Pharmacology

Cannabifuran belongs to the furan family of cannabinoids, the one in which the pyran ring of tetrahydrocannabinol is replaced by a furan ring. It was isolated from an Afghan hashish extract, together with dehydrocannabifuran and two other minor compounds, its structures being established by mass spectrometry and nuclear magnetic resonance. Its tricycle is entirely aromatised, which makes it one of the most oxidised plant cannabinoids known and explains why it is described in resin rather than in the fresh plant. No measured affinity for CB1 or CB2 has been published. The review by Anand and co-workers (2024) notes that the furan class has not given rise to reports of psychoactivity and proposes to see in it a structural switch towards non-psychoactive cannabinoids, a hypothesis that no affinity measurement has yet come to support. The compound finally carries a lesson in method: ElSohly and Slade (2005) report that a cannabinoid published as cannabinodiol by Van Ginneken corresponded in reality to cannabifuran, which invalidates the references that rely on that assignment.

Biosynthetic pathway

The molecule belongs to the furan family of cannabinoids, the one that replaces the pyran ring of THC with a furan ring. The tricycle is here completely aromatised, which presupposes an extensive dehydrogenation of the cannabinoid skeleton and explains why it is described in resin rather than in the fresh plant. Cannabinoïde de type cannabifurane, bâti sur un noyau dibenzofuranique entièrement aromatique portant un hydroxyle en position 1, une chaîne pentyle en position 3, un méthyle en position 6 et un groupe isopropyle en position 9. Cette aromatisation complète le distingue de la cannabielsoïne, dont le noyau dibenzofuranique reste partiellement saturé, et du THC, bâti sur un benzo[c]chromène. Le déshydrocannabifurane, qui porte un groupe isopropényle au lieu d'isopropyle, en est le proche parent.
