Pharmacology
Cannabidibutol (CBDB) is the butyl-chain homologue of cannabidiol, isolated and characterised in 2019 by the team of Citti and Linciano during an analysis of the impurities of CBD extracted from hemp, where it features alongside cannabidivarin among the two major impurities. Its structure was established by nuclear magnetic resonance, ultraviolet and infrared spectroscopy, circular dichroism and high-resolution mass spectrometry, and its absolute configuration (1R,6R) confirmed by comparison with the synthetic stereoisomer. Its pharmacology at the cannabinoid receptors remains unexplored: no CB1 or CB2 affinity has been published. The only activity data come from a 2021 study in breast cancer cell lines, in which CBDB causes oxidative stress, a loss of mitochondrial membrane potential and a disorganisation of the endoplasmic reticulum, with a potency comparable to that of cannabidiol.

Biosynthetic pathway
Biosynthetic pathway: CBDB belongs to the butyl series of cannabinoids, a minor series in hemp, in which the polyketide precursor bears a four-carbon chain instead of pentyl olivetolic acid. Prenylation followed by cyclisation by CBDA synthase leads to the corresponding acid, whose decarboxylation releases CBDB. Its constant presence in CBD extracts makes it an analytical marker of the plant origin of cannabidiol.
Legal framework
France
CBDB is not listed by name on the French narcotics list. A homologue of cannabidiol, it does not share the benzo[c]chromene nucleus of the THC-type derivatives covered by the generic clause of the ANSM Decision of 22 May 2024. Its presence as an impurity of CBD falls under the framework applicable to hemp.
European Union
Not listed in the European Union narcotics schedules. Concentrated preparations intended for food fall under Regulation (EU) 2015/2283 on novel foods (Novel Food).

Phytocannabinoïde de type cannabidiol, homologue à chaîne butyle (C4) du CBD. La chaîne latérale du noyau résorcinol compte quatre carbones au lieu des cinq du cannabidiol. La configuration absolue (1R,6R) a été confirmée par comparaison avec le stéréoisomère obtenu par synthèse.

Aucune donnée pharmacocinétique n'est publiée pour le CBDB, chez l'animal comme chez l'humain.
Metabolism
Les voies métaboliques propres au CBDB ne sont pas documentées. Une prise en charge hépatique par les cytochromes P450 comparable à celle du cannabidiol est attendue mais non démontrée.
Toxicology and risks
Aucune étude de sécurité humaine n'est disponible. Sur cellules de cancer du sein, le CBDB induit une hausse significative des espèces réactives de l'oxygène, réversible par la N-acétylcystéine, une perte du potentiel de membrane mitochondriale, une baisse de l'ATP d'environ 35 à 40 % et une désorganisation du réticulum endoplasmique accompagnée d'une accumulation de vacuoles autophagiques. Ces observations sont in vitro et ne préjugent pas d'un profil de sécurité humain.
Detection and analysis
Le CBDB se quantifie par chromatographie liquide haute performance couplée à la spectrométrie de masse. Sa présence, aux côtés du cannabidivarine, sert de critère analytique pour distinguer un cannabidiol d'origine végétale d'un cannabidiol de synthèse, lequel en est dépourvu.
