- Isoflavone connue de la littérature phytochimique du milieu du XXe siècle, décrite chez le trèfle des prés (Trifolium pratense) et le pois chiche (Cicer arietinum) ; les sources secondaires ne s'accordent pas sur la date exacte de la première isolation, aucune année ne peut donc être avancée avec certitude. Son entrée dans la pharmacologie du système endocannabinoïde est en revanche précisément datée : en 2010, l'équipe de Christopher Fowler à l'université d'Umeå, avec Benjamin Cravatt, Roger Pertwee et Ruth Ross parmi les coauteurs, publie dans le British Journal of Pharmacology la caractérisation de la biochanine A comme inhibiteur naturel de la FAAH, l'hydrolase qui dégrade l'anandamide. Une étude de 2022 a repris cette activité dans un contexte comportemental chez la souris.

- A natural product of legumes, absent from cannabis. Biochanin A is, together with formononetin, one of the major isoflavones of red clover (Trifolium pratense); it is also present in chickpea, soya, alfalfa, groundnut, rose apple and several Baptisia species. It therefore belongs to the ordinary diet and is found in red clover extracts sold as food supplements. No plant of the genus Cannabis produces it.

In plain terms
Biochanin A is a molecule from red clover, also present in chickpea, soya and groundnut. It does not come from hemp and bears no structural relation to CBD. In the laboratory it slows the enzyme that destroys anandamide, a cannabinoid the body makes itself, but the amounts reached in the blood after a meal or a supplement remain far too low for this effect to occur in humans.

Pharmacology
Described in 2010 as a natural FAAH inhibitor, biochanin A blocks the hydrolysis of anandamide with IC50 values of 1.4 µM in rat brain, 1.8 µM in mouse liver and 2.4 µM at the human enzyme, according to a mixed-type inhibition. Selectivity clearly leans to the enzymatic side: no effect on FAAH-2, and negligible binding to the CB1 and CB2 receptors. In the mouse, a local injection into the paw reduces the spinal phosphorylation of ERK induced by formalin, an effect abolished by the CB1 antagonist AM251; given intravenously, the molecule potentiates the effects of anandamide without raising the brain concentration of anandamide, its brain-to-blood distribution ratio being very low. The authors conclude that FAAH inhibition is peripheral, not central. A 2022 study nonetheless reports a rise in anandamide in the mouse prefrontal cortex and a reduction of immobility in the forced swim test, which leaves the central question open. Two limits deserve stating: biochanin A also activates PPARα and PPARγ, inhibits aromatase and acts as a phyto-oestrogen, which forbids attributing its effects to the endocannabinoid system alone; and the plasma concentrations measured in humans after a red clover supplement, of the order of 170 nM at most, remain about tenfold below its potency at FAAH. Human data are lacking: no clinical trial has assessed this endocannabinoid activity.

Origin (research tool)
The isoflavone pathway specific to the Fabaceae: p-coumaroyl-CoA arising from the phenylpropanoid pathway is condensed with malonyl-CoA by chalcone synthase, then chalcone isomerase leads to naringenin. A 2-hydroxyisoflavanone synthase of the cytochrome P450 family (CYP93C) performs the aryl migration characteristic of isoflavones, and the dehydration that follows yields genistein. An S-adenosylmethionine-dependent isoflavone 4'-O-methyltransferase finally methylates the hydroxyl at position 4', delivering biochanin A. The molecule is thus methylated genistein, which explains why its demethylation in the body regenerates genistein.
Legal framework
France
Biochanin A is named in no annex of the decree of 22 February 1990 setting the list of substances classified as narcotics. Nor is it captured by the generic clause of annex IV, which covers tetrahydrocannabinols, their esters, ethers and salts as well as the salts of the aforementioned derivatives: it is a plant isoflavone, with neither a cannabinoid skeleton nor any chemical kinship with THC. Its status is therefore that of an unclassified substance, a normal constituent of red clover and chickpea. A concentrated extract falls under food law and the regulation of food supplements: the 2005 report of AFSSA and AFSSAPS on phyto-oestrogens sets an intake limit of 1 mg per kilogram of body weight per day of aglycone isoflavones, and recommends labelling advising against these products for women with a personal or family history of breast cancer.
European Union
No European control measure targets biochanin A: it appears neither in the schedules of the 1961 Single Convention on Narcotic Drugs nor in those of the 1971 Convention, and it has never been the subject of a risk assessment as a new psychoactive substance. It is treated as a food constituent. EFSA assessed in 2015 the safety of supplements of isolated isoflavones, a category to which biochanin A belongs, in peri-menopausal and post-menopausal women: the human data examined did not establish any harmful effect on the mammary gland, the uterus or the thyroid at the usual doses of these products. An isolated and highly concentrated ingredient would fall under Regulation (EU) 2015/2283 on novel foods.

