradent la molécule très vite, la valeur de clairance intrinsèque rapportée dépassant la borne supérieure du test. Les métabolites n'ont pas été identifiés et aucune voie enzymatique précise n'est documentée.
Toxicology and risks
Aucune étude de toxicologie n'est publiée et aucune exposition humaine n'est rapportée. Chez le rat, un test d'Irwin a relevé moins de signes centraux qu'avec le WIN 55,212-2, agoniste qui pénètre le système nerveux central, mais ces signes n'étaient pas absents. Le point important pour la sécurité est l'efficacité intrinsèque : la molécule active le récepteur CB1 humain avec un effet maximal supérieur à celui du WIN 55,212-2, donc en agoniste complet, alors que le THC n'est qu'un agoniste partiel. C'est cette activation pleine du CB1 qui explique la gravité des intoxications aux agonistes cannabinoïdes de synthèse, tachycardie, vomissements incoercibles, convulsions et confusion. Ces accidents ont été décrits avec d'autres composés de cette classe, et non avec l'AZ-11713908, pour laquelle il n'existe aucune donnée clinique.
Detection and analysis
Aucune méthode analytique dédiée à cette molécule n'est décrite dans la littérature, et elle ne figure ni dans les listes de substances surveillées par l'EUDA ni dans celles de l'ONUDC. Un travail publié en 2020, fondé sur un robot d'exploration des forums d'usagers, la recense parmi les cannabinoïdes de synthèse cités en ligne, sans saisie ni cas d'intoxication documenté. Les laboratoires de toxicologie ne la recherchent donc pas en routine.
