Pharmacology

Cannabichromevarinic acid (CBCVA) is the native acid precursor of CBCV and the varinic representative of the cannabichromene series. It was isolated in 1977 by Shoyama and his collaborators from the Thai cannabis “Meao variant”, in the work that yielded the first four propyl-chain acid cannabinoids ever isolated. Like the other acid cannabinoids, it is not psychoactive, its free carboxylic function strongly degrading affinity for the orthosteric site of CB1. It nevertheless possesses a measured pharmacological activity: in the Scn1a+/- mouse, a model of Dravet syndrome, CBCVA significantly raises the thermal threshold for the onset of generalised tonic-clonic seizures, an effect also observed for CBC and CBCA but not for CBCV. These results are exclusively from animals and support no recommendation in humans.

Biosynthetic pathway

Biosynthetic pathway: divarinolic acid, the propyl homologue of olivetolic acid, condenses with geranyl pyrophosphate to form cannabigerovarinic acid. CBCA synthase then cyclises this precursor into cannabichromevarinic acid. Like the other acid cannabinoids, CBCVA decarboxylates into CBCV under the effect of heat, time or light: it is the form actually present in the living plant.

Pharmacokinetics

Chez la souris, après administration intrapéritonéale, le CBCVA est bien absorbé et présente une pénétration cérébrale substantielle ; les rapports cerveau sur plasma mesurés pour la série CBC vont de 0,2 à 5,8. Aucune donnée pharmacocinétique humaine n'est disponible.

Metabolism

Le métabolisme du CBCVA n'est pas documenté spécifiquement. La décarboxylation non enzymatique en CBCV, provoquée par la chaleur lors du séchage, du stockage ou de la combustion, constitue sa principale voie de transformation avant même toute prise en charge métabolique.

Toxicology and risks

Aucune étude de toxicologie dédiée n'est publiée pour le CBCVA. Aucun événement indésirable humain ne lui est attribué. Les doses actives rapportées le sont chez l'animal uniquement, et aucune dose humaine caractérisée ne peut en être déduite.

Detection and analysis

Le CBCVA se quantifie par chromatographie liquide couplée à la spectrométrie de masse, méthode qui préserve la fonction acide. La chromatographie en phase gazeuse le décarboxyle en CBCV à l'injection et ne permet donc pas de le distinguer de sa forme neutre sans dérivatisation préalable.