Isoflavone O-méthylé : 3-phénylchromén-4-one portant deux hydroxyles en positions 5 et 7 et un méthoxyle en position 4' du cycle phényle. C'est l'éther 4'-méthylique de la génistéine, à laquelle elle revient par déméthylation dans l'organisme. Aucune parenté avec le squelette terpénophénolique des cannabinoïdes végétaux ; par sa planéité et ses hydroxyles phénoliques, elle appartient à la famille des flavonoïdes alimentaires, dont plusieurs membres inhibent faiblement la FAAH.
Pharmacokinetics
La biodisponibilité orale est très basse : inférieure à 4 % chez le rat, pour la biochanine A comme pour la génistéine issue de sa déméthylation, la fraction plasmatique libre n'étant que de 1,5 %. Les formes circulantes majoritaires sont des conjugués glucuronides et sulfates, retrouvés dans le plasma et dans la bile, et des pics de réabsorption traduisent un cycle entéro-hépatique. La pénétration cérébrale est faible, avec un rapport de distribution cerveau sur sang de l'ordre de 0,04, ce qui explique l'absence d'élévation de l'anandamide cérébral dans l'étude princeps. Chez l'être humain, les concentrations plasmatiques maximales rapportées après prise d'un complément de trèfle rouge sont d'environ 170 nM, très en dessous des concentrations actives sur la FAAH in vitro. Aucun jeu complet de paramètres humains, demi-vie ou volume de distribution, n'est établi.
Metabolism
La transformation principale est une 4'-O-déméthylation qui régénère la génistéine, catalysée par les cytochromes P450 1A1 et 1B1, le CYP1B1 privilégiant cette déméthylation sur les hydroxylations. Suivent des conjugaisons rapides, glucuronidation et sulfatation, portant sur la biochanine A comme sur la génistéine, avec l'urine comme principale voie d'excrétion après cycle entéro-hépatique. Des dizaines de métabolites ont été inventoriés chez le rat, dans les microsomes hépatiques et sous l'action du microbiote intestinal, parmi lesquels la formononétine, la daidzéine et des dérivés glycosylés. La molécule est donc pour partie une prodrogue de la génistéine, ce qui complique l'attribution de ses effets à l'une ou à l'autre.
Toxicology and risks
Aucune étude de toxicité réglementaire n'existe pour la molécule isolée. Chez le rat, une administration orale répétée pendant vingt-huit jours n'a produit ni signe de toxicité clinique ni variation du poids des organes dans les travaux rassemblés par les revues récentes. Les points de vigilance sont endocriniens et métaboliques plutôt que neurologiques : la biochanine A est un phyto-œstrogène, elle inhibe l'aromatase et l'expression du gène CYP19 in vitro, et elle inhibe les cytochromes P450 1A1 et 1B1, ce qui ouvre un risque d'interaction médicamenteuse. C'est à ce titre que l'AFSSA a fixé en 2005 une limite d'apport en isoflavones et déconseillé ces compléments aux femmes ayant des antécédents de cancer du sein, tandis que l'EFSA n'a pas retenu, en 2015, d'effet nocif documenté aux doses usuelles des compléments chez les femmes ménopausées. Les données humaines manquent sur l'activité endocannabinoïde elle-même, ainsi que sur les conséquences éventuelles d'une inhibition prolongée de la FAAH.
Detection and analysis
Le dosage se fait par chromatographie liquide couplée à la spectrométrie de masse ou à une détection par barrette de diodes, sur les extraits végétaux comme sur le plasma, la bile et l'urine ; les protocoles publiés recourent également à la chromatographie gazeuse couplée à la spectrométrie de masse et à l'UHPLC haute résolution pour l'inventaire des métabolites. Une hydrolyse enzymatique préalable des conjugués est nécessaire, faute de quoi la fraction libre est largement sous-estimée. Aucun dépistage toxicologique ou forensique ne la recherche : elle n'appartient à aucune liste de substances contrôlées et, sans parenté structurale avec le THC, elle échappe aux immunoessais cannabinoïdes de routine.
