Pharmacology
Cannabisol is a dimer of Δ9-tetrahydrocannabinol: two THC units linked by a methylene bridge, that is, the first carbon-bridged dimeric cannabinoid described in the literature. Zulfiqar and co-workers isolated it in 2012 from high-potency cannabis samples rich in cannabigerol, pooled to reach a sufficient quantity of material: thirty-six batches totalling 630 g yielded 10 mg of purified compound, its structure established by high-resolution mass spectrometry and nuclear magnetic resonance. The authors propose a biogenesis proceeding through decarboxylation of tetrahydrocannabinolic acid then enzymatic installation of the one-carbon bridge, without identifying any enzyme. No pharmacology has been published: the isolation paper includes neither binding assays nor activity measurements, and no later work has filled this gap. Its mass, close to twice that of THC, places it outside the usual range of cannabinoid ligands, though no experiment has established this.

Biosynthetic pathway
The molecule is a dimer: two units of Δ9-tetrahydrocannabinol linked by a methylene bridge between their positions 2, that is, on the aromatic ring of each unit. The authors of the isolation propose an initial decarboxylation of Δ9-tetrahydrocannabinolic acid, followed by nucleophilic attack of the resulting anion on the electrophilic carbonyl carbon of a coenzyme A ester, then an enzymatic reduction that installs the one-carbon bridge. This pathway is presented as plausible, not as demonstrated: no enzyme has been identified.

Pharmacokinetics
Aucune donnée d'absorption, de distribution, de biodisponibilité ou d'élimination n'a été publiée, ni chez l'animal ni chez l'humain. La masse moléculaire élevée et la très forte lipophilie attendue d'un dimère de THC laissent supposer un comportement éloigné de celui du monomère, mais aucune mesure n'existe.
Metabolism
Aucune voie métabolique n'a été décrite et aucun métabolite n'a été identifié. L'implication éventuelle des cytochromes P450 ou des enzymes de conjugaison n'a jamais été étudiée pour ce composé.
Toxicology and risks
Aucune étude de toxicité expérimentale n'existe et aucun cas d'exposition humaine n'a été rapporté. Le composé n'ayant jamais été commercialisé ni produit en quantité, il n'apparaît dans aucun signalement de pharmacovigilance ni dans aucune série d'intoxications. Le profil de risque reste inconnu.
